Peripheral naive CD4+ T cells selectively differentiate to type1 helperT (Th1), type2 helper T (Th2), and interleukin (IL)-17 producingT (Th17)cells, depending on the priming conditions. Since these subsets develop antagonistically to each other to elicit subset-specific adaptive immune responses, balance between these subsets can regulate the susceptibility to diverse immune diseases. The present study was under taken to determine whether poly-γ-glutamic acid (γ-PGA), an edible and safe exopolymer that is generated by microorganisms such as Bacillus subtilis, could modulate the development pathways of Th subsets. The presence of γ-PGA during priming promoted the development of Th1 and Th17 cells but inhibited development of Th2 cell. γ-PGA up-regulated the expression of T-bet and ROR-γt, the master genes of Th1 and Th17 cells, respectively, while down-regulating the level of GATA-3, the master gene of Th2 cells. γ-PGA induced the expression of IL-12p40, CD80 and CD86 in dendritic cells and macrophages in a Toll=likereceptor-4-dependent manner, and the effect of γ-PGA on Th1/Th2 development was dependent on the presence of antigen-presenting cells. Furthermore, γ-PGA-stimulated dendritic cells favored the polarization of naive CD4+ T cells toward Th1 cells rather than Th2 cells. Incontrast, γ-PGA affected Th17 cell development, regardless of the presence or absence of antigen presenting cells. Thus, these data demonstrate that γ-PGA has the potential to regulate the development pathways of naive CD4+ T cells through antigen-presenting cell-dependent and -independent machanisms and to be applicable to treating Th2-dominated diseases. In the second part of the study, we investigated the effect of γ-PGA on dendritic cells, and its potential for treating Th2-mediated allergic asthma. γ-PGA selectively signaled bone marrow-derived conventional but not plasmacytoid dendritic cells to activate NF-kB and mitogen-activated protein kinases, leading to upregulation of CD86, CD40, and IL-12. These effects of γ-PGA were dependent on Toll-like receptor (TLR)-4 and independent of TLR-2. Importantly, intranasal administration of γ-PGA into ovalbumin-sensitized/challenged mice reduced airway defects such as airway hyper-responsiveness, leukocyte influx, goblet cell hyperplasia, and eosinophilia. Treatment of mice with γ-PGA significantly reduced the multiplication and Th2 polarization of mediastinal lymph node T cells upon allergen-specific restimulation. These anti-asthmatic effects of γ-PGA were abolished in TLR-4-defective mice. Our observations highlight the potential of γ-PGA in the treatment of asthma and other allergic disease in which Th2 polarization plays an important role.
