1) CD4+ T 세포에 자궁내막증 환자 복강액을 처리하면 대조군의 복강액의 처리에 비교하여 IP-10 생성이 증가하였음. 자궁내막증 환자 복강액의 IL-8과 IP-10의 농도 사이에는 positive- correlation 상관성이 있었음. 자궁내막증 환자 복강액의 IP-10 농도는 복강액을 처리한 호중구, 단핵구, 및 CD4+ T 세포에 의하여 분비되는 IP-10의 양과 positive correlation을 보였음.
2) 자궁내막증 환자 복강액의 MCP-1과 MIP1-a의 농도는 서로 상관관계를 보였음. 단핵구 및 T세포

3) 자궁내막증 환자의 복강액을 처리한 단핵구와 T 세포에서 분비되는 MCP-1dm의 농도는 estradiol의 처리에 의하여 감소되지 않았으나 대조군의 복강액이나 LPS로 처리한 세포에서 분비되는 MCP-1의 양은 estadiol에 의하여 현저히 감소하였음.

Purpose Chemokines have been in plicated in the pathological process of endometriosis, Therefore, we investigated the effects of peritoneal fluid obtained from patients with endometriosis (ePF) on the release of angiogenesis-related chemokine, such as IP-10 and IL-8 and monocyte-specific CC chemokines such as monocyte chemotactic protein-1 (MCP-1), regulated upon activation normal T cell expressed and secreted (RANTES), and macrophage inflammatory protein-1a (MIP-1a) by neutrophils, monocytes, and T cells, in addition, we evaluated the correlation between the levels of chemokines in ePF and their release by these cells, Secondary, Hormone, chemokine, and inflammation have been implicated in the pathological process of endometriosis, Therefore, we investigated combined effects of peritoneal fluid obtained from patients with endometriosis (ePF) and estrogen or progesterone on the release of chemokines by neutrophils, monocytes, and T cells.
2. Experimental contents - Cells were obtained from healthy young volunteers and cultured with ePF or a control PF (cPF)obtained from patients without endometriosis, The chemokine levels in the ePF and the superratants of cultured cells with ePF were then evaluated, The correlation between chem okine levels in the ePF and the supernatants of cultured cells with ePF were evaluated, In addition, it was investigated if E2 and PF could modulate secretion of chemokines by leukocytes, neutrophils, monocytes and T cells.
3. Results - 3-1) The addition of ePF to cultures of CD4+ T cells led to a significant increase in the release of IP-10 when compared to cPF, There was a posit ive correlation between the levels of IL-8 and IP-10 in ePF (R=0.89, P=0.041). but not between the levels of IP-10 and IL -8 released by neutrophils, CD4+ T cells, and monocytes, The levels 01 IP-10 in ePF were positively correlated with the release of IP-10 by ePF-treated neutrophils (R=0.89, P<0.001), CD4+ T cells (R=0.93, P<0.001), and monocytes (R=0.70, P=0.01), Moreover, the addition of ePF significantly enhanced the interferon-g-induced release of IP-10 by nuetrophils and CD4+Tcells, 3-2) There was a positive correlation between the levels of MCP-1 and MIP1-a in ePF, The addition of ePF to the cell cultures failed " increase 1he release of MCP-l, RANTES, and MIP-1a when compared to cPF, but the levels of RANTES in ePF were positively correlated with the release of RANTES by ePF-treated monocytes and T cells, Moreover, there was a positive correlation between the levels of RANTES and MIP1-a released by neutrophils and between the levels of MCP-1 and MIP-1a released h, T cells, Finally, the levels of RANTES released by monocyte-derived macrophages and monocytes cultured with ePF were positively correlated, 3-3) The production of MCP-1 by cPF-treated monocytes and T cells is down-regulated by E2 but ePF-induced production of MCP-1 is not inhibited by E2.
4. Expected outcome- Immune cells such as monocytes, neutrophils, and T cells release differential levels of MCP-1, RANTES, and MIP-1a in response to stimulation with ePF. Therefore immune modulation may be one of strategy in treatment of the endometriosis, These findings also suggest a cross-talk between steroid hormones and ePF in production of chemokines by leukocytes, which is implicated in pathogenesis of endometriosis.
