□ 1세부과제명 : 내생 미생물 유래 유도 물질분리, 기전연구 및 실용화

Pseudomonas chlororaphis 06는 뿌리에 관주하였을 때 식물병과 가뭄에 대해 내성을 유도한다. 또한 P. chlororaphis 06가 생성하는 휘발성 물질인 2R,3R-butanediolO] 병 저항성 유도 물질임이 동정되었다. 하지만, 아직까지 06균주와 2R,3R-butanediolO] 식물의 병 저항성과 가뭄 내성을 유도하는 기작에 대한 연구는 아직까지 밝혀진 바 없다. 따라서 이들 미생물과 물질에 의한 가뭄 내성 유도기작을 밝히기 위해 이들 처리에 의한 식물의 기공의 변화를 관찰하고, 이들 처리에 의한 식물의 신호전달 체계를 밝히기 위해 다양한 신호전달 돌연변이체를 이용하여 가뭄 내성 유도 여부를 조사하였다. 또한 병 저항성과 가뭄 내성에 연관성을 알아보기 위해 병 저항성에 중요한 신호물질인 salicylic acid, jasmonic acid, ethylene의 신호 전달 돌연변이체를 이용하여 가뭄 내성 유도 여부를 확인하였다. 또한 p.chlororaphis 06와 butanediol을 처리한 후 식물체내의 salicycli acid와 ABA농도를 측정하여 이들 처리에 의해 식물체내에서의 변화를 측정하였다. 또한 기공개폐에 중요한 신호물질인 Nitric oxide와 과산화수소의 변화를 confocal microscope을 이용하여 측정하고 이들 물질의 억제제를 처리하여 가뭄 내성 유도 여부를 밝혀 이들이 가뭄내성 유도에 어떤 역할을 하는지를 밝힌다.

Pseudomonas chlororaphis 06를 뿌리에 접종하면 식물체내에서 병 저항성과 가뭄에 대해 내성이 유도된다. 또한 이런 스트레스를 처리하기 전에도 06균주만을 처리하였을 때에도 기공이 폐쇄되는 식물의 생리적인 변화가 일어난다. 아직까지 이러한 ISR유도 미생물에 의해 유도되는 priming관련 유전자들은 밝혀진 바 없다. 따라서 06균주 처리에 의해서만 유도되는 potential priming관련 유전자들을 분리하기 위해 06균주를 뿌리에 처리하고 3일 혹은 5일 후에 잎에서 total RNA를 분리하여 Differentially expressed genes들을 분리하였다. 이들 분리된 유전자들의 발현양상을 다양한 처리를 통해서 분석하였고 또한 가뭄 처리시 병저항성 관련 마커 유전자들의 발현 양상을 비교하여 이들 분리된 유전자와 병 저항성 관련 마커 유전자들의 가뭄 내성 유도에서의 발현 양상을 분석하였다.
4. 본 과제 에 서 선 발한 미 생 물, P. chlororaphis 06, K. oxytoca C1036, T. harzianum, B. amyloliqeufaciens IN937a와 미생물 유래 병 저항성 유도 물질, 4-aminobenzoic acid, pyroglutamate, EPS, 3-pentanol의 병 저항성 예방 및 방제 효능을 기존에 알려진 제품인 BTH과 엑스텐을 대조 약제로 검정하였다. 이들의 식물병 예방 및 방제 스펙트럼을 조사하기 위해 고추 세균성반점병원균 갯빛곰팡이병원균 역병균을 처리하여 방제 효능을 비교하였다. 또한 포장에서의 이들 시제품들의 효능을 검정하기 위해 농가 포장에서 고추를 공시작물로 하여 식물병해충 발생 생장 및 수확량 등을 조사하였다.
□ 국제 공동 과제명: 유용-내생균 (Trichoderma) 및 내생균유래 유도불절의 병저항성과 내한발성유도 불질분리 및 설용화
1. 고추역 병 에 저 항성 을 유도하는 유용 내생균 (Trichoderma) 선발
- 미국농무성 에 서 보유하고 있는 Trichoderma 내생균 중 19개 균주를 고추역 병균 CPhythophthora capsici)에 대해 저항성을 유도하는 균주를 colonization 정도, mycoparasitim, antibiosis, metabolite처리, real-time PCR등의 방법을 이용하여 최적의 효과를 갖는 6개의 내생균을 선발하였다.
- 선발된 6개의 내생균을 온실수준에서 처리하여 disease progress curve를 작성하여 내생균이 고추역병균에 저항성을 유도함을 밝혔으며 그 종 최적의 내생균 2 개 균주를 선발하였다.
- 식물체내의 균사체 존재 및 식물세포에 미치는 영향을 규명하고자 내생균을 고추뿌리에 처리한 후 전지-현미경 및 GFP-Trichoderma를 이용하여 내생균의 colonization 패턴을 연구하였다.
2. Real-Time RT -PCR 및 고추 nucroarray 분석에 의한 유도 저헝-성 관련유전자분석
- 위와 같이 선발된 6개 내생균의 고추역병저항성 발생기작을 규명하고자 유도저항성에 관련된 유전자 307H 정도룹 여러 처리후 고추잎과 뿌리에서 total RNA를 추출한 후 Real-Time RT-PCR을 이용하여 Trichoderma 균주에 의해 유도되는 식물의 유전자를 분석하고 이들의 발현패턴을 규명하였다.
- 고추묘에 디음과 같은 처리를 한 후 Real-Time RT-PCR 분석을 하였다:
Trichoderma접종후 2주/5wn 후, Trichoderma 균의 culture filtrate로부터 분리된 metabolite 처리 후, Trichoderma 균의 휘발성물질 처리 후, 고추역벙균(Phythophthora capsici) 처리 2, 4, 6일 후
- 또한 최적의 효과를 갖는 2개균주의 고추역병저항성 발생기작을 규명하고자 고추에 Trichoderma를 접종하여 48시간 후 고추뿌리로부터 total RNA를 추출하여 고추 microarray를 이용하여 고추전체 genome의 반응 기작을 연구 하였다.
3. 유용 내생균 (Trichoderma)으로부터 고추역병균 성장억제물질의 탐색
- 여러 Trichoderma 균의 culture filtrate를 고추역병균 (Phythophthora capsici)의 배지에 첨가하여 배양한 후 고추역병균의 생장을 억제하는 metabolite 및 휘발성물칠룹 탐색하였다.

In order to apply ISR into agricultural field, we use three different microbial groups including endopytic bacteria, soil bacteria and endophytic fungus. Firstly using endopytic bacteria, bacterial volatile-elicited ISR was assessed. Analogous to the role of volatiles in insect resistance volatile organic compounds (VOCs) play a significant role in the induction of plant host resistance. We observed that, both Bacillus subtilis strain GB03 and B. amylolique/aciens strain IN937a produced volatiles that significantly reduced the severity of disease on Arabidopsis thaliana caused by ET11Jinia carotovora subsp. carotovora. Further study of these VOCs identified 2,3-butanediol and 3-hydroxy-2-butanone (also referred to as acetoin) as elicitors of induced systemic resistance (ISR) in A. thaliana against P. carotovora subsp. carotovora. Bacterial VOCs were shown to elicit ISR in several A rab idops is mutants, including a jasmonic acid-insensitive line (coil), an ethylene-insensitive line (ein2), a salicylic acid-degrading transgenic line (NahG), and a nonexpressor of PR proteins (nprl). VOCs from strain IN937a elicited ISR on all of these lines, whereas VOCs from strain GB03 failed to elicit ISR on ein2 plants. The importance of the ethylene-dependent signaling pathway for elicitation of ISR by VOCs from strain GB03 was confirmed in tests using GUS fusions to the PDF1.2 gene, which is an indicator for the ethylene response. Recently, the relationship between ethylene signaling and bacterial VOC-elicited ISR was further substantiated by proteomic analysis that demonstrated significant up-regulation of three major ethylene biosynthesis proteins (i.e. aspartate aminotransferase, S-adenosylmethionine synthetase 2, and methionine adenosyltransferase 3) following bacterial VOC treatment.
Klebsiella oxytoca CI036 (C1036) causes induced systemic resistance (ISR) activity against the soft-rot pathogen Pectobacterium carotovorum subsp. carotovorum SCCl (SCCD. However, microbial metabolites from CI036 involved in ISR activity remain unknown. In order to determine the structures of microbial traits involved in induced systemic resistance of K. oxytoca C1036, tobacco soft rot pathogen, Pectobacterium carotovorum subsp. carotovorum SCCI was used to test induced systemic resistance activity of C1036. The CI036 cultures grown on LB media was extracted with organic solvents, and the resultant extracts were subjected to column chromatography and instrumental analyses. ISR bioassay-guided fractionation of the cultural extracts was involved in the isolation of the microbial determinants from C1036. Mass spectrometric and NMR analyses were performed to determine the structure of the purified induced systemic resistance metabolite.
Root colonization of plants with certain rhizobacteria, such as Pseudomonas chlororaphis 06, induces tolerance to biotic and abiotic stresses. In order to determine mechanism involved induced drought tolerance mediated by P. chlororaphis 06 and 2R,3R-butanediol, we determined correlation with reduced water loss in 06-colonized plants with stomatal closure. Stomatal closure and drought resistance were mediated by production of 2R,3R-butanediol, a volatile metabolite of P. chlororaphis 06. In addition, wild-type bacterial strains and butanediol-deficient mutants were used to determine role of 2R,3R-butanediol in induced drought tolerance. Plant signal pathways involved in 06 and butanediol-mediated induced drought tolerance were determined using Arabidopsis mutant lines related to salicylic acid (SA)-, ethylene-, and jasmonic acid-signaling pathways. Free SA amount was measured in 06-colonized and drought stressed plant to further determine relationship between induced drought tolerance and plant disease resistance. In addition, nitric oxide and hydrogen peroxide productions in guard cells of 06-colonized or butanediol-treated plant were investigated to determine role of NO and activated oxygen species in 06-mediated induced drought tolerance.
Root colonization of Arabidopsis thaliana with Pseudomonas chlororaphis 06 induces systemic tolerance against pathogenic challenge, as well as drought and salt stresses. In order to isolate priming related genes upon root colonization of P. chlororaphis 06, we performed PCR based-differentially expressed genes approach in plants only colonized by P. chlororaphis 06. To confirm the selected potential priming genes upon root-colonization of P. chlororaphis 06, serial real-time RT-PCR analysis were conducted in plants that treated with or without 06, and followed with or without water withholding. Transcripts of the jasmonate-marker gene, VSPl, the jasmonate- and ethylene-regulated gene, PDF1.2, the salicylic acid-regulated gene, PRl, and the ethylene-responsive gene HEL, were investigated upon root colonization with P. chlororaphis 06 or water withholding to determine correlation of plant defense and drought tolerance.
In order to evaluate control efficacies of the selected microbes or metabolites against various plant diseases, we used red pepper as refemce crop plant. Bacterial leaf spot caused by Xanthomonas campestris pv. vesicatoria, gray mold caused by Botrytis cinerea, or phytophthora blight caused by Phytophthora capsici were used as target diseases. In pot experiments, Control efficacies of the formulated products against plant diseases divided in two different treatments, preventive effect, that the formulated products were sprayed 5 days before pathogen inoculations, and curative effect, that the products were sprayed a day before pathogen treatments. The biocontrol efficacies of the products were examined in 2010 in red pepper cultivated farm field. Disease incidence, plant growth, and yield were investigated.
In addition, Endophytic Trichoderma isolates collected in tropical environments were evaluated for biocontrol activity against Phytophthora capsici in hot pepper (Capsicum annuum). Six isolates were tested for parasitic and antimicrobial activity against P. capsici and for endophytic and induced resistance capabilities in pepper.
