고품질 로얄제리 생산을 위하여 화분공급에 따라 로얄제리의 생산량을 조사하였으며, 이에 따른 로얄제리의 물리화학적 분석을 하였다. 도한 생산시기에 따른 기능성을 구명하였으며, 국내산과 중국산 로얄제리의 차별화를 위하여 국내산 로얄제리의 기준물질을 확립하였다. 국내산 로얄제리의 다양한 기능성을 알아보기 위하여 항균, 항산화, 항당뇨, 멜라닌억제효과 등을 조사하였다. 또한 기능성식품원료 재등록을 이한 연구로서 광노화가 유도니 유도된 무모생쥐에서 로얄제리의 식이 공급에 의한 피부 노화 (주름/보습) 개선 및 항산화 효능과 폐경기 노화가 유

Royal jelly is the exclusive food of the queen honeybee larva which is made from the hypopharyngeal and mandibular glands of the worker honeybees. RJ consists of water, proteins, carbohydrates, lipids, mineral salts and vitamins. RJ has several pharmacological properties of anti-angiogenesis, antioxidation, antibacterial activity, vasodilative activity, hypotensive activity, anti-tumor activity and anti-hypercholesterolemic effects among others. Thus RJ has been widely used as a functional food because it contains some health-promoting compounds beyond traditional nutrients. To determine the efficacy of royal jelly (RJ) as a functional dietary food source for anti-aging and anti-oxidation of skin, the aged animal models caused by either UV-irradaion, ovariectomy or natural againg were fed 1% of various domestic RJs (group CW, PC, GP and etc) in diet. Also the content of 10- HDA (trans-10-hydroxy-2-decenoic acid), the standard component of RJ, and anti-oxidant activities were determined in each RJ sample. The domestic RJ contained the 10-HDA in the range of 1.8 ~ 2.1 /g, which was significantly higher than that in Chinese RJ. When RJs were fed in UV irradiated hairless mice for 6 wks, epidermal hydration of group CW was significantly higher than of UV irradiated control group (group UV+). In parallel, epidermal levels of Cer2, a major ceramide (Cer) in epidermis as well as of GlcCer (glucosy,ceramides) and SM (sphingomyelin), precursor lipids of Cer generation, in group CW were higher than in group UV+. The epidermal levels of Cer, GlcCer and SM in other RJ groups (PC, GP and etc) were less than in group CW or similar with those in group UV+. Furthermore, the protein expression of collagen type 1 and transforming growth factor-1 (TGF-1), anti-wrinkle biomarkers, were increased in group CW, but of matrix metalloproteinase (MMP-1), a degradative enzyme of collagen type 1, was not altered in all groups. In the analysis of antioxidant activity in each group, DNA damage (assessed by tail DNA, tail length and tail monent) and induced DNA damage were significantly increased in group UV+. However, the degrees of these DNA damage in all RJ groups were decreased to the similar levels of non-UV irradiated group. However, the activities of catalase (CAT), glutathione peroxidase (GSH-Px) and superoxide dismutase, antioxidant enzymes, were not altered in the epidermis of all groups. In ovariectomized (OVX) rats, epidermal hydration and total Cer did not differ with those in group SHAM. However, epidermal levels of Cer 1 as well as of total GlcCer and SM in group OVX were significantly lower than in group SHAM. Notably, epidermal levels of GlcCer, SM and Cer1 in group CW were higher than in group OVX, which was paralleled with an increased protein expression of -glucosylcerebrosidase (-GlcCer’ase) , an enzyme of GlcCer hydrolysis into Cer. Furthermore, the epidermal level of collagen type 1 in group CW was increased to a level similar in group SHAM. When RJs were fed in 6 month retired C57BL/6J mice for 16 wks, epidermal hydration of group CW was significantly higher than of control group and even higher than of 6 month control group, which paralleled with increased protein expression of collagen type 1. In in vitro experiments of UV-irradiated dermal fibroblasts, CW treatment protected the cell viability from UV irradiation, and futher enhanced the protein expression of collagen type 1 and TGF-1. Also, CW treatment enhanced the migration of human dermal fibroblasts, which was paralleled with a decrease of triglycerol and cholesterol, and an increase of sphingoid bases such as sphingosine and sphinganine, basic components of ceramide synthesis in an wound scratched model. Taken together, our data demonstrate that with high content of 10-HDA, domestic RJs, especially RJ produced in CW area have anti-aging and anti-oxidant efficacies in skin of aged animal models by increasing levels of Cer, procollagen type 1 or TGF-1. The royal jelly CW could be a functional dietary food source for anti-aging and anti-oxidation of skin.
