연구결과
막유화법에 의한 신규 도세탁셀 함유 나노입자의 제조법을 확립하여 제조한 도세탁셀 나노입자는 다음과 같은 특성을 가짐.
- 나노입자는 20 nm 크기로서 yield 및 봉입효율이 각각 약 90% 및 60%로서 생산화가 가능하며 40 ℃에서 6개월 동안 유의할 만한 차이가 없어 적어도 실온 2년간 안정함.
- 도세탁셀 함유 나노입자의 정맥투여시 기존제품과 AUC 및 Cmax 등이 유의할만한 변화를 나타내지 않아 동일한 약효를 나타내냄,
- 도세탁셀 함유 나노입자의 경구투여시 기존제품의 경구투여보다 AUC가 크게 증가하고 절대생체이용율이 6.5배 증가하며 기존제품의 정맥투여보다는 항암효과는 좀 떨어지나 탁월한 항암효과를 나타냄과 동시에 체중 감소 중 독성이 나타나지 않음.
- (추가연구) : P-glycoprotein 억제제로 도세탁셀과 동시 투여시 도세탁셀의 생체내 이용율을 증대시키고 항암효과를 동시에 가지고 있는 cucrcumin을 선정하였으며 curcumin 함유 고상 SMEDDS 15-30분 투여후의 본 연구에서 개발한 도세탁셀 함유 나노입자 투여를 하면 도세탁셀의 절대생체이용율은 약 30%로 증가함.

- 1st year : Preparing of docitaxel-loaded nanoparticles (effect of surfactants on the solubility of docetaxel, phase diagram, preparation of emulsion via membrane emulsification technique, preparation of nanoparticles, assay of docitaxel),
Physicochemical characterization of nanoparticles (yield and encapsulation efficiency, solubility, morphology and size)
- 2nd year : dissolution, efficacy (pharmacokinetic study after intravenous administration of nanoparticles) and patent pending
- 3rd year : efficacy (pharmacokinetic study after oral administration of nanoparticles), antitumor efficacy (after oral administration of nanoparticles), stability (at accelerated condition for 6 months), toxicity (body weight loss after oral administration of nanoparticles) and scale-up

Result
A novel docitaxel-loaded nanoparticle using membrane emulsification technique has the following properties.
- The nanoparticle with the particle size of about 20 nm, yield of 90% and encapsulation efficiency of 60% might be stable at least 2 years at room temperature, resulting from no significant differences in drug contents and appearance at 40 ℃for 6 months
- At intravenous administration, the docitaxel-loaded nanoparticle gave no significant differences in AUC and Cmax compared to commercial injectable product.
- At oral administration, the docitaxel-loaded nanoparticle increased 6.5-fold oral bioavailability compared to oral administration of docetaxel solution, and gave excellent antitumour activity, even if it had lowerantitumour activity compared to commercial injectable product. Furthermore, it induced no body weight loss compared to commercial injectable product.
- additional research : docitaxel-loaded oral nanoparticle after oral administration of SMEDDS containing curcumin, a P-glycoprotein inhibitor gave 2-fold absolute bioavailabilty of 30% compared to sing dose.
