The family Gracilariaceae has been well known as the economic agarophytes. Although they have been extensively investigated and well defined from both an anatomical and a molecular point of view, the species boundary and classification are very difficult because of the high morphological plasticity, the lack of diagnostic chatacteristics and great species diversity. The main goal of this study is to verify the systematic position of Gracilariaceae from Asia-Pacific region, and to make the DataBase for agarose-producing red algae resource. ◎ Biodiversity of Gracilariaceae from Asia-Pacific region • Collection sites: Korea, Japan, Malaysia, Philippines, China, Thailand, Australia • 22 species, 662 specimens were collected: making a distribution and flora list from Asia-Pacific region • Defined five gracilarian species from Korea: Morphology and molecular analysis • Discovered three undescribed species: Gracilaria perplexa from Japan, Hydropuntia fisheri from Thailand and Vietnam, and Gracilariopsis heteroclada from China • Detected five cryptic species: potential new species inferred from morphological and sequences characters ◎ Molecular phylogeny of Gracilariaceae • 335 total genomic DNA were extracted and analyzed rbcL sequences • We observed diagnostic characters to evaluate phylogenetic relationships of Gracilariaceae • We confirmed phylogenetic relationships and taxonomic position of 22 Gracilariaceae species • We provided basic information of cheklist and distribution for phylogenetic study in worldwide ◎ DNA barcoding of Gracilariaceae • Applying DNA barcoding approach using mitochondrial COI sequences • We identified 15 species of Gracilaria, 2 of Gracilariopsis, 5 of Hydropuntia from Asia-Pacific region • Sequence variation within Gracilariaceae: Intraspecific divergence ranged between 0 to 1.5%, and interspecific divergence was 2.8-16.9% • Species boundaries of five Korean Gracilariaceae: Intraspecific divergence ranged between 0 to 0.9%, and interspecific divergence was 9.2-16.% • Gracilaria salicornia from South-east Asia: 0-1.3% intraspecific divergence and eight haplotypes • Our study demonstrated that DNA barcoding can provide an efficient method for species identification and contribute powerfully to taxonomic, biodiersity and phylogeographic research • We showed the species boundaries within and among the members of the family Gracilariaceae ◎ GenBank of economic red algae • We made the DabaBase for Agarose-producing red algae resource • We provided the baseline for the future study of the conservation of economic seaweeds
