(개요) 최근, 산화환원(redox)조절자로서 Redox factor-1 (Ref-1)의 역할이 주목 받고 있으나, 활성산소종 (ROS)과 관련된 심장손상관련기전에 있어서는 그 역할이 아직 명확히 알려지지 않고 있다. 본 과제에서는 산화스트레스 상태의 심장에서 산화 환원기전을 제어하는 Ref-1의 역할을 규명하기 위해, ① 산화스트레스하의 심장세포 기능과 관련된 단백질의 profile 및 Ref-1과 관련 가능성 단백질을 동정하고, ② 산화스트레스 심장에서의 Ref-1 과 동정 단백질의 상호관련 기능을 검토하였다. (결과) 1) hypoxia에 의한 심장세포에서의 산화스트레스 관련단백질의 변화:Ref-1발현은 hypoxia(6시간)에 의해서 증가하였으며, 이것은 reoxygenation(2시간)에 의해 약화되었다. DJ-1 및 Prx1 발현 또한 비슷했다. ERK2/1 및 p38 MAPK의 인산화는 hypoxia에 큰 영향을 받지 않았으나, reoygenation에 의해 증가하였다. JNK인산화는 hypoxia 및 reoxygenation에 의해 증가되었다. Annexin II 발현은 hypoxia에 의해 감소하며, 이 반응은 reoygenation에 의해 약화되었다. H2O2(100 μM)는 Ref-1 및 DJ-1의 발현을 증가(10분 처리) 및 감소 (30, 60분처리)시키며, p38MAPK인산화를 증가시켰다. 2) Hypoxia에 노출된 심근세포의 단백체 변화 확인 및 profiling: ①단시간 [6 시간hypoxia/2시간 reoxygenation]노출은 다음과 같이 단백질 변화를 유도했다: 정상군과 hypoxia 군 비교(변화된 수:12 개, 동정된 수: 8개), 정상군과 reoxygenation 군 비교(변화된 수:10 개, 동정된 수: 9개), hypoxia군과 reoxygenation 군 비교(변화된 수:6개, 동정된 수: 5개)②장시간 [24 시간hypoxia/6시간 reoxygenation]노출 노출은 다음과 같이 단백질 변화를 유도했다: 정상군과 hypoxia 군 비교(변화된 수:14 개, 동정된 수: 13개), 정상군과 reoxygenation 군 비교(변화된 수:23 개, 동정된 수: 22개), hypoxia군과 reoxygenation 군 비교(변화된 수: 8개, 동정된 수: 6개) ③ hypoxia (돼지심근경색모델)에서의 단백질 변화 확인 및 profiling: 정상심근과 경색심근 비교 (변화 된수:14 개, 동정된 수: 3개) 3)hypoxia에 의한 심근세포기능변화: hypoxia및 reoxygenation에 노출한 심근세포는 정상군에 비해 생존율이 감소되었다. hypoxia (6시간)시 심근세포는 H2O2발생 증가 및 심근세포의 자발성 수축의 빈도억제를 유도했으며, reoxygenation(2시간)된 세포는 H2O2및O2-발생증가, apoptosis 발생, 심근세포자발성수축의 회복등을 유도하였다. 4)산화스트레스 유발 심근기능 이상기전에서의 Ref-1 역할: ROS 발생에 의한 산화성 세포손상 및 apoptosis를 유도하는 물질로 알려진miconazole로 자극 후 ROS에 의한 심근세포의 산화스트레스 반응에 있어서 Ref-1 역할을 검토하였다. miconazole은 심근세포 Ref-1 발현의 감소, OO2- 발생의 증가 및 apoptosis발생 증가, 심근세포의 박동 억제등을 유도했다. 이들 반응은 Ref-1 과발현에 의해 억제되었다. 이 결과는 Miconazole에 의해 유도되는 ROS/apoptosis 발생에 의한 산화스트레스성 심장기능이상에 있어서 Ref-1이 중요한 역할을 하는 단백질인 것을 시사한다. 또한 이것은 ROS관련 산화성 스트레스와 관련된 심장기능이상에 Ref-1이 중요 조절자일 가능성을 제시한다. 5)Ref-1과상호작용관련가능성단백질: 심장세포에 Ref-1을 과발현 시킨 후 변화되는 gene을 검토한 바, Prx3가 특이적으로 증가하는 것이 확인되었다. hypoxia 및 reoxygenation상태에서 Prx3는 Ref-1과 비슷한 변화를 보였고, Ref-1억제는 Prx3 변화를 억제했다. hypoxia 에 의해 발생되는 beating 및 apoptosis 변화는 Prx3의 knockdown 및 과발현에 의해 영향을 받았다. 이 결과는 산화스트레기전에 있어서 Prx3와 Ref-1의 상호작용관련성을 시사한다. (결론) 그러므로, 본 과제의 연구결과들은 산화스트레스 관련된 심혈관 질환 연구 분야 및 심혈관질환의 예방/진단/치료를 위한 분야에 새로운 정보를 제공할 것으로 사료된다.

Recently, the role of Ref-1 as a redox regulator, is interested in researches associated with cellular oxidative stress. However, a role of Ref-1 in heart exposed to oxidative stress has not been fully elucidated. This study was designed to explore the role of Ref-1 in regulation mechanism of redox system in heart exposed to oxidative stress. Cardiomyocytes exposed to hypoxia showed the increment of Ref-1 expression and this was reduced by reoxygenation treatment. Similar to Ref-1, DJ-1 and Prx I were also altered by hypoxia and reoxygenation. Hypoxia stimulation did not affect the phosphorylation of ERK2/1 and p38 MAPK but these proteins showed an increased phosphorylation by reoxygenation. JNK phosphorylation is increased in hypoxia and reoxygenation-stimulated cells. Annexin II expression was attenuated by hypoxia and this recovered by reoxygenation treatment. H2O2 (100 μM) increased Ref-1 and DJ-1 expression at treatment for 10 min and decreased their expression at treatment for 30 and 60 min. The phosphorylation of p38 MAPK was increased in H2O2-treated cells. In hypoxia exposure of cells for short time (6 h), 12 proteins were differentially expressed in comparison between hypoxia and normal cells. Of them, 8 proteins were identified. 10 proteins showed different expression in comparison between reoxygenation(2 h) and normal cells. Of them, 9 proteins were also identified. In comparison between hypoxia and reoxygenation cells, 6 proteins revealed different expression. Of them identified proteins were five. Moreover, in hypoxia exposure of cells for long time (24 h), cells exposed to hypoxia compared with normal cells have alteration of 14 proteins and 13 of these proteins were identified. When compared with normal cells, cells with reoxygenaion showed different expression in 23 of proteins and 22 proteins of them were identified. In cells with reoxygenation, 8 proteins were differentially expressed compared with cells exposed to hypoxia and 6 proteins of them were identified. Using model animal of cardiac infarction, fourteen proteins were differentially expressed in infarcted muscle compared with normal one. Of them 11 proteins showed an increase expression and 2 proteins did a decreased one. There were three identified proteins: heat shock protein 27, annexin A2, and hemoglobin alpha chain. Moreover, Cells exposed to hypoxia and reoxygenation showed the decrease of cell viability and increased generation intracellular H2O2 and superoxide anion (OO2-). Apoptosis in cells increased by reoxygenation exposure and the number of beating in cardiomyocytes was suppressed in cells exposed to hypoxia and this was restored by reoxygenation. To explore the role Ref-1 in mechanism of heat function abnormality by oxidative stress, miconazole was used as oxidative stress stimulant of cardiomyocytes. Miconazole induced the reduction of Ref-1 expression, the increment of OO2- and apotosis, and the suppression of beating number in cardiomocytes. These responses were inhibited in cells with Ref-1 overexpression. This indicates that Ref-1 may play an important role in mechanism of cardiomyocyate function abnormality mediated by ROS/apoptosis. Finally, interaction proteins with Ref-1 were investigated in cardiomyocytes using overexpresion and knockdown system. Ref-1 overexpression in cells increased Prx3 expression. Prx3 showed expression alteration similar to Ref-1 in cells exposed to hypoxia and reoxygenation. This alteration was inhibited by inhibiton of Ref-1 expression. Moreover, Beating number and apoptosis response in cells with hypoxia were affected by Prx3 knockdown and overexpression. This show that Prx 3 may interact with Ref-1 in oxidative stress-induced resposes in cells. The results from this project will provide useful information for developing a potential target for therapeutic strategies and mechanism research for prevention/treatment/care of cardiovascular disorders relative to oxidative stress.
