과거의 연구사례 및 연구 동향에서는 변성프리온 단백의 축적 자체가 신경퇴행성 프리온질환을 유발한다고 알려져 왔지만 비교적 최근의 논문들에서는 단순한 변성 프리온의 축적보다는 neuroinflammation과의 연관성을 통해 프리온 질환이 유발된다는 연구결과들이 나오고 있음. 때문에 이후 여러 연구기관에서 다양한 inflammatory 단백의 발현, 또는 체내에서의 농도 등을 모니터링하여 프리온 질환과의 연관성을 찾는 연구들을 진행하고 있음.

이번 연구에서는 마우스 변형 프리온 생물자원을 확보하고, 각종 inflammatory marker들과 프리온 질환과의 연관성 및 발병 메커니즘을 밝히고 치료 가능성을 탐색하고자 하였음. 따라서 다양한 연구를 계획하였고, 여기에 인지기능이 저하되는 시기 등을 조합하여 인지기능과의 연관성 또한 함께 관찰하고자 하였음.
실험상 계획대로 마우스들을 실험군과 정상군으로 나눠 실험군에는 Chandler/RML, 정상군에는 ICR brain homogenate를 각각 주입하여 날짜에 따른 경과를 지켜보고 주기적으로 burrowing activity를 수행하였으며, 정해진 날짜에 마우스를 sacrifice하여 샘플을 확보하였음.
샘플을 모두 확보한 후 샘플들을 이용해 WB, ELISA, IHC를 진행하였음.
실험 결과, WB를 통한 PrPSC의 관찰에서 마우스 샘플의 dpi가 증가함에 따라 PrPSC의 축적이 진행되었음을 확인 할 수 있었고, ELISA를 통해서 마우스 plasma 내의 mPTX3의 농도의 변화가 발생하였음을 확인하였음. IHC를 통해서는 실험군 마우스의 해마에 mPTX3가 분포하고 있음을 확인하였고, multiplex를 통해서 plasma내의 cytokine의 농도를 분석한 결과 23개의 cytokine 중 IL-1α, IL-1β, IL-2, IFN-γ, RANTES에서 실험군과 대조군 사이에 차이를 발견할 수 있었음. 마지막으로 Burrowing Activity를 통해 PrPSC의 축적 시기와 어느 정도 상관관계가 있다는 것을 발견 할 수 있었음.
이번 연구결과를 통해 변성프리온 단백과 inflammatory 단백 간의 연관성과 인지기능 장애와의 연관성을 발견하였음. 이를 통해 앞으로의 프리온 질환에 대한 biomarker로서의 inflammatory 단백의 역할의 재조명 및 치료제로서의 가능성을 발견하였음.

Previous research reports in prion diseases provided accumulative concrete evidences of misfolded abnormal prions in causing neurodegenerative disease. Recently, other interesting findings revealed that inflammations are being triggered in prion disease in addition to the accumulations of misfolded abnormal prion proteins. Hence, several research institutes have started in monitoring the expressions and concentrations of inflammatory proteins to find the correlation with the prion disease.
In this study, we investigated inflammatory biomarker by transmitting Chander/RML strain of abnormal prion protein (PrPSC) into mouse, which was adapted to murine through several passages in previous study. This strain was provided by KCDC through ‘Pathogenic material transfer agreement’ with original provider. Animal experiment was approved by animal test ethics committee (KCDC-058-12-2A). Burrowing activities of mice were measured periodically, and animal specimens, brain, spleen and plasma, were collected at 1, 7, 14, 56, and 105 days post inoculation (dpi) interval. Mouse brains and plasmas were tested with antibodies against inflammatory biomarkers through ELISA and immunohistochemistry. Another object of this study was to regenerate murine denatured prion resources, along with pathological studies of prion diseases in correlations with diverse inflammatory markers.
Accumulations of PrPSC in time course(dpi) were observed and confirmed by WB, and changes of mPTX3 levels in plasma was confirmed by ELISA. Dispersion of mPTX3 in the hippocampus of mouse in experimental groups was also confirmed by IHC. And IL-1α, IL-1β, IL-2, IFN-γ, RANTES among 23 cytokines showed differences between experimentally PrPSc transmitted and control groups. Lastly, correlations between PrPSc accumulation in the time course and burrowing activity was also found.
From the current study, correlations of PrPSC, inflammatory proteins and burrowing activity were evident. Hence, inflammatory proteins should be revisited as potential surrogate biomarkers for the diagnosis of prion disease and the mediators for the drug discovery.
