연구의 목적 및 내용
오늘날, 전 세계적으로 많은 사람들이 자폐증, 주의력 결핍 및 과잉 행동 장애 (이하 ADHD), 정신분열, 우울증, 정신 지체 등의 다양한 정신 질환으로 인해 고통받고 있으며, 이들 정신 질환의 유병률 또한 점차 증가하고 있는 추세이다. 본 연구자는 유전학적 분석을 통하여 정신 질환과 관련된 유전자를 발견하고, 이 유전자를 유전학적으로 제거한 결손 생쥐에 대한 생화학적, 전기생리학적, 행동학적 분석을 통해 정신 질환의 원인을 규명하며, 나아가 정신 질환에 대한 치료제를 제시하고자 한다. 본 연구자는 이를

오늘날, 전 세계적으로 많은 사람들이 자폐증, 주의력 결핍 및 과잉 행동 장애 (이하 ADHD), 정신분열, 우울증, 정신 지체 등의 다양한 정신 질환으로 인해 고통받고 있으며, 이들 정신 질환의 유병률 또한 점차 증가하고 있는 추세이다. 본 연구자는 유전학적 분석을 통하여 정신 질환과 관련된 유전자를 발견하고, 이 유전자를 유전학적으로 제거한 결손 생쥐에 대한 생화학적, 전기생리학적, 행동학적 분석을 통해 정신 질환의 원인을 규명하며, 나아가 정신 질환에 대한 치료제를 제시하고자 한다. 본 연구자는 이를 위해 Git1 결손 생쥐와 Shank2 결손 생쥐를 각각 ADHD와 자폐증의 모델 동물로 이용하여, ADHD와 자폐증의 기전을 연구하고 치료제를 제시하고자 했다.
연구결과
ADHD 환자의 GIT1 유전자의 단일염기변이(이하 SNP)에 관한 연구를 통해, GIT1 유전자의 특정 SNP이 ADHD 환자에서 더 높은 빈도로 관찰됨을 확인했다. ADHD와 연관되어있는 SNP은 GIT1 유전자의 발현을 감소시킬 수 있었으며, 이는 GIT1의 감소가 사람과 생쥐 모두에게서 ADHD를 일으키는 데에 중요한 역할을 수행함을 의미한다. 실제로 Git1 결손 생쥐는 ADHD의 주요 증상에 속하는 기억장애와 과다행동을 보였으며, 이와 같은 증상들은 ADHD의 주요 치료약인 암페타민과 메틸페니데이트에 의해 개선되었다. Git1 결손 생쥐를 이용한 전기 생리학적, 생화학적 연구를 통해 Git1 결손 생쥐는 기존에 ADHD와 관련이 되어있다고 밝혀진 도파민 회로에 이상이 없으며, 억제성 신경세포의 분화 및 신호 전달에 이상이 있는 것이 밝혀졌다. 이는 신경세포의 억제 기전에 문제가 발생했을 때에 ADHD가 발생할 수 있음을 시사한다.
Shank2 결손 생쥐가 사회성 결핍, 의사소통 결여, 상동증과 같은 자폐증의 모든 행동학적 증상을 보임을 밝힘으로써 이 생쥐가 자폐증 모델 동물로서 기능할 수 있음을 보였다. 또한 Shank2 결손 생쥐를 이용한 생화학적, 약리학적 연구를 통해 Shank2 결손 생쥐가 NMDA 수용체의 기능 저하를 보이며, 이 NMDA 수용체의 기능 저하를 회복시켰을 경우, Shank2 결손 생쥐의 사회성 결핍 증상이 회복되는 것을 보였다. 이는 신경발달장애에 해당하는 자폐증 또한 치료가 가능하며, 자폐증에 대한 치료제로서 NMDA 수용체의 기능을 향상시키는 방법론을 제시한다.
연구결과의 활용계획
Git1 결손 생쥐는 이후에도 ADHD의 기저 원인 및 신경 회로를 밝히고, ADHD의 새로운 치료제를 개발하는 데에 사용될 수 있을 것이다. 이 뿐만 아니라 ADHD와 연관성을 가지는 GIT1 유전자 상의 SNP은 ADHD를 진단하는 데에 활용될 수 있다.
Shank2 결손 생쥐 또한 이후에 자폐증의 기저 원인 및 신경 회로를 밝히고, 자폐증의 새로운 치료제를 개발하는 데에 사용될 수 있을 것으로 기대된다. 또한 Shank2 결손 생쥐의 사회성 결핍을 회복시켰던 D-cycloserine과 CDPPB는 현재 마땅한 치료법이 없는 자폐증 환자에 대한 치료제로서 제시될 수 있을 것이다.

Attention deficit hyperactivity disorder (ADHD) is a psychiatric disorder that affects ~5% of school-aged children; however, the mechanisms underlying ADHD remain largely unclear.
Autism spectrum disorder (ASD) is a group of conditions characterized by impaired social interaction and communication, and restricted and repetitive behaviours. Although ASD-associated genes are being increasingly identified and studied, further efforts are required to delineate important causal mechanisms with the potential for therapeutic application.
Here I report two mouse models, Git1 KO mice and Shank2 KO mice for ADHD and autism respectively, and show the underlying mechanisms and possible therapeutics for two neurodevelopmental disorders.
Result
Here I report a previously unidentified association between GIT1 and ADHD in humans. An intronic single-nucleotide polymorphism (SNP) in GIT1, the minor allele of which causes reduced GIT1 expression, shows a strong association with ADHD susceptibility in humans. Git1-deficient mice show ADHD-like phenotypes, with traits including hyperactivity and impaired learning and memory. Hyperactivity and impaired memory in Git1 KO mice are reversed by amphetamine and methylphenidate, psychostimulants commonly used to treat ADHD. GIT1 deficiency in mice leads to decreases in Rac1 signaling and inhibitory presynaptic input; furthermore, it shifts the neuronal excitation-inhibition balance in postsynaptic neurons toward excitation.
Moreover, regarding the study of Shank2 KO mice, I show that Shank2-mutant mice exhibit ASD-like behaviours including reduced socialinteraction, reduced social communication by ultrasonic vocalizations, and repetitive jumping. These mice show a marked decrease in NMDAR function. Direct stimulation of NMDARs with D-cycloserine and indirect stimulation of NMDARs via mGluR5 positive allosteric modulator with CDPPB normalizes NMDAR function and improves social interaction in Shank2 KO mice. Furthermore, treatment of Shank2 KO mice with a positive allosteric modulator of mGluR5, which enhances NMDAR function via mGluR5 activation, also normalizes NMDAR function and markedly enhances social interaction.
Expected Contribution
My study on GIT1 identifies a previously unknown involvement of GIT1 in human ADHD and shows that GIT1 deficiency in mice causes psychostimulant-responsive ADHD-like phenotypes. Moreover, these studies suggest excitatory-inhibitory imbalance as another mechanism for ADHD.
Moreover, the works on Shank2 suggest that reduced NMDAR function may contribute to the development of ASD-like phenotypes in Shank2 KO mice, and mGluR modulation of NMDARs offers a potential strategy to treat ASD.
