연구의 목적 및 내용
본 연구의 최종 목표는 고성능 선택특이성 생체흡착소재의 설계기술 개발에 있다.
1단계에서는 신개념 생체흡착소재의 설계를 위한 핵심요소기술을 개발하기 위하여 효소반응 경로를 설계하고 조건을 최적화시켰으며, 선택특이성 펩티드를 선정하여 유전자를 합성하고 유전자 발현조건을 최적화시켰다. 2단계에서는 부상-침전 제어가 가능한 선택특이성 흡착소재 시작품과 응용공정을 개발하기 위하여 금속추출 가능물질을 선정하고, 소재의 고정화 지지체를 선발하여 다양한 형태의 선택적 흡착소재를 제조하였으며 소재 제조 조건을

본 연구의 최종 목표는 고성능 선택특이성 생체흡착소재의 설계기술 개발에 있다.
1단계에서는 신개념 생체흡착소재의 설계를 위한 핵심요소기술을 개발하기 위하여 효소반응 경로를 설계하고 조건을 최적화시켰으며, 선택특이성 펩티드를 선정하여 유전자를 합성하고 유전자 발현조건을 최적화시켰다. 2단계에서는 부상-침전 제어가 가능한 선택특이성 흡착소재 시작품과 응용공정을 개발하기 위하여 금속추출 가능물질을 선정하고, 소재의 고정화 지지체를 선발하여 다양한 형태의 선택적 흡착소재를 제조하였으며 소재 제조 조건을 최적화하였다.
연구결과
1단계 연구 결과, 세포막에 존재하는 phospholipid를 phospholipase A2로 처리하여 다량의 음전하기(-COOH)를 갖게 함으로써 양이온성 금속종의 결합력을 획기적으로 증가시켰다. 또한 유전자 재조합을 통하여 세포막 내부에 중금속을 흡착할 수 있는 다량의 단백질을 발현하도록 제조된 소재는 원료균체에 비하여 약 50%의 Cd 흡착량 증가를 보였고, 세포 외부에 금속 나노 입자를 부착시킬 수 있는 단백질을 발현시킨 소재는 bio-TEM, FE-SEM 분석을 통하여 Pt와 Pd 나노입자를 흡착한 것을 확인하였다. 이온성 액체에 용해시킨 후 anti-solvent로 재배열하여 제조한 재배열 생체흡착소재의 경우, 조건을 최적화시켰을 때 Ru 흡착성능이 원료균체에 비하여 약 4배 증가하였다.
2단계 연구 결과, 특정 금속을 선택적으로 추출할 수 있는 물질을 선발하여 환원력을 평가하고 선발된 추출제를 고정화하기 위하여 비드형, 섬유형, 캡슐형 소재를 제조하였다. 금속환원물질인 ascorbic acid, metal chelater인 melamine, L-cystein, phosphatidylcholine, metal extraction 물질인 alamine 336, aliquat 336 등을 함유한 alginate capsule의 경우 특정 금속에 대한 선택적 흡착이 가능하다는 것을 알았다. 또한, pH, 금속의 초기 농도에 따라 금속의 흡착선택성이 변하였다. Melamine 소재는 Pd(II)에 대한 흡착 선택성이 Pt(IV)에 비하여 높았으며, L-cystein 소재는 pH 3 ~ 6, 9 이상에서 Pd(II) 선택성이 높았다. Phosphatidylcholine 소재는 pH 전 영역에서 100%에 가까운 Au(III) 회수율 보였으며 pH 4와 12에서 Pt(IV)나 Pd(II)에 대비하여 선택도가 가장 높았다. Ascorbic acid를 함유한 bead는 Au(III), Pt(IV), Pd(II) 혼합용액에서 Au(III), Pd(II), Pt(IV) 순으로 높은 선택성을 나타냈다 (KAu/Pd: 322.7, KAu/Pt: 2781.5). 또한 metal chelator인 alamine을 함유하는 alginate capsule은 흡착과 탈착을 진행하면서 침전-부상하는 현상을 관찰할 수 있었으며 흡착-침전 (24시간)보다 탈착-부상 (2시간)의 속도가 상대적으로 빨랐다.
본 연구에서 개발된 소재와 운전공정 방식은 기존 흡착소재 제조방식이 아닌 새로운 방식으로써 선택적 소재 설계에 있어서 원천기술을 확보하였다.
연구결과의 활용계획
고성능 선택특이성 생체흡착소재의 설계기술은 생체흡착 연구 분야에서 breakthrough 기술로서 인정받을 수 있을 것이며, 생체흡착소재의 본격적인 실용화를 앞당기는데 기여할 것이다. 또한 선택적 소재를 제조함에 있어 사용되는 금속추출 가능물질의 고정화 기술은 기존 생체흡착소재의 고정화, 약물전달 시스템의 구축을 위한 캡슐제조 등에 확대 활용할 수 있을 것이다.
개발된 소재는 선택적으로 금속을 회수할 수 있는 특성을 지니면서도, 흡·탈착 과정에서 비중이 변화함에 따라 소재가 부상·침전하는 특징을 갖는다. 따라서 실제적인 금속회수 공정이 단순화될 수 있으며, 반응 진행을 눈으로 확인할 수 있다. 이러한 선택성 생체흡착소재로 학술적, 기술적 가치가 높을 것으로 기대된다. 또한 생체흡착소재는 대부분 수입에 의존하는 이온교환수지와 활성탄을 대체할 수 있는 저비용의 환경소재가 될 것이다.

The final goal of this work is to design high-performance and selective biosorbents. In Phase I, core technologies for providing high sorption capacity and selectivity to biosorbents were extensively investigated. To increase the binding sites, the enzyme reaction paths were designed and optimized. Some useful peptides which have selective metal binding property were screened and fused in anchoring motifs. Metal-selective peptides bearing E. coli was cultured and conditions for expression of peptides were optimized. In addition, metal selective chelating agents and reducing agents were encapsulated within hydrogel or polymer matrixes. In Phase II, high-performance selective biosorbents were designed on the basis of outcomes from Phase I. The developed biosorbents were immobilized or granulated. It was found that floatation-settlement of some biosorbents were possible to control and a fill-and-draw type reactor was designed and optimized.
Result
From the Phase I study, the cell membrane was treated with phospholipase A2, leading to formation of negatively charged functional groups that can act as binding sites for divalent metal ions. Cell surface display of metal binding peptides resulted in enhanced Cd uptake. Furthermore, it was found that the genetically engineered E. coli had affinity toward Pt and Pd nanoparticles. Through cell dissolution with ionic liquids and precipitation of the cell components with anti-solvents, rearranged cells were constructed and showed 4 times higher uptake of Ru than the original cells.
In Phase II, selective chelating and reducing agents were extensively screened in multi-metal systems. The screened chelating and reducing agents were used for making biosorbents in the various shapes such as beads, fibers, and capsules. As a result, ascorbic acid bearing capsules showed very good selective reduction property. Au selectivity of ascorbic acid alginate capsules was evaluated to be 322.7 (KAu/Pd) and 2781.5 (KAu/Pt). Some chelating agents like melamine, L-cystein, phosphatidylcholine, alamine 336, and aliquat 336 were successfully encapsuletd inside the alginate shell and the chelator-bearing alginate capsules were proved to be excellent selective sorbents. It was also noted that the metal selectivity could be tuned according to the solution pH, which is likely because metal speciation with chloride alter. Especially, phosphatidylcholine-capsules led to almost 100% of recovery of Au from the metal mixture. It was also found that floatation-settlement of alamine-bearing alginate capsules was be able to be controlled according to the amount of metal loading. Metal free capsules were floated and as metal sorption proceeded, the capsules began to be settled down. The relationship between floatation-settlement and metal uptake was investigated. By using this property, a novel sorption process was designed and optimized. The fill-and-draw sorption process was expected to be much simpler that conventional column-type process.
Expected Contribution
The developed technology enables biosorbents to have high-performance and selective sorption characteristics and will be considered as ground-breaking idea. At the same time, the developed biosorbents and biosorption processes can accelerate the commercial practice of biosorption technology. Especially, the high-performance and selective biosorbents are expected to be used in the field of precious metal recovery from urban mines and of heavy metal removal from industrial waste solutions. The encapsulation method which was developed in this work may be used in other fields like immobilization of cells and enzymes and drug delivery. In addition, the developed biosorbents can be alternative sorbents to conventional ion exchange resins which are expensive and mainly imported.
