(1) Research Objectives Concerns about the shortage of available fossil fuels in the future, coupled with environmental problems resulting from their extraction, refining and use, have spurred increased efforts to synthesize biofuels from renewable resources. Among biofuels, butanol has recently received special attention as a renewable resource fuel alternative on the basis of a number of attractive attributes, including its comparable energy content to gasoline, low hygroscopicity, and ability to be fermented from a wide variety of carbon sources. Butanol can also be blended with gasoline or added to diesel fuel to reduce viscosity, and is compatible with existing supply infrastructures. Typically, butanol is produced by Clostridium acetobutylicum in a mixed-product fermentation, which limits the economical mass production of butanol because of its poor productivity. To improve butanol production, host strains (E. coli and C. acetobutylicum) with an improved butanol tolerance and a high production yield will be constructed by reprogramming the whole gene expression profiles of the strains using novel aritficial transcription factors and restructuring the cellular machinery of membrane and cell wall biosynthesis. Moreover, we will engineer the metabolic network to be suitable for the high-yield butanol production from low cost substrates such as corn steep liquor, molasses and cellulosic biomass. Taken together, our strategy can lead to the economical mass production of bio-butanol for commercial uses. (2) Contents For the mass production of the next-generation biofuel, bio-butanol, a synthetic genomics approach will be explored to genetically modify host microorganisms (E. coli and C. acetobutylicum) to become more efficient bio-butanol synthesizers. 1. Construction of butanol producing microorganisms with an improved butanol tolerance - by restructuring the cellular machinery of membrane and cell wall biosynthesis - by reprogramming the whole gene expression profiles of the strains using novel aritficial transcription factors 2. Construction of the high-yield butanol producing microorganisms, in which expression of genes responsible for the butanol production will be artificially regulated and optimized, directing toward the efficient production of bio-butanol - by reprogramming the metabolic newtork toward the production of butanol using novel aritficial transcription factors 3.
