Title (1st subtitle) : Genome Variation Screening Program for Swine Influenza Virus and Studies on Characterization and Pathogenesis of SIV (Seroprevalence of swine influenza in pig herds in Korea in 2009 and 2010) Seroprevalence of swine influenza in pig herds in Korea was investigated using the sera from slaughter houses in 2009 and 2010. Total 3,775sera and 3,282 sera collected in 2009 and 2010 respectively were examined for the antibodies to swine influenza virus subtype H1 and H3 using hemagglutinin inhibition test. Test results showed that 2,201 (58.3%) out of 3,775 sera from 2009 were positive against swine influenza. Specifically, 1,860 (49.3%) sera were positive for H1 subtype and 658 (17.4%) sera were positive for H3 subtype. Sera from 2010 showed slightly different results. Total 1,359 (41.4%) out of 3,282 sera were positive to swine influenza;920 sera (28%) were positive for H1 and only 27 sera (0.8%) were positive for H3. Screening for pandemic 2009 H1N1 virus was also done for the same samples. Only 512 samples (13.6%) were positive for pandemic 2009 H1N1, which we guess was the cross reaction with general swine influenza virus H1 subtype such as H1N1 or H1N2. Among 2010 sera, 950 sera (28.9%) were positive against pandemic 2009 H1N1, which was believed to be genuine positive since the seropositive reactions were decreased significantly in H1 and H3 subtype (920 and 27 sera, respectively.) Our data showed that distribution of swine influenza in Korea in pig herds was extensive and the need to take effective measure to control swine influenza. Title (2nd subtitle) : Genome Variation Screening Program for Swine Influenza Virus and Studies on Characterization and Pathogenesis of SIV (Differential effect of prior influenza infection on Alveolar Macrophage phagocytosis of Staphylococcus aureus and E scherichia coli: Involvement of Interferon-gamma production) The influenza A virus is one of the main causes of respiratory infection. Although influenza virus infection alone can result in pneumonia, secondary bacterial infection combined with the virus is the major cause of morbidity and mortality. Interestingly, while influenza infection increases susceptibility to some bacteria including Streptococcus pneumoniae (S. pneumoniae), Staphylococcus aureus (S. aureus), and H aemophilus influenzae (H . influenzae) , other bacteria such as E scherichia coli (E. coli) and K lebsiella pneumonia (K . pneumonia) are not associated with influenza infection. The reason for this discrepancy is not known. In this study, we find that prior influenza virus infection inhibits the murine alveolar macrophage phagocytosis of S. aureus but not E. coli. Here, we elucidate the mechanism for this inhibition, showing that prior influenza virus infection strongly increases Interferon gamma (IFN-γ) production. Furthermore, we show that IFN-γ differentially affects alveolar macrophages phagocytosis of S. aureus and E . coli. Our study explains how influenza virus infection increases susceptibility to only some bacteria, such as S. aureus, and provides evidence that IFN-γ might be a promising target for protecting the human population from secondary bacterial infection by influenza. Title (3nd subtitle) : Genome Variation Screening Program for Swine Influenza Virus and Studies on Characterization and Pathogenesis of SIV (Involvement of NF-κB in changes of IFN-γ-induced CIITA/MHC-II and iNOS expression by influenza virus in macrophages) Type II interferon (IFN-γ) plays an important role in defense against viral infection. Although this cytokine is found during influenza virus infection, it seems to have no protective function against the virus, and the reasons for this are not clear. To determine how the influenza virus overcomes the antiviral effects of IFN-γ, we examined the effect of A/Puerto-Rico/8/34 (H1N1) (PR8) infection on the expression of various IFN-γ inducible genes involved in defense against virus infection. The results showed that PR8 selectively affects IFN-γ induced MHC-II in both the murine macrophage-like cell line, Raw264.7, and in primary alveolar macrophages. Infection of IFN-γ treated macrophages with PR8 resulted in decreased expression of CIITA/MHC-II. These changes correlate with activation of NF-kB but not with JAK/STAT signaling. The data indicate one possible mechanism underlying the ineffectiveness of IFN-γ against influenza virus, and suggest that NF-κB may be a promising target for anti-influenza drugs.
