연구결과:
1) Lipogenesis와 지질방울 생성에 관여하는 114개의 세포 단백질로 구성된 siRNA library를 제작하고 신규 세포 타겟을 발굴하기 위한 screening system 구축
2) siRNA screening의 결과 C형간염바이러스 증식 억제 효과를 나타내는 10개의 신규 타겟 유전자를 선별하였음. 이들 신규 유전자중 C형간염바이러스의 복제 및 방출에 현저한 감소효과를 보이는 4개의 유전자 SCD1, FDFT1, ARFRP1, METTL7A 등의 기능에 대한 연구를 지속하고자 siRNA-mediated knockdown, membrane fractional assay, 신규단백질의 저해제의 처리 및 면역형광염색법등의 다양한 실험적 기법을 통해 C형간염바이러스의 증식과정에 관여하는 기전을 심도 있게 분석하였음.
3) C형간염바이러스 NS4B 단백질과 결합하여 증식을 조절하는 METTL7A
METTL7A 단백질의 발현을 siRNA knockdwon의 방법을 사용하여 억제하였을 때 C형간염바이러스 복제 및 증식이 현저히 감소하였음. METTL7A의 바이러스 증식에 있어 기능을 연구하기 위해 바이러스 단백질들과의 결합을 조사하였을 때, NS4B와의 상호작용을 통해 C형간염바이러스의 증식 조절에 관여하는 것으로 판단됨.
4) C형간염바이러스 증식에 필수적인 세포 단백질 FDFT1
siRNA mediated knockdown 및 저해제의 처리를 통해, FDFT1이 C형 간염바이러스의 life cycle 중 복제 및 바이러스 입자의 방출과정 조절에 중요한 역할을 수행함을 관찰함.
5) C형간염바이러스의 복제 복합체의 일원으로 복제를 조절하는 세포단백질 SCD1
SCD1은 siRNA mediated knockdown 및 저해제의 처리에 의해 바이러스가 감염된 세포와 subgenomic replicon 세포에서 C형간염바이러스의 증식과정에 관여하는 것을 확인하였으며,SCD1의 대사산물인 oleic acid와 palmitoleic acid는 C형간염바이러스의 복제과정에 중요한 산물임을 입증하였음. SCD1이 C형간염바이러스의 복제 및 방출의 감소를 유발하는 기작을 탐색하기 위해, 면역침강법과 면역형광염색법을 수행하여 확인한 결과, NS3, NS4B, NS5A, NS5B과 상호작용됨을 입증하였음. membrane fraction assay를 수행하여 SCD1의 localization의 분석한 결과, SCD1은 lipid raft에서 비구조단백질과 co-localization을 통해 replication complex의 형성에 관여할 것으로 사료됨. SCD1의 활성억제제를 처리하였을 때 바이러스 증식이 현저히 억제됨이 관찰되어 치료제의 타겟으로의 활용가능성이 큼.
6) C형간염바이러스의 복제 및 조합에 중요한 LD 생성에 관여하는 세포단백질 ARFRP1
ARFRP1 단백질 발현 저해 연구를 통해 C형간염바이러스의 복제 및 방출과정에 필수적인 세포단백질임을 입증하였고, ARFRP1은 C형간염바이러스 NS5A와 core 단백질과 결합하는 것으로 확인되었음. ARFRP1은 C형간염바이러스 감염이후에 간세포의 LD 생성에 중요한 역할을 담당하여, ARFRP1 단백질 발현 저해는 LD 형성을 억제하고 NS5A가 지질방울과 colocalization을 저해함을 규명함.
7) C형간염바이러스의 조합에 중요한 LD의 융합에 관여하는 세포단백질 SNAP23
C형간염바이러스의 감염은 지질방울의 융합을 위해, SNAP23의 재분포를 유도하는 현상을 면역형광염색법을 통해 관찰하였으며, 간세포내의 ARFRP1 단백질 양을 제한하였을 때 C형간염 바이러스에 의해 생성되는 지질방울 내로의 SNAP23의 이동이 억제되었음. C형간염바이러스 감염 후, ARFRP1은 adaptor 단백질인 SNAP23을 활성화시켜 지질방울의 융합을 유도함.
8) C형간염바이러스 단백질의 tm모화 (sumoylation)
본 과제에서는 바이러스 단백질의 스모화가 C형간염바이러스의 복제 및 증식에 미치는 기작을 밝혀내고자 하였음. 스모화 기작에 필수적인 UBC9 단백질의 knockdown을 통해, 스모화가 C형간염바이러스의 증식과정에 필수적인 현상임을 확인하였음. 스모화와 C형간염바이러스의 복제기작과의 연관성을 구체적으로 밝혀내기 위해, 면역침강법을 수행하여 스모화되는 C형간염바이러스 단백질을 탐색한 결과, C형간염바이러스가 감염된 간세포 및 subgenomic replicon세포에서 비구조단백질 NS5A가 스모화되었음. NS5A의 스모화에 의해 NS5A의 유비퀴틴화가 억제되어 단백질의 분해과정으로부터 보호되었으며, NS5A의 스모화는 NS5A의 인산화와 상호의존적인 작용을 통해 NS5A의 안정화가 유지되었음. 또한, NS5A의 스모화는 C형간염바이러스의 비구조단백질인 NS5B와의 결합에 관여하며, C형간염바이러스의 복제과정에서 replication complex에서 NS5B와 NS5A와의 상호작용에 중요한 역할을 함을 규명함.
본 과제를 통해, C형간염바이러스의 증식에 관여하는 10여개의 신규 타겟을 발굴하였으며, 이들 타겟의 발현 및 활성을 조절함으로써 C형간염바이러스의 증식을 조절하는 기전을 연구하여 바이러스와 사람세포와의 상호작용을 관찰하였음. 또한 스모화되는 바이러스단백질의 발굴과 C형간염바이러스의 증식과정과 숙주세포의 스모화 기작과의 연관성을 연구하였음. 일련의 연구를 통하여 높은 돌연변이 때문에 완치기 어려웠던 C형간염바이러스의 치료를 위해, 숙주세포단백질을 표적으로 하는 효과적인 치료제의 개발을 위한 기반 기술을 제공하였으며, 새로운 치료 기술 및 치료제의 개발을 통한 국민건강의 증진과 경제발전에 기여할 것으로 사료됨.

Results:
1) Establishment of siRNA screening system for novel cellular factors of lipogenesis and lipid droplets involved in HCV propagation.
2) We screened a siRNA library targeting 114 genes that was involved in lipogenesis and lipid droplets in HCVcc-infected cells. From 114 siRNA pools, 10 pools were identified as candidate hits. These pools were retested in a secondary screen. Of these, functional roles of SCD1, FDFT1, ARFRP1, METTL7A in HCV propagation were further investigated by siRNA-mediated knock down, membrane fraction assay, inhibitor treatments, and immunofluorescence assays.
3) We demonstrated that siRNA-mediated depletion of METTL7A significantly inhibited HCV replication and propagation. We showed that HCV modulated METTL7A via NS4B protein for its own propagation.
4) We showed that siRNA-mediated knockdown of FDFT1 significantly inhibited HCV replication and egress of infectious HCV virion. We further verified that FDFT1 inhibitor impaired HCV assembly and release.
5) By employing small interfering RNA (siRNA) library screening in cell culture-grown HCV (HCVcc)-infected Huh7.5 cells, we identified stearoyl-CoA desaturase 1 (SCD1) as a host factor required for HCV propagation. SCD1 is the rate limiting enzyme catalyzing the biosynthesis of monounsaturated fatty acids oleate and palmitoleate. Here we demonstrated that knockdown of SCD1 attenuated HCV replication up to 60% and extracellular HCV RNA level was decreased ~80% in HCV infected cells. The treatment of 3-[4-(2-chloro-5-fluorophenoxy)-1-piperidinyl]-6-(5-methyl-1,3,4-oxadiazol-2-y l)-pyridazine, an inhibitor of SCD1, inhibited HCV replication in both subgenomic replicon cells and in HCVcc-infected Huh7.5 cells. Moreover, the treatment of oleate and palmitoleate resurrected HCV replication in SCD1 knockdown cells. We showed that SCD1 was coimmunoprecipitated with HCV nonstructural proteins, including NS3, NS4B, NS5A, and NS5B, but not core protein, in HEK293T cells. We further demonstrated that SCD1 was colocalized with both dsRNA and HCV nonstructural proteins, indicating that HCV may recruit SCD1 to the RNA replication complex.
6) HCV exploits lipid metabolism for its propagation. HCV replication and assembly occur in close proximity to lipid droplets (LDs). To elucidate the functional roles of lipids in HCV propagation, we have screened the small interfering RNAs (siRNA) targeting genes involved in lipid metabolism and LD formation in HCVcc)infected cells. In this study, we have selected and characterized the gene encoding ADP-ribosylation factor-related protein 1 (ARFRP1). ARFRP1 is essential for LD growth and is involved in the regulation of lipolysis. We demonstrated that downregulation of ARFRP1 by RNA interference significantly inhibited HCV replication in both HCV subgenomic replicon cells and HCVcc-infected cells. Moreover, ARFRP1 interacted with both HCV core and nonstructural 5A (NS5A) proteins. ARFRP1 interacted with NS5A derived from both genotype 1b and 2a via domain I of NS5A and this interaction was required for HCV replication. We further showed that HCV-induced accumulation of LDs was reduced in ARFRP1-knockdown cells.
7) We demonstrated that synaptosomal-associated protein 23 (SNAP23), a downstream effector of ARFRP1 which was known to be required for LD assembly, was also required for HCV virion production. HCV may regulate ARFRP1 via SNAP23 for fusion of LDs and thus ARFRP1 may be a therapeutic target for HCV.
8) Viruses exploit cellular SUMOylation machinery to favor their own propagation. We show that NS5A is a target protein of small ubiquitin-like modifier and is SUMOylated at lysine residue 348. SUMOylation and phosphorylation of NS5A are interdependent, indicating that there is a crosstalk between posttranslational modifications. SUMOylation increased protein stability of NS5A and was required for protein interaction with NS5B.
These data imply that SUMO modification may contribute to HCV replication. Indeed, HCV replication level was significantly reduced in SUMO-defective replicon cells. We provide evidence for the first time that HCV exploits cellular SUMO modification system to favor its own replication.
In conclusion, we identified ten novel cellular factors involved in HCV propagation. Our data provide new insights into the innovate technology for development of highly effective therapeutic agents for HCV. This technology will contribute to the economic development in Korea as well as the improvement of public health worldwide.
