본 연구단은 당화와 발효가 한 reactor에서 수행되는, 바이오에탄올 생산라인의 차세대 기술인 통합공정개발을 위하여 고온, 고 에탄올 내성 및 바이오매스의 전처리 부산물로 저해제인퍼퓨랄에 대한 내성, 고농도 당에서 발효 기능을 가진 균주개발을 시도하였다.
1단계에서 분자유전학(1세부;BT) 및 전사체 분석 기술(3세부:IT)을 융합하여 최적의 균주를 개발하고자 연구를 수행하였다.
2단계에서는 다음의 연구를 수행하였다.
1) 고온 강화, 목질계 부산물인 아세트산 내성, 내성 통합균주 개발. 활성산소 신호전달 기작규명
본 연구단은 당화와 발효가 한 reactor에서 수행되는, 바이오에탄올 생산라인의 차세대 기술인 통합공정개발을 위하여 고온, 고 에탄올 내성 및 바이오매스의 전처리 부산물로 저해제인퍼퓨랄에 대한 내성, 고농도 당에서 발효 기능을 가진 균주개발을 시도하였다.
1단계에서 분자유전학(1세부;BT) 및 전사체 분석 기술(3세부:IT)을 융합하여 최적의 균주를 개발하고자 연구를 수행하였다.
2단계에서는 다음의 연구를 수행하였다.
1) 고온 강화, 목질계 부산물인 아세트산 내성, 내성 통합균주 개발. 활성산소 신호전달 기작규명
2) 추후 당화시스템을 겸비하는 최적의 통합공정용 균주 개발을 위한 고분비 목질계 분해시스템
1세부: 통합공정용 균주개발 기술로는 gTME 기술, Transposon-중개 변이체의 library screen 및 SGKO (single-gene knock-out) library screen, 순환형 카셋트를 이용한 gene knock-out과 유전자 과벌현 기술을 활용하여 아래와 같이 균주를 개발함(표). 이들의 성장능 및 발효능은 각 조건하에서 wild 대비 평균 15% 이상 증가하여 좋은 수율을 보임. 개발된 균주의 일부는 전사체 분석(3세부와의 융합)을 통하여 내성 관련 인자 다수 발굴.
1. 바이오에탄올 생산라인의 통합 공정용 균주개발에 필요한내성인자 발굴
1-1: 고온

2. 다중내성 신규 효모 설계

2-2: 고온 + 고에탄올 + furfural
- 20 % glucose, 43℃ 발효시 control2 대비 발효 성능 30% 증가

3. 자연형 균주제작용 분자유전학 도구 개발
3-1: 자연형 균주 작업을 위한 분자유전학 도구 확보 (pScDAL22-25)
- 과발현과 유전자 결손 (gene knockout)을 동시에 수행
- 유전자의 연속적인 결손 가능

4. 내성 기작 규명

- 두 signal pathway가 다를 것으로 짐작
4-2: 아세트산 내성 균주
- 전사체 분석을 통한 8종의 아세트산 내성인자 발굴
4-3: 고온 내성 균주
- 전사체 분석을 통한 13종의 고온 내성인자 발굴
- Dfg5 deletion에 의한 내성기작 model 제시
4-4: 세포내 활성산소 산호전달 기작 규명
- Hydrogen oeroxide에 의한 HOG pathway 활성 기작 구명
2세부: 효모의 대사체 분석을 위한 대사체 분석방법 최적화하여 quenching 방법으로 fast filtration 방법을 선정하였으며, 추출 용매로 acetonitrile/water mixture를 선정하였음.
• 에탄올 및 고온, furfural 내성 변이주의 대사적 표현형 규명 및 내성 인자 발굴하여 glucose-6-phosphate, trehalose, proline, fatty acids, lactic acid 등의 내성 관련 인자를 찾음.
• 에탄올 및 고온, furfural 내성 변이주의 대사적 표현형 규명을 통하여 내성 인자와 관련된 pentose phosphate pathway와 trehalose 합성경로, 아미노산 및 지방산 생합성 경로가 내성 메카니즘에 큰 변화가 있는 것을 확인함.
• 에탄올 및 고온, furfural 내성 변이주의 글로벌 탄소대사 특성 규명
• 다중 내성 변이주의 고온 조건, 고에탄올 조건, 고 furfural 조건에서의 글로벌 탄소 대사 특성 규명
• 3세부: 바이오에탄올은 저탄소 바이오에탄올은 저탄소녹색성장을 위한 핵심 에너지원으로서 다양한 형태로 자동차연료에 첨가되어 대기 내 이산화탄소를 저감시킬 수 있는 물질임
• 자연계에 풍부한 섬유소를 이용하여 상업적으로 가능한 바이오에탄올을 생산하기 위해서는 고에탄올・고온・고삼투압・발효독소에 내성을 갖는 발효균주의 개발이 필수적임
• 이에 본 연구에서는 (1) 현재 개발된 각 내성인자를 보유한 내성균주의 발효능력을 평가하였고, (2) 글로벌최적화기술을 통하여 각 균주에 적합한 맞춤형 발효공정을 개발하였으며
(3) 정량적으로 에탄올 농도 15%와 생산성 1.4 g/L-hr, 이론수율의 80% 이상의 발효공정을 개발하였음

The multiple steps of bioethanol processes including pretreatment, saccharification, fermentation are one of the main reasons for high operating cost for cellulosic ethanol.
Therefore, it is highly demanding to integrate enzymatic hydrolysis and ethanol fermentation processes and the intergrated process are so called "Consolidated Bioprocessing (CBP). In order to develop an ethanol-fermenting yeast to be used in the CBP, the primary causes for the sensitivity and resistance to high ethanol, inhibitors such as furfural and hydroxymethyl furfural (HMF) and high temperature are necessary.
Sub-project 1
The purpose of this research is to develop strain(s) ideal for bioethanol production line proper for consolidated bioprocess (CBP).
At the first stage, we developed thermo-, ethanol-, osmo-, furfural-tolerant strains was performed using molecular biology technology (sub-project 1) and bioinformatics (sub-project 3). Highly efficient cellulolytic system for combined bioprocess was also studied by eco-technology (sub-project 2).
At the second stage, we fortified thermo-tolerant strains and developed acetic acid-tolerant strains and stress tolerant factor-integrated strains. We also clarify the signal transduction pathway activated by hydrogen peroxide.Development of strains proper for CBP has been achieved by using the gTME technology, screen of transposon-mediated mutagenized library and SGKO (single gene knockout) library, gene overexpression and sequential gene knockoutwith a reusable cassette. The developed strains exhibited increased growth rate and fermentation capacity more than 15% compared to wild-type control. RNA-seq analysis was conducted (with sub-project 3 group) to identify heat and acetic acid tolerance factors. We also revealed the signal transduction pathway activated by hydrogen peroxide.
Sub-project 2
In this study, metabolomics approch will be undertaken for the phenotyping of Saccharomyces cerevisiare mutants generated in the first research division of this project for the possible increase of such resistances.
In the first step, the optimization of metabolome sample analysis will be conducted. The optimal quenching or rapid filtration method will be developed for yeast, and also optimal metabolite extraction solvent mixture will be selected based on the unbaiased and quantitative recovery of intracellular metabolites of yeast cells. Yeast mutants will be cultivated in different conditions in the aspect of ethanol concentration, furfural concentration and cultivation temperature, and the cells were recovered and their metabolites and transcripts were sampled and analyzed for the metabolomics and transcriptomics analyses. These information will be fedback to the first division of the current project for molecular genetic manipulation of the mutant. The manipulated mutant will be further analyzed by the metabolomics and the transcriptomics, and the information will be handed to the first division for the strain improvement and enforcement of such resistances to high temperature, inhibitors and ethanol.
In the subcontract with Ewha Womans Univ., in the first year, a genome-scale metabolic network model of S. cerevisiae will be reconstructed and its prediction capacity will be evaluated. In the second and third years of the project, the metabolic targets essential for construction of robust ethanologenic yeast will be examined with the model.
The microbial metabolomics emerging area, and it is not at the mature stage yet. In particular, the metabolomics has not been applied to elucidate the cellular and molecular causes and mechanism for the resistance to fufural, ethanol and high temperature. The results from the integrative study using metabolomiccs and transcriptomics will promote the efficiency of finding the possible mechanisms and responsible genes for rendering the resistance to yeast will be great boost for the strain development study of consolidated bioprocessing.
Sub-project 3
The final goal of this study is to evaluate the fermentation performances of pre-constructed tolerant alcohol yeasts and to develop an efficient process for bioethanol production by global optimization with more than 15% ethanol content and 1.4 g/L-hr productivity. Osmotic stress caused by high sugar and ethanol concentrations is one of the major reasons for reduction of bioethanol productivity during yeast fermentation. To overcome this hurdle, global transcription machinery engineering (gTME) by introducing SPT15 variants encoding S. cerevisiae TATA-binding proteins was applied and S. cerevisiae ETS3 was selected to possess enhanced ethanol producing ability under high osmotic pressure. Its fermentation properties were evaluated in batch and fed-batch cultures. Among various culture types and conditions including using various glucose concentration and aeration, a batch culture of ETS3 with 300g/L glucose at oxygen-limited condition resulted in 22.2 g/L dry cell mass, 98.1 g/L ethanol concentration and 1.82 g/L-h ethanol productivity which were about 13–.16% increases relative to those of the S. cerevisiae BY4741 host strain. A micro-aerobic condition elevated ethanol productivity to 2.49 g/L-h without significant changes of other fermentation parameters. In a fed-batch fermentation with 400 g/L glucose, ETS3 produced up to 151.7 g/L ethanol with a maximum ethanol yield of 0.402 g / g and showed a 23% increase in dry cell mass and 16% increase in ethanol concentration and productivity in comparison to the host strain.
