(1) 연구개발의 목표
지구 온난화 및 세계 에너지 수요의 급속한 증가와 함께 화석에너지의 고갈에 대비하여 새로운 대체 에너지의 개발과 이용확대가 더욱 절실한 상황이다. 특히, 신ㆍ재생에너지 중 태양에너지를 식물체에 고정시켜 이를 에너지원으로 전환하는 지속가능한 바이오 에너지는 현재 세계 에너지 시장의 15%를 점유하고 있고 향후 2035년경에는 전체 시장의 23%에 이를 것으로 전망되고 있다. 그 중 바이오 부탄올은 최근 가솔린과 유사한 정도의 높은 에너지 밀도를 가질 뿐만 아니라, 흡습성 및 휘발성이 낮아 바이오 디젤 및

따라서 본 연구팀은 부탄올의 경제적인 대량생산을 위한 고효율 생산 균주를 개발하여 가솔린 및 유용 화합물의 전구물질을 경제성있게 상업적으로 대체할 수 있는 부탄올을 공급하고자 한다.
(2) 연구 내용
본 연구진은 고효율 생산 균주(E. coli와 Clostridium)를 개발하여 차세대 연료로 각광받고 있는 부탄올을 대량생산하고자 한다. 이를 위한 최적화된 바이오 부탄올 생산 균주 구축을 위해 미생물의 유전자 발현 시스템을 조절하여 바이오 부탄올 생산을 위한 세포내 대사회로를 재설계하고, 새로운 DNA scaffold 시스템을 개발하여 바이오 부탄올 생합성 관련 효소작용의 최적화 및 바이오 부탄올 생산 대사회로의 최적화를 통해 고효율 바이오 부탄올 생산 균주를 구축하여 바이오 부탄올을 산업적으로 대량 생산하고자 한다.
① 효과적인 바이오 부탄올 생산을 위한 대장균 및 Clostridium 내 대사회로 재설계
- 바이오 부탄올 생산 유전자의 효율적 발현시스템 구축을 위해 바이오 부탄올 생산 관련 유전자들을 operon으로 구축
- 바이오 부탄올 생산 관련 유전자들의 발현 순서 최적화.
- 인공회로제어 시스템을 이용하여 바이오 부탄올 생산 대사회로를 재설계하여 기존의 한계를 뛰어넘는 바이오 부탄올 생산회로 구축.
② DNA scaffold system을 이용한 바이오 부탄올 생산 대사회로 최적화
- 바이오 부탄올 생합성에 관련된 유전자에 DNA scaffold 기술을 도입하여 대사회로 상에 필요한 각각의 효소간의 반응을 최적화하고 중간대사산물의 diffusion에 의한 반응속도저하의 최소화 및 생산효율을 증가시키고 독성 중간대사산물의 축적을 막아 바이오 부탄올의 생산을 최적화.

Concerns about the shortage of available fossil fuels in future, coupled with environmental problems resulting from their production and use, have spurred increased efforts to synthesize biofuels from renewable resources. Among biofuels, butanol has recently received a special attention because of its great advantages over other biofuels such as ethanol. Butanol has higher energy density and lower hygroscopicity than ethanol. In addition, it is less corrosive and is easier to blend with gasoline or diesel fuels. Typically, butanol is produced by Clostridium acetobutylicum in a mixed-product fermentation, which limits the economical mass production of butanol because of its poor productivity. Moreover, the cellular and metabolic reactions in biological systems of a living organism are orchestrated as complex networks of individual reactions which seem like precisely designed and ordered for the survival of the living organism. However, the actual reaction depends on the concentration of substrates, simple-diffusion of metabolites and enzymes causes the inefficiency of metabolic reactions by lowering the local concentration around the enzymes. Furthermore, release of intermediates with toxicity to the host cell by simple-diffusion also inhibits the activity of many cellular functions.
Herein we will engineer a novel butanol synthesis pathway into Escherichia coli that has a fast growth rate, allowing for a flexible and economical process design for the large-scale economical production of butanol. Moreover, host strains (E. coli and C. acetobutylicum) with a high production yield will be constructed by reprogramming the whole gene expression profiles of the strains using novel artificial transcription factors and DNA scaffold systems. A DNA scaffold system can promote the proximity of metabolic enzymes and increase the local concentration of intermediates turn out to be the most promising approach.

For the mass production of the next-generation biofuel, bio-butanol, a synthetic genomics approach will be explored to genetically modify host microorganisms (E. coli and C. acetobutylicum) to become more efficient bio-butanol synthesizers. To produce butanol in E. coli, a recombinant butanol production pathway will be introduced and expressed in E. coli. To further improve butanol production, host strains (E. coli and C. acetobutylicum) with a high production yield will be constructed by reprogramming the whole gene expression profiles of the strains using novel artificial transcription factors.
Furthermore, optimization of butanol synthetic pathway using the DNA scaffold system which can promote the proximity of metabolic enzymes and increase the local concentration of intermediates. Taken together, our strategy can lead to the economical mass production of bio-butanol for commercial uses.
