1. Procurement of polar biological samples and metagenomes from extreme environments
Polar organisms have developed their own survival mechanisms particularly in the extreme environment, which is not very common in most of general temperate organisms. They possess variety of novel materials and unique genetic resources, which could be utilized for BT industry. In order to study the usefulness of polar organisms and to obtain genetic resources, we have procured the biological samples from extreme environment. We collected about 180 biological samples from surrounding Antarctic King Sejong and Arctic Dasan stations. Also, 88 samples were procured from Kamchatka and lake Baikal, Russia. They were preserved in deep-freezer and/or in liquid nitrogen tank, and by freeze-drying method. To investigate the enzymes for use in industry and to gain the genomic information of useful enzymes, using plasmid vector, metagenome libraries were constructed from samples obtained from polar seawater and soil.
2. Preparation of tissue mass culture of polar plants
Natural compounds are esteemed new drug candidates because they are easily searchable matters than synthetic compounds, and for development of new pharmaceuticals, researchers can estimate their safety and validity. Thus, using the natural compounds they can shorten the development term and cost. Those natural compounds have been obtained from the plants cultivated in ground or collected by herbalists. Recently, the compounds can also be obtained from the plant tissues which were cultivated by cell culture technique. In this study, we could obtain the tissue cultures of 8 mosses and 2 lichen species, which is able to produce some biologically active compounds such as antioxidant, antimicrobial, antidiabetic, anticancer and antiwrinkle materials.
3. Isolation of microorganisms from Lake Baikal in Russia
(A) Isolation of psychrophilic organism by new cultivation method, I-tip from Baikal sponges
Using standard cultivation methods resulted in the isolation of 16 specIes belonging to 3 taxonomic groups: Betaproteobacteria, Firmicute and GanJIDaproteobacteria. Using the I-tip method, 34 species were isolated belonging to 5 taxonomic groups: Actinobacteria, Alphaproteobacteria, Betaproteobacteria, Firmicute, and GanJIDaproteobacteria. The number of isolates from the I-tip method accounted for 71% of the listed taxonomic groups by pyrosequencing and 42% of the taxonomic groups from conventional methods.
(B) Analysis of microbial community in Lake Baikal
The microbial community distribution is easily flexible according to environmental variation. The microbial community were different by thermocline and depth and classified cleary between lake-water and sponge samples. The microbial community was not similarity according to the season. The actinomycetal community has higher similarity between each samples, we suggest it is likely more useful to understand the environmental ecology. The retrieved band from lake-water sample were identified with uncultured bacterium. We believe that it is important to isolate and research about uncultured bacteria for understanding the ecological meaning of Lake Baikal. Therefore, we consider that it will be important to understand the other functional gene by molecular methods and exploit to cultivate bacteria containing these gene for comprehending the rapidly changing ecosystem.
(C) Isolation and Characteristics of hallophilic bacteria from Lake Alginskoe
Total 43 species bacteria belonging to 3 taxonomy groups (Proteobacteria, Actinobacteria, Firmicutes) were isolated from Lake Alginskoe and most isolates were halophilic bacteria.
Enzyme activities, amylase, cellulase, lipase and protease of isolated bacteria from Lake Alginskoe were analyzed. At the results, twenty two isolates had one or more enzyme activities at least. And four isolates had three enzyme activities.
The growth rate of isolates in different salinity were analyzed. Cupriavidus pauculus showed high growth rate at 0% salinity. But Cupriavidus pauculus showed the growth rate of decreased significantly with the increase of salinity. On the other hand, Bacillus sc;ifensis showed faster growth rate than other speCIes in highly saline condition. Enzyme activity measurement results, Brachybacterium paraconglomeratum, Bacillus aryabhattai had three enzyme activities. These bacteria increased the growth rate in 0-7% of salinity, the growth rate was faster than the other species.
4. Analyses of photosynthetic activities and growth of polar mosses
Plant stress contains both destructive and constructive elements and IS a selection factor as well as a driving force for improved resistance and adaptive evolution. Photoinhibition of photosynthesis in a changed physiology of the organization from drought condition is characterized by a reduction III the quantum yield of photochemistry and a decrease in chlorophyll fluorescence. In addition, expression of LEA proteins has been shown to be induced by dehydration and ABA, which is synthesized during dehydration stress. However, their precise function is unknown, but they are assumed to protect cellular or molecular structures from the damaging effects of water loss. Here, we observed change that the maximum quantum efficiency of photosystem II primary photochemistry and the identification of the specific roles of the genes expressed during the development of environmental stress tolerance in a polar moss species Aulacomnium turgidum and the model moss species Physcomitrella patens.
5. Procurement and utilization of genetic resources from polar organisms
We analysed the DNA sequence and function study of small plasmid from cold-adapted Pseudomonas sp. strain MCI (KOPRI 26573) and carried out the acquisition and analysis of cold-active chitinase, new gene for cold-active catechol 1,2-dioxygenase, and cold-active protease. In order to produce the cold-active enzyme, vector-host system was constructed. Also, we analysed the productivity, characterization, and commercial applications of R-Pr02l717.
Obtaining polyketide synthase genes from polar lichen species Polar areas are largely unexplored and contain many undescribed organisms. Plants, animals, lichens, and microorganisms are managing their lives together by competition and cooperation to withstand the severe environment. Polar organisms have evolved to SurvIve severe environments, including extremely cold temperatures, dry climate, and concentrated ultraviolet rays. Such harsh conditions compel the consideration of the existence of specific agents or factors that enable them to maintain such a unique physiology. Considering this hypothesis, we performed experiments on identifying cell growth factors and their working mechanisms to develop potentially valuable polar bioresources, including enzymes and growth factprs. We could screen 15 PKS genes, encoding a critical enzyme of polyketide biosynthesis, from polar lichen species Stereocaulon alpinum and analyzed their gene expression.
6. Genetic transformation of polar plant genes
Plants, animals, lichens, and microorganisms from polar regions are managing their lives together by competition and cooperation to withstand the severe environment. Polar organisms have evolved to survive severe environments, including extremely cold temperatures, dry climate, and concentrated ultraviolet rays. Such harsh conditions compel the consideration of the existence of specific agents or factors that enable them to maintain such a unique physiology. Considering this hypothesis, we performed experiments on identifying cell growth factors and their working mechanisms to develop potentially valuable polar bioresources and corresponding genes. Our researches are focused on functional study on genes from antarctic organisms using genetic approach. For this aim, transformants analysis using model plant Arabidopsis thaliana and chloroplast transformation method were established and utilized to investigate gene functions from polar moss species. Among the trasformants, Arabidopsis plants overexpressing genes of Polytrichastrum alpinum, such as MBF1c, FKBP12, GST6, and MnSOD, have shown enhanced tolerace to environmental stresses.
7. Alzheimer's disease modulatory effects of a natural compound from Antarctic lichen
Alzheimer's disease (AD) is an age-related disorder characterized by deposition of neurotoxic froms of Abeta and degeneration of neurons. The production of Abeta requires sequential clesvages of APP by β-secretase (BACE1) and Υ-secretase. The causes of Abeta accumulation in the majority of AD patients remain unclear, but may include age-related increases in oxidative stress. A screening of a active materials of polar origin with modulatory ability on amyloidogenesis leads us to identify several compounds altering BACE1 expression and activity. Theses reduce protein levels of BACE1 and BACE1 promoter activity. We also demonstrated that in vivo treatment with Ramalin, as a active material of polar origin, improved cognitive functions and reduces BACE1 mRNA and Aβ in the brains of AD mice. Consequently, our studies suggest that Ramalin may be efficacious for the treatment of AD.
8. The immunomodulatory effect and anti-inflammatory effect of polar lichen extracts
Our data demonstrate that HO-1 and PI3K are important factors that modulate NO prouction, iNOS protein expression and autophagy-related protein expression in Rapamycin-induced Raw 264. 7 cells. Ramalin treatment inhibited autophagy through suppressing NO production.
Ramalin suppressed TNF-α-induced production of intracellular reactive oxygen species (ROS), PADI4 expression, and phosphorylation of p38, ERK, and JNK. Moreover inhibition of PADI4 expression by small interfering RNA or the PADI4-specific inhibitor C1-amidine markedly attenuated TNF-α-induced activation of NK-kB and AP-1 and VCAM-1 expression in VSMCs.
To investigate the apoptotic effect of ramalin on cancer and to understand the underlying mechanisms, tow different kinds of human breast cancer cell lines were used in this study, Estrogen receptor (ER)-positive MCF-7 (with wild type p53) and ERα-negative MDA-MB-231 (with mutated p53). The results revealed that cell viability is decresed and apoptotic cells identified by annexin V/PI analysis are increased after ramalin treatment of both breast cancer cell lines. Further studies indicated that treatment of the tow breast cancer cells with ramalin activates the mitochondrial apoptotic pathway via release of cytochrome c and AIF from the mitochondria to the cytosol due to the up-regulation of bax and the down-regulation of bcl-2. In addition, ramalin induced apoptosis though caspase activation in MCF-7 and MDA-MB-231 cells. Moreover, our data demonstrate that Ramalin decreased the expreSSIOn of NLRP3 m LPS-induced Raw 264. 7 cells. Taken together, the present data indicate that components and extracts from polar biological resources have anti -inflammatory effect, anti-cancer effect and inhibited autophagy. These new finding might provide a new therapeutic strategy for treating pathological diseases.
9. Structure elucidations of Secondary Metabolites from Antarctic Bio-resources
Isolation of various secondary metabolites from relatively untapped plants from Kamchatka area, employing bioassay-guide fractionation investigation
1) Conducting various in vitro assays targeting anti-neuroprotective and anti-inflanunatory agents
2) Isolation of secondary metabolites from extracts of plants from Kamchatka
3) Isolation of secondary metabolites from plant organisms by various chromatographic methods
4) Structure elucidation of isolated metabolites by spectroscopic and chemical methods
5) Evaluation of biological activities of identified metabolites by a number of biochemical methods
Our data will contribute to the bio-active secondary metabolites depository and provide lead compounds for the developments of drugs and/or other functional agents
10. Screening and characterization of useful biomaterial
The Korea Polar Research Institute (KOPRI) has assembled a culture collection of cold-adapted bacterial strains from both the Arctic and Antarctic, which include cold-active chitinase-, protease-, and oxygenase-producers. To identify excellent cold-active enzyme-producers among them, significant strains were selected in advance according to their relative activities and were subsequently re-examined for their extracellular enzyme activity on each proper plates supplemented with corresponding substrates at vanous temperatures. This rapid and direct screening method permitted the selection of a small group of cold-adapted bacterial strains, which showed enzymatic activities at temperatures ranging between 5-15℃. The finally selected strains were identified and classified based on their 16S rRNA DNA sequences.

Characteristic analysis of cold-active lipase For the screemng of useful enzyme, several lipase-producing bacteria was tested and P AMC 21333 was selected. Lipase activities using synthetic substrates were compared in various conditions. When the P AMC 21333 was cultivated in 1% mixture of olive oli/Gum arabic solution, more than 130 UIL of lipase activity was obtained. Lipase was purified using butyl and Q-column, but clear one-band was not obtained.
Psychrobacter okhotskensis P AMC21333 was selected as the candidate strain producing a psychrophilic lipase useful for industrial application. The lipase coding gene was cloned in genomic library screening and sequencing. The lipase enzyme for strain P AMC21333, named as Lip21333, The recombinant enzyme showed Ca-independent activity. In substrate specificity, the enzyme prefer to hydrolyze p-nitrophenyl palmitate with psychrophilic activity.
11. Production modeling
By using production modeling, 53 UIL of protease originated from polar region was produced. The amount of the produced P-21653 by fed-batch culture was 3.76 gIL, 4 times higher than that by batch culture. Preussia sp. was used for the production of Preussia-C, a antioxidant agent, at various culture conditions.
12. Cryoprotective properties of exopolysaccharide (P-21653) produced by the Antarctic bacterium Pseudoalteromonas arctiw (KOPRI21653)
P-21653 polysaccharide have high viscosity and higher M.W. over 2 M Da. Acid treatment (O.1M TFA) was effective method to produce L. M. polysaccharide with high solubility and low viscosity and enhance the cryoprotective effect of P-21653 by decreasing their molecular weight. L. M. P-21653 (1.8x105 Da.l pennitted reduction of freeze-thaw induced Hemolysis of Red blood cells(REC). And the method using L.W. P-21653 as a cryoprotectant is a simple, quick inexpensive way to make frozen red blood cells (RECs) for an extended period. To evaluate the biochemical qualities of frozen-REC with L.M. P-21653, the levels of were tested. the levels of ATP and 2,3-DPG were maintained close to the levels of non-frozen-RBC. The cryopreservation of RBCs for clinical use can be categorized based on the techniques used to achieve biologic stability and ensure a viable state after long-term storage. L.M. P-21653 acts as an percipients to protect red blood cells during long term storage (5 month over).
P-21653 is effective to reduce repeated freezing and thawing damage to Mammalian cells. 1-2% L.M. P-21653 combination with 1% DMSO may be considered to effective cryoprotectant for human stem cell and mammalian cell cryopreservation. And its combination may be a better alternative standard 10% DMSO protocol in primary human stem cell cryopreservation. The biochemical characteristics of L.M. P-21653 reflect that this compound may be developed as a useful cryoprotectant for use in medical and biological applications
