B형과 D형은 바이러스 표면항원이 거의 동일하여 현존하는 B형 백신으로 일차적 예방은 가능하다 D형은 B형과 동반감염 또는 B형 감염자에게 중복감염 되어 간암으로의 전이율이 현저히 증가된다. 아직 D형에 대한 백신은 개발되어 있지 않기 때문에 이미 B형에 감염된 3억 5천만 이상 B형 보유자의 D형 중복감염에 대한 예방 대책이 시급하다. 이에 본 연구의 목적은 단 1회 접종으로 科(family)가 서로 다른 2개의 간염바이러스를평생 면역할 수 있는 최첨단 백신을 개발하는 것이다. 개발 전략은 B형의 표면항원과 D형의 델타항원을 혼합 백신용 항원으로 특수 제조하는 것이다. 현존하는 B형 백신은 안정성의 문제(혈장에서 분리된 백신)와 면역항원성의 저하(효모에서 생산된 백신) 등으로 인하여 수 차례 반복 접종해야하는 불편과 이로 인한 비경제적인 결점이 있다. 세계에서 가장 큰 간염백신 시장을 고려할 때 상기 결점이 보완되고, 하나의 혼합백신으로 2개의 간염바이러스를 예방할 수 있는 차세대 백신 개발이 절실히 요구된다.
이러한 연구목표를 수행하기 위하여 각각의 간염바이러스 항원 및 B형 간염바이러스 표면항원과 D형 간염바이러스 델타 항원을 동시에 발현하는 많은 벡터를 제조하였고 이들 벡터를 이용하여 stable cell 및 여러 개의 재조합 바이러스(recombinant viruses)를 제조하였다. 재조합 바이러스는 Sf9 곤충세포를 감염시켜 발현이 높은 바이러스를 다시 선발하였고 세포에 감염 시킨후 발현된 항원을 부리하여 다각도로 특성을 규명하였다.
본 연구를 위하여 20개 이상의 재조합 바이러스를 제조하였고, 항원발현 분석결과 SHDAg-HBsAg, SHDAg-HBsAg (90-175), LHDAg-HBsAg, LHDAg-PreS2, HBsAg-SHDAg, HBsAg-LHDAg 등의 혼합백신용 항원발현이 우수하였다. 그러나 항원의 상당부분이 insoluble하였다. 이중에서 LHDAg-PreS2와 HBsAg-LHDAg을 분리 정제하여 mice에 면역주사 한 후 immunogenicity를 조사한바 HBsAg에 대한 면역반응은 미약하였으나 HDAg에 대한 면역반응은 상당히 우수하였다. 그러나 혼합백신의 용도를 충조기키기 위하여 HDAg의 길이를 조절한 mutant 형태의 항원을 만들어 면역원성을 조사하는 등 보강연구를 수행중이다. 결론적으로 본 연구는 하나의 혼합 백신으로 두 개의 서로 다른 간염바이러스를 동시에 예방할 수 있는 증거를 제시하고 있다.

Among these, B, C. and D cause chronic hepatitis and hepatocellular carcinoma. More than 400 million people are currently infected with hepatitis viruses worldwide and these viruses kill at least one million people annually.
To date, only A and B vaccines are available. Hepatitis D virus (HDV) shares hepatitis B virus surface antigen (HBsAg) with hepatitis B virus (HBV) as an envelope. The primary infection of HDV can be prevented by HBV vaccine. However, secondary infection of HDV from HBV carrier cannot be prevented because HDV either coinfects with HBV or superinfects with HBV carrier. HDV causes fulminant hepatitis and hepatocellular carcinoma. The goal of this project was to develope a divalent vaccine against both HBV and HDV. For this purpose, a chimera antigen consisting both HBsAg and hepatitis delta antigen(HDAg) was designed and expressed in insect and mammalian cells. Since current HBV vaccine has some problems including safety and low immunogenicity, a divalent vaccine is required to improve the quality of HBV vaccine and to prevent HDV infection from HBV carrier.
To perform this project successfully, we have constructed many plasmids expressing either hepatitis antigen alone or chimera proteins of viral antigen. Using these plasmids, we established stable cell lines and generated more than 20 recombinant baculoviruses. All recombinant proteins were expressed in Sf9 cells and analyzed by Coomassie blue staining and Western blot. The results showed that the following chimera proteins were expressed highly: SHDAg-HBsAg, SHDAg-HBsAg (90-175), LHDAg-HBsAg, LHDAg-PreS2, HBsAg-SHDAg, HBsAg-LHDAg. However, most of these chimera proteins were insoluble.
Among these LHDAg-PreS2 and HBsAg-LHDAg were chosen for further studies. We therefore partially purified these proteins and mice were immunized to exarrune immunogenicity. Most mice respond weakly to HBsAg but strongly to HDAg. To improve the immunity against HBsAg so as to function efficiently as a divalent vaccine, we are currently modifying chimera proteins. Taken together, these data provide an evidence showing a divalent vaccine against two different hepatitis viruses.
