- Th2 및 Th1 분화조절능 평가
․ 더덕의 각 분획별 추출물을 Balb/c mice에서 얻은 비장세포에 전처리 한 후, T세포의 mitogen인 concanavalin A 및 phytohemaglutinin (PHA)를 처리하여 T세포의 증식을 유도하였다. 그리고 그 상층액을 취하여 T세포가 분비한 IL-4와 IFN-γ의 양을 ELISA 법을 이용하여 정량하였다. 그 결과 더덕의 열수추출물과 Hexane 분획에서 IL-4와 IFN-γ의 양이 증가하였다. 따라서 더덕의 열수추출물과 He

- Th2 및 Th1 분화조절능 평가
․ 더덕의 각 분획별 추출물을 Balb/c mice에서 얻은 비장세포에 전처리 한 후, T세포의 mitogen인 concanavalin A 및 phytohemaglutinin (PHA)를 처리하여 T세포의 증식을 유도하였다. 그리고 그 상층액을 취하여 T세포가 분비한 IL-4와 IFN-γ의 양을 ELISA 법을 이용하여 정량하였다. 그 결과 더덕의 열수추출물과 Hexane 분획에서 IL-4와 IFN-γ의 양이 증가하였다. 따라서 더덕의 열수추출물과 Hexane 분획은 T세포의 Th2/Th1 분화조절능이 있을 것으로 생각된다. 또한 더덕의 지상부/지하부 추출물을 이용하여 T세포 분화 유도에 중요한 역할을 하는 대식세포에서 Th2/Th1반응에 관여하는 유전자인 COX-2와 IL-12의 발현 정도를 RT-PCR 방법을 통해 확인하였다.
그 결과 지상부의 경우 COX-2와 IL-12의 mRNA 발현이 모두 억제되었으며, 지하부는 IL-12의 mRNA 발현만을 억제하였다.
- 비만세포/대식세포 기능 억제 효과 및 더덕 효능성분의 항알러지/항아토피 효과 기전연구
․ 더덕의 각 분획별 추출물의 효능을 비만세포에 처리 후 분비되는
β-hexosaminidase의 양을 β-hexosaminidase 정량법을 이용하여 정량하였다. 그 결과 더덕의 각각의 분획들이 β-hexosaminidase의 방출을 유의적으로 억제하는 것을 확인하였다. 또한 더덕의 각 분획별 추출물을 대식세포에 처리 후 FITC-dextran 탐식 정도를 Flow cytometry를 이용하여 정량한 결과 더덕 부탄올 분획의 사포닌 성분이 대식세포의 탐식능을 강력히 억제하였다. 그리고 대식세포에서 분비되는 사이토카인중 알러지/아토피 반응에 관여하는 TNF-α와 PGE2의 생성능을 ELISA 법을 이용해 측정하였으며, 더덕의 부탄올 분획내 사포인 성분에 의해 강력히 억제된다는 것을 확인하였다.
- 동물실험 및 예비임상 실험 연구
․ 이전의 연구를 바탕으로 피부염에 대한 동물 실험 및 예비임상 실험을 진행 한 결과, PGE2에 의한 염증 반응 및 인체를 대상으로 한 임상 실험에서 더덕 유래의 사포닌 성분에 대한 효과를 확인할 수 있었다. 더덕 유래 사포닌 성분은 ICR mice에서의 경구 투여와 인체에 대한 피부 처리에서 효과를 보였으며, 이러한 것은 피부를 통한 처리뿐만 아니라 경구 투여를 통해서도 효과가 있음을 나타낸다. 본 연구팀은 이러한 결과들을 통해서 더덕 유래의 사포닌 분획이 PGE2 유래의 염증 반응 및 알러지/아토피 피부염에 대해 효과가 있음을 확인하였고, 인체에 대한 부작용이 없음 또한 확인하였다.

- 생리활성물질 추출 및 단일성분 분리
․ 각 용매별로 추출물을 제조하여 각각의 수율을 확인하고, 가장 수율이 좋은 열수추출물을 이용하여 다당체를 분리하였다. 또한 HPLC를 사용한 다당체의 구성성분과 특성검토를 통해 더덕유래 다당체는 fructose와 glucose로 이루어짐을 확인하였다. 각 추출물별 NO억제능 실험을 통해 최적 활성분획이 부탄올 추출물임을 확인하였다.
- 생리활성물질 분리정제 기술개발
․ 인삼 saponin 추출방법을 응용하여 생리활성물질인 crude saponin를 추출 및 분리하였으며, 함량분석 및 HPLC를 사용하여 구성성분이 ginsenoside-Re와 ginsenoside-Rg1과 유사한 성분임을 확인하였다. 또한 각 layer 별 NO억제능을 확인하고, 가장 효능이 좋은 cascade butanol saponin 활성분획의 구성성분과 활성물질 함량 분석을 하였으며, 활성물질의 특성을 검토하였다. 그 결과 crude saponin의 구성성분과 다른 물질로 구성되어 있음을 확인하였다.
- 생리활성물질 분리공정 최적화
․ 가장 좋은 활성분획의 제조 및 추출수율 최적조건 확립을 위해 시간 및 voulme에 변화를 주어 단계별 최적화 조건을 검토하여 최적 조건을 확인하였으며, 그 결과로 얻어진 추출물의 함량을 확인하였다. 그 결과 시간이 증가함에 따라 비례적으로 추출수율이 증가하였지만, 효율적인 면에서는 크게 차이가 나지 않았다. Volume 역시 양의 증가에 따라 추출수율도 가하였지만, 그 정도는 크게 증가하지 않았다. 또한 생리활성물질 분리시 일부가 소실됨을 확인하였으며 이에 따라 추출 과정에서 소실되는 생리활성물질의 양을 줄이는 방안을 마련할 필요가 있었다.
• 협동과제
- 더덕 유래 사포닌 함유 향장제품 개발
․ 더덕추출물 사포닌 분획을 함유하는 기능성 화장품 제형 설계에 필요한 물성을 평가하고 이를 기반으로 기능성 화장품 제형 3종을 개발하였다. 개발된 기능성 화장품 제형으로는 크림제, 에센스제, 및 팩제로 이 제형들 모두 물을 함유하고 있어 물에 대한 친화성이 큰 더덕추출물 사포닌 분획울 용이하게 탑재할 수 있었다. 또한 이들 3가지 기능성 화장품 제형의 실험실적 처방을 수립하고 스케일업 시험 생산을 거쳐 제조공정을 수립하였다.

Part 1
1. Evaluation of functional activation of T and antigen presenting cells The molecular immunological effects of fractions from Codonopsis lanceolata on cell viability, the production of cytokines and cytotoxic molecules as well as their mechanism were evaluated using splenic T cells. Fractions fdid not block normal cell viability of splenic T lymphocytes. Hot water extract and hexane fraction strongly up-regulated IL-4 and IFN-gamma production. In addition, Fractions from odonopsis lanceolata negatively modulated the expression of IL-12 and COX-2, which are know to be critical factors in atopic dermatitis symptoms.
2. Evaluation of functional activation of mast cells and macrophages The effect of fractions from Codonopsis lanceolata on the production of histamine, a critical pathophysiological factor in atopic symptom, was examined using hexosaminidase assay. Of fractions, saponin fraction showed significant inhibition of beta-hexosaminidase release. In addition, fractions strongly suppressed FITC-dextran uptake, assessed by flow cytometry. Saponin fraction from Codonopsis lanceolata strongly suppressed PGE2 and TNF-alpha production as assessed by ELISA. Finally, the fractions also modulated cell-cell adhesion seen in skin inflammatory conditions.
3. Evaluation of anti-atopic activities using animal model and pre-clinical study The in vivo efficacies of saponin fraction from Codonopsis lanceolata were evaluated using animal and human volunteer. Saponin fraction nicely suppressed skin rash induced by allergen DNCB. Furthermore, this fraction also inhibited in vivo ear edema-induced by arachidonic acid, a strong inflammatory inducer. In agreement with previous results, this fraction also normalized atopic symptoms of atopic dermatitis patient without skin irritation phenomena. Interestingly, saponin fraction also showed anti-stress effect. Thus this fraction recovered higher serum levels of GOT and GPT, parameters seen under stress-induced liver damage conditions. Therefore, our data suggest that saponin fraction from Codonopsis lanceolata can be developed as potent and valuable anti-atopic dermatitis biomaterial with anti-inflammatory and anti-stress properties.

1. Extraction and purification of biological response modifier
The extraction yields were examind with various solvents. Polysaccharide was purified from hot-water extract showing the best extraction yield. By using HPLC, it was confirmed that polysaccharide consisted of glucose and fructose. The optimal active fraction was known to be the butanol extract by the experiment of the inhibition activity against NO production.
2. Development of purification technology
The crude saponin was extracted and purified with the modified extraction method of ginseng saponin. It was confirmed that the constituents were ginsenoside-Re and ginsenoside-Rg1 by HPLC. And the inhibition activity against NO production was studied with each fraction. The cascade butanol fraction of crude saponin showed the best inhibition activity. The compositions and contents of this fraction were examined, resulting in containing the crude saponin compositions and other compounds.
3. Optimization of purification process
In order to establish the optimal conditions for the active fraction and the extraction yield, extraction time and volume were varied. The content was examined from the established optimal conditions. The extraction yield was proportionally increased with increase in time. However, there was no significant difference in yield with respect to efficiency. The extraction yield was also increased a little with increase in volume with showing no great difference. There was a loss in purification process of biological response modifier. Thus, it was necessary to find the way to reduce the loss in purification process.

1. Preformulatory evaluation of the extract of Codonopsis lanceolata The extract of Codonopsis lanceolata was evaluated for aqueous solubility, time-stability profile together with chromatographic determination method. The possibility of being incorporated into the solid lipid nanoparticles was also examined. The extract of Codonopsis lanceolata was highly hydrophilic and this indicated that water-bearing formulation is suitable for the extract. Although the extract is hydrophilc material, it can be incorporated into the lipid nanoparticles.
2. Development of semisolid cosmetic dosage forms and evaluation of release properties
The extract of Codonopsis lanceolata was added to four different cosmetic dosage forms such as O/W cream, W/O cream, hydrophilc base, and lipophilic base. Then the release property of the extract was evaluated to select suitable bases for use in the treatment of anti-allergic/atopic dermatitis. O/Wcream formulation and hydrophilic base showed excellent release behavior of the extract of Codonopsis lanceolata.
3. Formulation determination and establishement of production procedures of cosmetic dosage forms
Formulation determination and production procedures of three types of cosmetic dosage forms were performed such as cream, essence and pack. All the formulation contained aqueous media, thus the extract of Codonopsis lanceolata could be stably incorporated in the cosmetic dosage forms.
