○ 연구결과
주요목재변색균 은 강한 방미효력을 보여주었다. 특히 실험된 Ophiostoma 속에 속한 균중에서 O. ulmi가 가장 활발한 생장을 나타내었다. 목초액의 방미효력은 몇몇의 균주에 대하여 강한 효력을 보인반면 죽초액은 모든 균주에 매우 뚜렷한 방부능력을 나타내었다. 이들 목초액 및 죽초액은 GC-MS분석에 의하여 2,6 dimethoxy phenol, dehydroacetic acid, 2,3,5-trimethoxytoluene성분을 함유하였다.

목재변색균들은 주로 목재의 표면에 포자 혹은 균사의 형태로 집락을 형성하며 주로 셀룰로오즈 층보다는 세포벽의 리그닌집적층을 주로 분해 파괴하였다. 이들이 세포벽을 집적분해하기도 하지만 주로 벽공의 마르고의 파괴에 의하여 이웃세포로 생장하였다.
silver bead + silver thiosulfate complex는 매우 안정적이며 열에 대하여 저항이 매우 높고 방미 표력 또한 매우 높아서 페인트, 섬유, 고무 등과 같이 물질에도 방부제로서 활용이 기대된다. chitosan-glutaldehyde-silver ions membranes은 매우 높은 항균력을 보여주어 목재 방미제는 물론 방미효력을 지닌 코팅제나 음식포장지 등의 높은 활용성을 보여주었다.
목재변색균에 대한 바실러스균의 길항물질을 판명하였으며 이를 부분 정제하였다. 이들은 3300cm-1 주위의 넓은 밴드는 NH그룹으로 여겨지고 amid I 과 amid II의 밴드로 여겨진다.
사용된 Ophiostoma 균주에 대하여 ITS 염기서열을 비교분석한 결과 이들은 6개의 그룹을 분류 할 수 있었다.

Most deterioration and staining problems in wood are caused by fungi. Ophiostoma sp. is a major genera to produce staining in wood. In this study, natural resource materials, biological agents and polymers synthesized by natural resources were chosen for the study of the antifungal activity against wood staining fungi, Ophiostoma sp. The application of natural resources and biological control agents I wood preservation has not yet been reported. The purpose of our study was to evaluate the action of natural resources, bio-control agents and polymers against sapstaining fungi and inhibition mechanism of products were discussed briefly.
"Chikusaku-eki" and "Mokusaku-eki" are natural resources and acidic liquid by-products of bamboo and broad leaved treescharcoal burner. These products contain more than 200 ingredients, including phenols, poly phenols and acetic acids. These by-products were tested for their fungicidal activity against sapstaining fungi. These extracts were more effective against sapstaining fungi at minimum concentrations (0.10-1.0 %) used in 2 % MEA medium. Three Chikusaku-eki (CE-I, CE-II and CE-III) and 2 Mokusaku-eki (ME-I, ME-II) extracts were tested against 4 sapstaining fungal samples to evaluate the inhibition range on sapstaining fungal growth. Ophiostoma flexuosum, Ophiostoma tetropii, Ophiostoma narcissi and Ophiostoma ips were the sapstaining fungi used in this study against bamboo and wood extracts. The chloroform fractions of these extracts were analyzed by GC-MS and both the extracts contained 2,6-dimethoxy phenol, dehydroacetic acid and 2,3,5-trimethoxytoluene. Results revealed that compounds of "Chikusaku-eki" and "Mokusaku-eki" had remarkable fungal growth inhibitory effect at lower concentrations. The Chikusaku-eki and Mokusaku-eki has both antifungal and antioxidant properties and has a potential for use as natural preservative in wood industries. These extracts are easily available, inexpensive and non-toxic to environment, which validates its applications in wood industries. Wood slice tests were displayed the effective role of both extracts in laboratory level. Colonization and penetration of fungal hyphae into wood elements was evaluated by scanning electron microscope.
Another study was carried out to evaluate the exact role of phenolic compounds existing in wood vinegar towards the sapstaining fungal growth inhibition. To find out the clear mechanism behind antifungal activity of phenolic compounds in wood vinegar, we neutralized (pH 7) wood vinegar with NaOH and hot water extracts of Pinus ensiflora and Quercus serratasaw dusts were used in the media instead of sterilized distilled water to enhance the fungal growth inhibition property of neutralized wood vinegar. Ophiostoma polonicum, O. ips, O. flexuosum, O. narcissi and O. tetropii were the sapstaining fungi used in this study against neutralized wood vinegar, extracts of Pinus densiflora and Quercus serrata. Neutralized wood vinegar was active only above 2.5 % used. GC-MS analysis was performed to identify the phenolic compounds and other ingredients in both neutralized wood vinegar and acidic wood vinegar by chloroform extraction method. Eleven major compounds were identified in acidic wood vinegar and 7 major compounds in neutralized wood vinegar. The acidic wood vinegar contains 72.59 % phenolic compounds and neutralized wood vinegar contains 57.92 % phenolic compounds. The Scanning Electron Microscopy image of fungal growth on wood samples showed the phenomenon of fungal penetration into wood elements. Pinus densiflora was used as wood element for SEM analysis.
The antifungal activity of Bacillus subtilis EF 617317 and Bacillus licheniformis EF 617325 were demonstrated against sapstaining fungal cultures Ophiostoma flexuosum, Ophiostoma tetropii, Ophiostoma narcissi and Ophiostoma ips in both in vitro and in vivo conditions. The active supernatant fractions of 7 days old Bacillus subtilis and Bacillus licheniformis cultures inhibited the growth of sapstaining fungi in laboratory experiments. Four antifungal peptides were partially purified from Bacillus licheniformis and three antifungal peptides were
partially purified from Bacillus subtilis. The partial purification was done using sephadex gel filtration chromatography and the compounds masses were determined by LC-MS spectrometry as m/z 1023, 1038, 1066 and 1081 in B. licheniformis and m/z 1036, 1058, and 1090 in B. subtilis. FT-IR was performed to confirm the surface chemical bonds of lipoproteins. According to our results, the partially purified lipopeptides may belong to surfactin and iturin family. Thermostability and pH stability of lipopeptides were analyzed. The lipopeptides were thermostable and inhibited sapstaining fungal growth in in vitro analysis as methanol-soluble fractions. In vivo analysis of antifungal activity of lipopeptides on wood was conducted in laboratory. In addition, fungal inhibition potential of extracts on surface and internal part of wood samples was analyzed by Scanning Electron Microscopy.
Chitosan membrane was prepared by mixing sterilized chitosan with gluteraldehyde solution then the membrane was dried at 37 ºC at 48 hrs. Silve ions were embedded with membrane at different concentrations to produce antifungal activity. Wood staining fungi Ophiostoma flexuosum, O. tetropii, O. polonicum and O. ips were tested against Chi-Gla-Ag membrane for antifungal activity. Surface morphology of Chitosan, Chitosan-Gluteraldehyde and Chi-Gla-Ag membranes were studied by scanning electron microscopy. The quality and structure of membranes were analyzed by Fourier Transform-Infrared Spectroscopy. Thermostability of the membranes was confirmed by Thermo Gravimetric Analysis. pH 5 was maintained in chitosan and all concentrations of silver ion solutions. SEM results show that the uniform appearance of membrane and addition of gluteraldehyde increases the membrane thickness, thermostability and water insolubility of membrane. The antifungal activity of chitosan membrane increased with increase in concentration of silver ions. In addition, different concentrations of pure chitosan were tested against wood fungi to understand the contribution of chitosan towards the antifungal activity and different concentrations of silver ions were also added with various concentrations of chitosan to demonstrate the antifungal activity by radial diffusion assay method, individually.
