○ 연구결과
고정화 lipase를 이용한 효소전환기술을 활용하여 compound K 지방산 에스터 화합물을 개발하기위하여, 유기용매에서의 효소 반응조건을 결정하여 대량 반응을 진행하였다. HPLC를 이용하여 분리 정제한 시료를 LC-MS, MS/MS, 1H-NMR 스펙트럼을 통하여 분석하여 에스터 화합물 생성 반응이 정상적으로 진행되었음을 추정할 수 있었으나, 정확한 구조 확인을 위해서 추가적인 연구를 진행하고 있다. 분리 정제한 compound K 지방산 에스터 화합물의 피부생리활성 및 안전성 평가를 시행
○ 연구결과
고정화 lipase를 이용한 효소전환기술을 활용하여 compound K 지방산 에스터 화합물을 개발하기위하여, 유기용매에서의 효소 반응조건을 결정하여 대량 반응을 진행하였다. HPLC를 이용하여 분리 정제한 시료를 LC-MS, MS/MS, 1H-NMR 스펙트럼을 통하여 분석하여 에스터 화합물 생성 반응이 정상적으로 진행되었음을 추정할 수 있었으나, 정확한 구조 확인을 위해서 추가적인 연구를 진행하고 있다. 분리 정제한 compound K 지방산 에스터 화합물의 피부생리활성 및 안전성 평가를 시행한 결과 사람의 피부섬유아세포의 성장을 촉진시키고, collagen 합성을 촉진시키는 등 compound K 자체보다 훨씬 우수한 활성을 보였으며, 안자극성과 피부자극성이 거의 없는 화장품 소재로서 안전한 물질인 것으로 평가되었다. 주름개선 화장품 소재로서의 가능성을 확인하고, 기능성화장품 처방 개발을 통하여 시제품을 제작하였다.

The esterification reactions of a ginsenoside, compound K, with fatty acids, such as oleic acid and octanoic acid, were carried out using the enzyme lipase GF. The esterification reactions of a ginsenoside, compound K, with fatty acids, such as oleic acid and octanoic acid, were carried out using the enzyme lipase GF. In the optimized condition the esterification was carried out using lipase GF(4%, w/v), with compound K(0.2%, w/v) and octanoic acid(6.4%, v/v)(molar ratio, 1:125) in an organic solvent medium (isooctane:DMSO(31.25:1)), stirred at 1,150rpm at 65℃ for 5 days in the presence of 4Å molecular sieves. The reaction mixtures were analyzed by both TLC and HPLC. The reaction products were isolated with preparative HPLC into two major fractions (EK-001 and EK-002) and analyzed by LC-MS, MS/MS and 1H-NMR. One product of compound K octanoyl ester was proved as a compound with molecular weight of 748, probably esterified with a single molecule of octanoic acid. And the other product with a molecular weight of 738 was also proved to be 10 smaller than expected, yet probably modified during enzymatic esterification reaction. The chemical structures of these products eeds to be confirmed by further analysis.
The absorption of gensenosides themselves into body has been found to be poor in the range of 0.1-3.7%. However, the most of residual forms of these ginsenosides in the body of mice fed with gensenosides were found as esterified with fatty acids. The fraction containing products through enzymatic esterification reaction, Octanoyl Esterified Compound K (OECK), accelerated the growth of dermal fibroblasts, which contribute to the maintenance of connective tissue integrity, in a dose-dependent manner. OECK also increased the production of type 1 collagen, major component of ECM. In line with these results, the phosphorylations of p42/44 ERK and p38 mitogen-activated protein kinase, AKT and CREB were markedly increased by OECK, suggesting that the enhancement of collagen production may be linked to the activation of multiple intracellular signaling cascades. In addition, OECK has been demonstrated safe as a cosmetics ingredients by OECD guidelines of eye and skin irritation assay.
