본 연구과제는 SERS 나노입자 프로브, 생체 라만분광영상기술, 생체 영상 진단기술, 체외진단기술의 네 분야를 주된 연구 분야로 함
첫째, SERS 나노입자 프로브는 본 연구의 다른 세부과제에서 진행 중인 SERS 나노입자 플랫폼을 바탕으로 하며, 이는 다중진단이 가능한 여러 SERS 신호 특성을 가짐. 이 나노입자는 생체 및 체외진단을 위해 특정 바이오마커(단백질, 핵산, 생물학적 process 등)에 대해 표적화되며 복합영상을 위해 자성 또는 방사성동위원소로 표지함. 이어 생체적용에 최적화되도록 생체적합성 개선법이 연구되고

첫째, SERS 나노입자 프로브는 본 연구의 다른 세부과제에서 진행 중인 SERS 나노입자 플랫폼을 바탕으로 하며, 이는 다중진단이 가능한 여러 SERS 신호 특성을 가짐. 이 나노입자는 생체 및 체외진단을 위해 특정 바이오마커(단백질, 핵산, 생물학적 process 등)에 대해 표적화되며 복합영상을 위해 자성 또는 방사성동위원소로 표지함. 이어 생체적용에 최적화되도록 생체적합성 개선법이 연구되고 향후 생체적용이 가능하도록 약동학, 안전성, 독성 자료를 확보하였음.
둘째, 생체 라만분광영상기술을 구현하기 위하여 상용 장비를 도입한 생체 영상시스템 구현, 라만분광장치를 장착한 소동물 생체 라만영상기술 구현, 인체 적용을 위한 프로토타입의 개념 작성을 위한 연구를 진행하였음.
셋째, 생체 SERS 나노입자 영상 진단기술은 본 과제의 핵심으로서, 동물질환모델에서 표적화 SERS 나노입자를 이용한 라만분광영상의 유효성을 평가하였음. 또한 소동물 MRI 및 PET 등 복합영상과 함께 구현할 수 있는 기술을 연구하였음.
넷째, 체외진단기술은 SERS 나노입자를 이용한 체외진단의 개념증명 연구를 진행하고, 이어 여러 바이오마커에 대한 다중 체외진단기법을 구현할 수 있도록 연구하였음. 이어 실제 환자 질환 조직에서 SERS 나노입자 응용 진단기술을 시행, 유효성까지 평가하였음. 최종적으로 multiplex 조직진단 키트를 구현하여 임상실용화 기반을 구축하였음.
최종적으로, 주요 암에 대한 특성 SERS 신호를 가지는 특정 바이오마커 표적 SERS 나노입자를 개발하여 SERS 라만분광영상을 개발하고, 이들의 PET 또는 MRI 복합영상을 구현하였음. 도입 및 개발된 생체 라만분광영 상장비를 이용한 생체 SERS 영상을 시행하여 진단 유효성을 확립하고 이를 인체 적용하기 위한 각종 기초 전임상 자료를 확보하였으며, 진단기술은 multiplex 조직진단 키트로 임상실용화를 추구하였음.

2. Contents
(1) Part I: Development of surface-enhanced Raman spectroscopy tagging material and measurement technique for in vivo imaging
- SERS Dots previously developed by our group are photostable, quantitative, and able to be used in multiplex diagnosis
- SERS NP are tagged with specific targeting materials (e.g. antibody, aptamer, peptide) and also labeled with magnetic, fluorescent, or radioactive materials for multimodal imaging using PET/MRI/optical imaging. Size, shape, and surface-modulation are tried to enhance efficacy and bioavailability.
Endoscopy-guided in vivo detection is one of the final goals of the research.
- Laser-equipped intravital confocal microscopy is introduced and equipped with Raman spectroscopic devices to make in vivo Raman imaging system. Furthermore, conceptual prototype of in vivo Raman imaging devices for human application is designed and suggested.
(2) Part II: Development of in vivo Raman spectroscopy and application to diagnosis
- The diagnostic efficacy of biomarker-targeting SERS NP with in vivo Raman imaging is assessed in animal disease models, with aids of multimodal imaging.
- For the human application, bioavailability is improved and data on pharmacokinetics, safety, and toxicity are acquired.
- Multiplex tissue diagnostic techniques are developed and applied in patients' tissues.
Diagnostic efficacy is also assessed with comparison of conventional methods. The final goal is to make multiplex tissue-diagnosis kits for one-step diagnosis of multiple biomarkers
3. Expected Contribution
(1) Tailored therapy guided by multiplex tissue diagnosis and in vivo imaging
- In life-threatening diseases such as cancers, multiple biomarkers are tested in one-step diagnostic procedure with lower price and high efficiency, which enables selection of optimal drug easily and enhance the prognosis of patients.
(2) Development of in vivo imaging and tissue-diagnosis kits using nanobiotechnology
- With the successful results of this research, Raman image-guided surgery, non- or minimally invasive tissue diagnosis based on endoscopic probes or needle-shaped probes are enabled.
- Biomarker-targeting SERS NP for in vivo use and multiplex tissue-diagnosis kits may make new markets in biomedical industries.
