Implantable biomaterials serve mainly as physical support devices, carriers for bioactive molecules and guidance for tissue growth. Comprehensive tissue regeneration is needed for re-functioning the original biocompatibility and bioactivity in order to overcome the limitation of current biomaterials
Implantable biomaterials serve mainly as physical support devices, carriers for bioactive molecules and guidance for tissue growth. Comprehensive tissue regeneration is needed for re-functioning the original biocompatibility and bioactivity in order to overcome the limitation of current biomaterials. Structural with functional features of extracellular matrices implement novel scaffolds design employing nano-based biocompatible organic/inorganic materials. In addition, tissue regenerative potential can be synergistically improved through tailored multi-kinetics delivery of bioactive agents including drugs, growth factors and genes. Therefore, the present study aimed to evaluate related source of technology on novel scaffold composed of 1) new functional regenerative biomimetic biomaterials, 2) regenerative functional composite hydrogel for injectable carrier, 3) organic/inorganic nanohybrid biomaterials and 4) nano/micro composite regenerative scaffold design.
With an aim to mimic the natural extracellular matrix, we fabricated the nanoand microfibrous matrix by co-electrospinning of nanofibers and microfibers. This composite matrix was expected to provide a mechanically stable and porous matrix as well as mimic the collagen fiber network similar to that of ECM. The double-layered nano-/microfibrous membranes were rolled to produce three dimensional scaffolds that exhibited the interconnected open pore structure. In vitro cell culture experiment showed that this nano-/microfibrous three dimensional (3-D) matrix provided a significantly greater microenvironment for cells to proliferate and differentiate as compared with only microfibrous matrix. ,
We employed the electrospinning technique to prepare microporous, biodegradable and biocompatible polymer scaffold which are generally composed of nanosized fibers. PLLA microfibrous scaffolds with three dimensional (3-D) structures were fabricated using an electrospinning technique followed by a mechanical expansion process. The 3-D mat was optimized by adding functional drug/gene/proteins through controlled release system. Its regenerative potential on heart, neuron, liver and cartilage was evaluated in vivo.
Gene delivery from tissue engineering scaffolds has demonstrated the ability to promote gene transfer and stimulate new tissue formation. The delivery of genes within the biodegradable scaffold can target any tissue’s regenerative process and produce prolonged expression of tissue inductive factors. Gene delivery requires that DNA can overcome a series of extracellular and intracellular barriers that can limit their efficiency.
Injectable multi-arm regenerative hydrogel with bio relative substances were developed as drug carriers for controlled release and their pharmaceutical potentials were established. For cartilage, muscle and injury therapy, multi-arm hydrogel showed great efficiency and bio-regenerative potential.
The organic/inorganic hydroxyapatite regenerative composites were fabricated as a new family of bone grafting materials because of its ductile-tough fracture behavior, apatite-forming ability in bone defect model.
In addition, tissue regenerative potential can be synergistically improved through tailored multi-kinetics delivery of bioactive agents including drugs, growth factors and genes. Drug/growth factors could be loaded in either fibers or particles embedded on the surface of scaffolding materials. Multiple release kinetics could be achieved by employing nano-particular drug releasing devices for synergistic effect. Thus, the cocktailed release of drug could stimulate attachment/differentiation/proliferation of cells in harmony with their growth pattern and in vivo events.
