연구의 목적 및 내용
세포행동 (세포증식, 생존, 형태, 이동과 분화 등) 조절에 관여하는 세포외 기질 (extracellular matrix)은 발생, 분화, 창상치유 과정에서 정상적으로 합성 또는 분해되는 리모델링 과정을 겪게 됨. 이러한 세포외 기질 리모델링의 적절한 조절에 문제가 발생할 경우, 조직재생에 장애가 발생하며, 더 나아가 기관 기능 이상(섬유증, 암, 당뇨 등)이 발생됨. 그러므로 본 연구는 세포외 기질의 역학과 리모델링에 관여하는 인자들을 분리, 동정하고 이들이 세포행동 및 조직재생에 미치는 영향과 이와 연
연구의 목적 및 내용
세포행동 (세포증식, 생존, 형태, 이동과 분화 등) 조절에 관여하는 세포외 기질 (extracellular matrix)은 발생, 분화, 창상치유 과정에서 정상적으로 합성 또는 분해되는 리모델링 과정을 겪게 됨. 이러한 세포외 기질 리모델링의 적절한 조절에 문제가 발생할 경우, 조직재생에 장애가 발생하며, 더 나아가 기관 기능 이상(섬유증, 암, 당뇨 등)이 발생됨. 그러므로 본 연구는 세포외 기질의 역학과 리모델링에 관여하는 인자들을 분리, 동정하고 이들이 세포행동 및 조직재생에 미치는 영향과 이와 연관된 세포학적/분자생물학적 작용기전을 규명하고자 함.
연구결과
1. 세포기능조절인자가 세포외 기질 합성/분해에 미치는 영향: 다양한 세포기능조절인자가 (아미노산, 지방산, 성장인자, 호르몬, 미분화 유지인자) 세포외 기질 합성/분해에 미치는 영향을 확인하였음. 세포기능조절인자는 세포막 수용체 또는 핵 내 수용체에 결합한 후 다양한 세포내 신호전달경로의 활성을 유도하며 세포골격의 재배열을 일으킴. 뒤이어 세포의 이동 및 증식이 일어나며 이때 세포외 기질의 합성과 분해가 역동적으로 일어남을 밝힘.
2. 세포외 기질성분이 세포기능에 미치는 영향: 세포외 기질성분을 (라미닌, 파이브로넥틴, 콜라젠) 세포에 처리한 후 세포기능에 미치는 영향을 확인하였음. 세포외 기질 성분은 세포-세포간 연접의 와해를 유도하므로 세포이동 및 증식을 촉진시킴을 밝힘. 또한 세포외 기질성분은 세포의 포도당 이동에 관여하는 포도당운반체 (GLUT)의 발현과 self-renewal 유지에 관여함. 또한 분화 유도 시 분화속도와 효율을 증가시킴을 밝힘.
3. 세포외 기질 역학 및 리모델링 연구: 세포외 기질에 존재하는 수용성 물질이 세포막과 세포외 기질 리모델링에 미치는 영향을 확인하였음. 세포막에 구조적으로 존재하는 세포-세포 연접, 세포-세포외 기질 연접, 수용체 그리고 운반체의 기능을 변화시키며 뒤이어 세포의 이동 및 증식을 촉진하여 세포외 기질의 역동적 변화를 가져옴을 확인함.
4. 세포질환모델을 활용한 세포외 기질 기능 연구: Matrigel을 이용한 삼차원 배양 기법을 확립한 후 세포질환모델에서 (당뇨병, 허혈) 세포와 세포외 기질과의 상관관계를 확인함. 당뇨병 상태에서 단기간의 배양 시 세포외 기질 성분의 합성과 세포 증식을 촉진시켰으나 장기간의 배양 시에는 상피세포-중간엽 전이를 (EMT) 일으키며 세포외 기질의 분해를 유도하였음. 허혈 상태에서 (저산소) 단기간 배양한 세포는 세포외 기질과 사이토카인의 합성을 촉진시켜 세포외 기질의 합성 및 분해를 조절하므로 세포의 증식 및 이동을 촉진시켰음. 반면에 장시간의 허혈 상태에 노출된 세포는 세포자멸사를 일으킴을 밝힘.
5. 전임상 질환모델 확립 및 세포외 기질의 치료효과 분석: 전임상 질환모델을 (척추손상, 관절손상) 확립한 후 합성 담체 또는 삼차원 배양한 세포를 이용하여 세포 치료 효율을 확인함. 중간엽 줄기세포를 Matrigel과 함께 삼차원 배양한 후 척추가 손상된 개의 척추에 주입한 결과 염증 감소와 신경세포 재생을 촉진시켰음. 또한 개의 손상된 연골에 Matrigel과 중간엽 줄기세포를 주입한 결과 관절염이 감소되었으며 뒷다리의 절음도 감소됨을 밝힘.
연구결과의 활용계획
세포외 기질과 세포의 상호작용, 세포외 기질 리모델링과 관련 조절 기전 및 전임상 질환모델 결과를 안정적 조직 재생을 위한 세포치료법 개발에 활용하고자 함.

The objectives of this research proposal are to isolate and analyze genes and factors that affect extracellular matrix dynamics and remodeling, elucidate the effect of these genes and factors on organ development and cell function, and determine their related cellular/molecular action mechanisms.
Result
1. Effects of cell function regulatory factors on the remodeling of extracellular matrix Effects of the cell function regulatory factors (amino acids, fatty acids, growth factors, hormone, stemness factors) on the extracellular matrix (ECM) remodeling were investigated. After the binding of regulatory factors to cell membrane receptors or nuclear receptors, intracellular signaling pathways and actin cytoskeleton rearrangement were promoted. Cell migration and proliferation were subsequently promoted with the synthesis and degradation of extracellular matrix.
2. Analysis of the ECM components on regulation of cellular function Effects of ECM components (laminin, fibronectin, and collagen) on the functional regulation of cell were investigated. ECM components promoted cell migration and proliferation by inducing the disruption of cell-cell junctions. In addition, the ECM components induced the regulation of glucose transporter (GLUT) expression, the maintenance of stem cells self-renewal, and the efficiency of stem cells differentiation.
3. Study of the ECM remodeling and dynamics The effects of ECM soluble factors on cell membrane and its related ECM remodeling were investigated. Soluble factors regulated the structure and function of cell membrane; cell-cell junction, cell-ECM junction, receptors, and transporters. These changes resulted in promoting cell migration and proliferation through remodeling and dynamics of ECM.
4. Study of ECM function in cell-based disease model After in vitro 3D culture system was established using the Matrigel, interactions of cell-ECM in cell-disease model (diabetes and ischemic cell model) were investigated. In the diabetic environment (high-glucose), ECM synthesis and cell proliferation were promoted in short-term culture, whereas ECM degradation and epithelial mesenchymal transition (EMT) were induced in long-term culture. In the ischemic environment (hypoxia), ECM synthesis or degradation were regulated by the production of cytokines, which resulted in acceleration of cell proliferation and migration in short-term hypoxia culture, while apoptosis was induced in long-term culture.
5. Establishment of the pre-clinical animal model and analysis of therapeutic effects of cell therapy based on 3D culture system After establishment of the pre-clinical animal disease model (spinal cord injury model, cartilage injury model), therapeutic efficiency of cell therapy using in vitro 3D culture system (Matrigel) was investigated. The combination of Matrigel + MSC produced beneficial effects in dogs with regard to functional recovery following spinal cord injury through enhancement of anti-inflammation, neuronal extension and neuronal regeneration effects. Also, transplantation of Matrigel + MSC to the damaged cartilage reduced arthritis and lameness of hind legs.
