Exosome형태와 디스인테그린의 특이구조를 지닌 신규인자 exosomal ADAM15를 새로이 발굴하여 인테그린과 결합함을 확인하고 확립된 분석시스템으로 세포이동을 조절함을 입증함. 그리고 면역질환 마우스 모델에서 그 면역억제의 효능을 확인하여 면역조절의 조절기전을 가지고 있음이 본 과제에서 증명되었음.

Integrin is a transmembrane receptor for extracellular matrix (ECM) proteins and its physiological function is closely associated with cell adhesion, migration, and other cellular events. Specific aim of this study is to investigate and elucidate the molecular mechanism of novel factor(s) that modulates ECM remodeling of immune cells. Final goal of this work is to establish the basic principle for developing novel immunotherapy. Establishing the functional role of integrins in ECM remodeling of immune cells will provide an opportunity for developing novel immunotherapy agents. In this study, we established the analytical system of integrin function and investigated the regulation of proteinases and integrin-mediated signaling in immune responses. Proteases including matrix metalloproteinases MMP-2, MMP-9 and serine protease uPA were induced, activated, regulated by immunological stimulator via integrin-mediated signaling pathway. Immune cell migration through the activation of integrin-mediated signaling pathways increased. The new regulatory factors which is involved in this integrin-mediated cell migration were identified; exsosomal ADAM15, BIG-H3, PAI-1. Among these molecules, exsosomal ADAM15 is released as a component of exosome in macrophage and interacts with integrin αvβ3 in a RGD disintegrin domain- dependent manner. We investigated the function and regulation system of the novel factor, exosomal ADAM 15 in ECM remodeling in vitro and in vivo. Finally, we established the functional role of the novel factor exsosomal ADAM15 in animal model system of immunological disease for therapeutics agent development.
