연구의 목적 및 내용:
본 연구는 ASMT 유전자의 클로닝이란 선행 연구기반을 통하여, 벼에서 멜라토닌 생합성 첫 효소인 AANAT 유전자를 클로닝하고, 이들 두 유전자 (AANAT & ASMT)의 조절기작 연구를 통하여, 벼에서 멜라토닌의 생합성 mechanism, 스트레스 내성 기능 규명 및 관련 유전자원을 기능성작물 육성을 위한 육종소재로의 응용 등을 연구하고자 한다. 주요 연구내용으로서는 ASMT 과다발현 형질전환벼, RNAi 억제발현 형질전환벼, ASMT homologs 유전자 과다발현 형질전환벼, 멜라토닌 분해유전자
연구의 목적 및 내용:
본 연구는 ASMT 유전자의 클로닝이란 선행 연구기반을 통하여, 벼에서 멜라토닌 생합성 첫 효소인 AANAT 유전자를 클로닝하고, 이들 두 유전자 (AANAT & ASMT)의 조절기작 연구를 통하여, 벼에서 멜라토닌의 생합성 mechanism, 스트레스 내성 기능 규명 및 관련 유전자원을 기능성작물 육성을 위한 육종소재로의 응용 등을 연구하고자 한다. 주요 연구내용으로서는 ASMT 과다발현 형질전환벼, RNAi 억제발현 형질전환벼, ASMT homologs 유전자 과다발현 형질전환벼, 멜라토닌 분해유전자 발현 형질전환벼, 벼에서 AANAT 유전자 클로닝, AANAT 과다발현 형질전환벼, AANAT RNAi 억제발현 형질전환벼 등의 다양한 멜라토닌 생합성 변이체를 육성하여 이들 변이체 벼로 통하여 멜라토닌의 식물내 기능과 조절기작을 연구한다.
연구결과:
1. Rice N-acetylserotonin methyltransferase (ASMT) 과다발현 형질전환벼 육성과 벼에서 멜라토닌 함량 변화 측정 함.
2. 벼 ASMT 억제발현 형질전환벼 육성과 벼에서 멜라토닌 생합성 변화를 조사 함.
3. 멜라토닌 생합성의 2번째 효소인 T5H 유전자가 knock-out 된 Sekiguchi mutant 벼에서 멜라토닌 생합성을 연구 함.
4. 빛에 의한 멜라토닌 생합성 연구 유도 기작과 암처리에 따른 멜라토닌 생합성 연구 함.
5. 벼에서 멜라토닌 생합성의 penultimate 유전자인 SNAT 유전자의 세계 최초 클로닝 및 효소 kinetics 분석 함.
6. 벼에서 ASMT homologs 유전자 ASMT2 및 ASMT3 유전자 기능 분석 연구 함.
7. 멜라토닌 생합성 유전자의 cellular localization 및 고온저항성 효소 특성을 규명 함.
8. In vivo SNAT 효소활성 측정 방법 및 ASMT 효소발현에 관여하는 물리적 요인 규명.
9. Sheep SNAT 과다발현 형질전환벼의 포장에서 멜라토닌 증대에 따른 형질전환벼의 생장특성 및 수량성 지표 분석.
연구결과의 활용계획:
세계최초로 식물에서 멜라토닌 생합성의 rate limiting enzyme으로 여겨지는 SNAT 유전자를 클로닝 하였으며, 식물에서 멜라토닌 생합성의 최종효소인 ASMT 유전자의 homologs 유전자 2종을 추가로 클로닝하였고, 멜라토닌 생합성 유전자의 세포내 위치를 규명하였다. 또한 고온과 dark 가 멜라토닌생합성 유전자를 유도 할 수 있음을 규명하였음. 본 연구결과는 식물에서 멜라토닌 생합성 유전자의 발현기작 및 멜라토닌의 metabolic engineering 연구에 중요한 과학적인 지식을 제공하였고, 본 연구결과로 통해 다양한 식물에서 멜라토닌의 생합성 및 유전자 발현 기작을 연구할 수 있는 정보를 제공하였음. 추후 식물에서 멜라토닌 기능 연구 및 분화 연구에 크게 활용될 것임.

Melatonin is synthesized from tryptophan by four enzymatic reactions that are similar in plants and mammals. Unfortunately, the last two genes, AANAT and ASMT, have not yet been cloned where AANAT acetylates serotonin into N-acetylserotonin and ASMT methylates N-acetylserotonin into melatonin. Recently, our group cloned the last enzyme, ASMT in rice plant. Based on the discovery of ASMT, we try to examine the functional roles of melatonin in plant by using the ASMT gene at the level of molecular and genetic analysis. Our research contents include the loss and gain of function study of ASMT, analysis of ASMT homologs, cloning of AANAT, and its overexpression. Using these genetic resources, we may draw up the real functions of melatonin in plants. Finally, these useful rice genotypes might further be utilized as genetic resources for breeding a functional rice food.
Result:
1. Generation of transgenic overexpressing rice N-acetylserotonin methyltransferase (ASMT) and quantification of melatonin.
2. Effects on melatonin and its precursors in ASMT RNAi transgenic rice plants.
3. Melatonin and serotonin contents in tryptamine 5-hydroxylase(T5H) deficient rice, a Sekiguchi mutant.
4. Study on melatonin levels by light and dark conditions.
5. Cloning of serotonin N-acetyltransferase (SNAT) from rice plant and its characterization of enzyme kinetics.
6. Cloning of ASMT homologs ASMT2 and ASMT3 from rice and their functional roles in melatonin biosynthesis in rice seedlings.
7. Cellular localization of melatonin biosynthetic genes SNAT and ASMT and their thermotolerance features.
8. Development of in vivo SNAT and ASMT enzyme activity measurement in rice seedlings under light or dark condition.
9. Phenotypic characteristics of transgenic rice plants overexpressing sheep SNAT gene in the paddy field condition.
Expected Contribution:
For the first time, we can discover the hidden functions of melatonin in plants through the molecular genetic study. Also, the various transgenic rice plants generated throughout this research can be employed as valuable genetic resources for breeding better quality of foods which contain a high level of melatonin exhibiting antioxidant activity, providing sound sleep,and extending life span
