CCR3를 발현하는 대표적 allergic cells인 Th2 cells, mast cells, eosinophils에서 zinc finger transcription factors인 GATA-1, GATA-2, GATA-3에 의해서 세포-특이적으로 조절됨을 규명함 - J Immunol 2013에 발표 4. “Olig2”라는 전사인자 유전자가 백혈구 중에서 유일하게 eosinophils에서 선택적으로 발현함을 동정하였음 - 투고 후 게재 거절되어 후속 작업 진행 중 5. 2,600여개의 methylated regions을 동정하였고, 이들 중 약 70%가 common methylated regions이었으며 두 populations의 methylation pattern이 매우 유사함. - 말초혈액 호산구에서 제한적으로 발생된 DMR regions이 naturally occurring eosinophils의 분화/기능에 중요한 역할을 할 수 있을 것이라 추정함. 논문 작성 중 연구결과의 활용계획 호산구(eosinophils)는 말초혈액내에서 lymphocytes, monocytes 다음으로 많은 백혈구임. 사람의 질병과 관련하여 호산구가 연구되는 가장 큰 이유는 호산구가 알레르기 천식의 주요 effector cells로 작용하기 때문인데, 최근에는 근육재생, 지방조직 퇴화, 포도당 항상성 유지, 간재생 등의 대사과정에 필요한 세포로 알려져서 관심의 더욱 고조되고 있다. 본 연구를 통해서 J Immunol를 포함하여 4편의 논문 (3편 본 연구 단독사사; 1편 공동 사사)의 발표하였는데 특히 이 연구를 통해서 발굴한 호산구-특이 유전자는 세계 최초의 발견이어서 본 연구자를 매우 흥분케 하였다. 그러나 아직 이 유전자가 호산구에서 수행하는 기능은 모르는 상태이다. 현재 후속 연구가 진행 중인데 호산구에서 선택적으로 과량으로 발현된다는 사실만으로 호산구 생물학의 연구에 새로운 전기를 마련한 것이며 추가연구를 통해서 그 기능을 파악하면 그 학술적 가치가 매우 높을 것으로 기대한다.

To this end, we aimed to identify a novel transcription factor or epigenetic regulation that would be specific or unique to eosinophils using eosinophils. 1. Correlation of CCR3 transcription with CpG methylation within GATA elements of CCR3 regulatory region. 2. Regulation of CCR3 transcription by different GATA factors in a cell type-specific manner 3. Analysis of genome-wide methylation pattern of eosinophils Result 1. The CpG methylation within GATA elements of CCR3 regulatory region was determined by bisulfate pyrosequencing in peripheral blood eosinophils, CB-derived eosinphils and cell lines and correlated the degree of methylation with their CCR3 mRNA levels. The CpG methylation fine-tunes CCR3 transcription. - This study has been published. (Uhm TG et al. Exp Mol Med, 2012; Uhm TG et al. Allergy Asthma Immunol Res 2012). 2. Elucidation of the molecular basis of the CCR3 expression by the different GATA factors in a cell type-specific manner: GATA-1 for eosinophils; GATA-1 and GATA-2 for mast cells; GATA-3 for Th2 cells. - This study has been published elsewhere. (Kong SK et al. J Immunol 2013) 3. CD34+cells were isolated and cultured with hematopoietic cytokines for 24 days. Eosinophils that were positive for MBP, an eosinophil marker, were than 95% at day 24. This in vitro eosinophil model was utilized throughout this study. 4. Using Oligochip microarray and the in vitro eosinophil differentiation model, Olig2, a helix-loop-helix transcription factor, was found to be expressed selectively and abundantly in eosinophils among human leukocytes. Although this study has been rejected from a high-ranked journal due to its unclear function, we continue to decipher its molecular function for future. 5. We analyzed the difference in genome-wide methylation patterns between peripheral blood eosinophils and CB-derived eosinophils, and found a few important aspects. We are currently preparing a manuscript. Expected Contribution The main reason that eosinophils have been studied is because they serve as effector cells in the pathogenesis of allergic asthma, and it is now mounting evidence that they play active roles in homeostasis such regeneration of muscle and liver, glucose metabolism, and adipocyte degeneration. In this study, we have published 4 articles, including an article in the Journal of Immunology. Importantly, we identify a novel gene (Olig2), which encodes the first eosinophil-specific transcription factor. We surely keep studying the interesting gene function, and elucidation of its function in eosinophil will provide a new avenue in the field of eosinophil biology.
