Ⅳ. 연구개발결과

2. 뿌리털 특이 cis-element(RHE)를 이용하여 애기장대 전체 유전체로부터 새로운 뿌리털 특이 발현 유전자들을 선별하고 이들이 뿌리털 발달에 미치는 영향을 분석하였다. 이전 연구를 통해 우리는 뿌리털 특이적인 cis-element인 RHE를 선별하였고, 뿌리털 특이적으로 발현하는 유전자(RHS)또한 선별하였다. RHS 유전자에는 형태형성과 관련된 단백질을 코딩하는 유전자들이 포함되어있었다. 예를 들어 세포벽 조절 단백질,인산화효소, 신호전달 관련 단백질 등이다.
3. 세포의 생장은 적당한 크기를 이루기 위해 조절되어야 한다. 우리는 선행연구를 통해 선별된 RHS 유전자 중에서 뿌리털 길이생장 또는 정단 생장을 억제하는 특징을 갖는 유전자에 초점을 맞추어 연구하였다. 그 중에서 Pro-rich receptor-like kinase,ROOT HAIR SPECIFIC 10(RHS10)은 뿌리털 길이 생장 또는 정단 생장을 억제하는 특징을 갖는 것을 확인하였다(Won et al., 2009). 또한 RHS10의 extracellular Pro-rich domain을 분석하여 단백질의 기능을 밝혔으며, RHS10 orthologu와 paralog에서 그 기능이 보존되어있음을 확인하였다. 기존에는 뿌리털 발달 유전자 중에 Receptor-like kinase의 직접저인 하위 유전자가 명확하지 않았다. 우리는 선행연구를 통하여 RHS10의 직접적인 타깃 유전자를 선별하였으며, 하위 유전자의 조절 방식도 연구하였다.RHS10의 하위 유전자로 생각되어지는 RNS2는 RNase T2 그룹2에 속해있으며, 생물학적 역할은 이미 보고되었다. RNS2는 다양한 조직에 분포하지만 RNA가 존재하는 세포질에서 활발히 그 기능을 수행할 것으로 여겨지며(Hillwig et al., 2011), 이것은 막 단백질인 RHS10의 세포질 방향에서의 타깃이 될 수 있을 것으로 여겨진다.
4. 옥신은 식물의 성장과 발달을 조절하는 핵심 호르몬이다. 세포의 분열, 확장, 분화등의 과정을 조절한다. 옥신신호 전달과정은 짧아서 세 단계로 구성되어 있지만, 그럼에도 식물의 생장 촉진과 억제에 광범위한 영향을 미치고 있다. 뿌리에 있어서 옥신에관한 연구는 많은 부분이 진척되고 있는데, 배세포 분화(Radmacher et al., 2011), 뿌리의 생장, 곁뿌리의 발달(Okushima et al., 2007), 뿌리 생장세포 조절(Dello loio et al.,2007) 등이다. 특히, 옥신 극성수송전달체의 관한 연구(Ganguly et al., 2014)가 활발하다. 하지만 아직까지 뿌리털의 발달과 옥신 신호전사자조절인자에 관한 연구는 드물게보고되는 경향이다. 옥신 신호 전사조절인자 ARF는 전자조절촉진인자 activator(aARF)와 전사조절억제인자 repressor(rARF) 두 그룹으로 나뉜다(Guilfoyle et al., 2007). 본 실험실은 선행연구를 통해 11종의 옥신 신호전사조절인자가 뿌리털의 발달과 생장에 미치는 분석하였다. 뿌리털의 길이를 촉진하는 ARF 4종과 억제하는 ARF 7종을 구별하였으며, 옥신과 뿌리털 생장의 관계를 분석하고 있다.
5. 옥신신호전달과 옥신 수송은 뿌리털의 형태생성에 영향을 준다. 옥신에 의한 뿌리털의 형태변화는 단일세포 시스템인 부리털을 지표로 사용하여 옥신 메카니즘을 연구하는데 도움이 된다(Emons and Ketelaar, 2009). 옥신 반응이 식물의 변화를 유도하기위해서는 유전자의 발현을 조절해야 한다. 옥신신호전달 과정은 두 종류의 전사조절요소 그룹이 필요하다. 옥신 반응 유전자 프로모터에 존재하는 시스요소(AuxRE)에 결합하는 옥신 전사조절요소(ARF) 그리고 다른 단백질들과 결합할 수 있으면서 동시에ARF에 결합하는 전사조절저해요소(Aux/IAA)이다(Guilfoyle and Hagen 2007). 본 실험실은 기존에 수행하던 뿌리털 연구를 바탕으로 옥신 신호전달 과정 선행연구 하였다. 전사 촉진 도메인을 갖는 aARF는 Aux/Iaa에 의해 저해 받지 않을 경우 전사촉진 기능이 증대되는 것으로 알려져 있지만, 전사저해 도메인을 포함하는 rARF의 경우에는Aux/Iaa와의 관계가 아직까지 명확하지 않다. rARF의 단백질 도메인 분석을 시도하고,뿌리털의 발달에 미치는 영향 분석하려 한다.
6. 뿌리 표피세포의 운명결정 기작을 규명하기 위하여 그동안 뿌리표피세포 운명결정기작에 관한 연구에 많이 이용되어지지 않았던 유전체적 접근 방법을 이용하고자 한다. 이러한 접근 방법은 genetic redundancy나 lethality에 의해 나타나는 정방향 유전학의 한계를 극복할 수 있게 해 줄 것이다. 가장 중요한 것은 “후보유전자군을 어떤 기준으로 확보하느냐” 이다. 현재 가장 많이 시도되고 있는 것은 여러 뿌리표피세포운명결정에 이상을 보이는 돌연변이체와 과발현 식물체 등의 뿌리에서 microarray를 통해 발현되는 유전자를 비교 분석하여 후보유전자군을 확보하는 것이다. 또 운명결정과정에 관여하는 것으로 밝혀진 인자들 중 SCM과 TTG1을 제외하고 모두 전사조절인자이므로 전사조절인자의 하위인자 후보군을 찾기 위한 ChIP-Chip이 이용되어지고 있다.
7. 본 연구팀에서는 FACS를 이용하여 뿌리 정단부의 표피세포를 아래층의 피층세포와의 상대적인 위치에 따라 분리, 즉 H 위치 표피세포와 N 위치 표피세포를 각각 분리하여 각 위치의 세포에서 RNA를 분리하였다. 위치특이적으로 발현되는 유전자는 두 위치 표피 세포의 차이를 유발하는데 관여하는 유전자일 가능성이 높으며, 따라서 일차스크린된 유전자들 중 뿌리표피세포운명결정에 관여하는 인자들을 찾는 것은 성공가능성이 매우 높을 것으로 기대하고 있다. 여기서 확보된 위치특이적발현 유전자를 과발현 형질전환체로 제작하여 뿌리표피세포 표현형을 관찰하였다. 이것을 통해 앞에서 최근 새로운 인자 확보를 어렵게 하고 있는 genetic redundancy나 lethality 등의 문제로 생기는 정방향 유전학의 한계를 극복하고자 하였다.
본 과제에서는 뿌리정단부에서 유전자를 강력히 발현시킬 수 있는 Q2610 enhancer trap line을 이용하여 activation tagging line pool을 만들어 뿌리표피세포운명에 이상이생긴 gain-of-function돌연변이체를 찾고자 하였다. 이와 같은 돌연변이 탐색으로 다른연구팀에서는 찾을 수 없는 새로운 인자들을 확보할 수 있었다.

Besides, root hair grows at root surface, it is easily observable in vivo condition. Plant scientists take root hair as a model system for researching cell growth. We have studied the mechanism of root hair growth and plant hormone by using a root hair cell model.We select Arabidopsis thaliana root hair specific expression genes by Root hair cis element(RHE) which was identified in our preceding research. We also performed a similar work with rice. Rice is a useful monocotyledonous crop. We acquire Arabidopsis root hair specific transcriptome data including 857 genes and rice transcriptome data including 197 genes as results.Cell growth should be controlled proper size. The function of ROOT HAIR SPECIFIC 10(RHS10) is discovered in preceding research. RHS10 suppresses root hair tip growth. It belongs to Pro-rich receptor-like kinase.
We found that RHS10 protein has a extracellular Pro-rich domain and is conserved at ortholog and paralog.Direct target gene of receptor-like kinase is unclear among root hair growth genes. We screened out direct target genes of RHS10 by Yeast two-hybrid method. Considered as a target of RHS10, RIBONUCLEASE2(RNS2) belongs to the RNase T2 groups and is already reported biological function. Loss of function lines and over expression transgenic lines of RNS2 show root hair phenotype closely connected root hair elongation. RHS10 transgenic lines also have change of RNA level. Therefore RNS2 is regarded as a target of RHS10. It can be target of RHS10 on the cytosol.Auxin is crucial plant hormone. It regulates cell division, expansion and differentiation. In spite of only three components(TIRs-Aux/IAAs-ARFs) of auxin signaling, auxin has far-reaching growth effects on plant growth. However Relationship of root hair development and auxin signaling components is rarely reported.
We studied effect of 11 different auxin response factors(ARFs) on the root hair growth. By root hair specific expression promoter, we finally get 4 ARFs activating root hair growth and 7 ARFs suppressing root hair growth. Activator ARF(aARF) over expression results in increasing IAA19 and EXP A7 mRNA and repressor ARF(rARF) over expression decreases the two genes mRNA level at seedling’s root. IAA19 represent early auxin response genes. Level of EXP A7 mRNA is important root hair elongation.Auxin signaling and auxin transport can affect root hair morphology. Change of root hair morphology is helpful studying auxin mechanism. aARF is transcription activator cause of activation domain. aARF function is improved without repression transcription factor,Aux/IAA. rARF has repression domain and can work as a transcription repressor. But its relationship with Aux/IAA has yet to clear. We are trying to analyze protein domain of rARF and discover effect of root hair development within auxin signaling.
Plants uptake nutrients they need from soil through their root. Root hairs account for about 70% of the root surface, which are responsible for the nutrient uptake. The Arabidopsis root epidermis is the outermost tissue in the root, and it is composed of two cell types: root-hair bearing cells (hair cells) and non-hair cells. Both cells originate from the epidermis/lateral root cap initial in the root meristem, and differentiate according to their relative position to the underlying cortical cells. The epidermal cells located on the periclinal cell wall of the underlying cortical cells and contacting a single cortical cell (the N position) differentiate into a non-hair cell, while the epidermal cells located over the anticlinal cell wall of the underlying cortical cells and contacting two cortical cells (the H position) differentiate into a hair cell. The root hair and non-hair cells in the Arabidopsis (Arabidopsis thaliana) root epidermis are specified by a suite of transcriptional regulators.
Two of these are WEREWOLF (WER) and CAPRICE (CPC), which encode MYB transcription factors that are required for promoting the non-hair cell fate and the hair cell fate, respectively. A lateral inhibition mechanism mediated by an R3 single-repeat MYB protein, CAPRICE (CPC), has been proposed to explain the specification of the two types of root epidermal cells (hair cells and non-hair cells). However, the precise function and relationship between these transcriptional regulators have not been fully defined experimentally. Furthermore, it is not clear how CPC acts preferentially in the H-position cells, rather than the N-position cells where its gene is expressed, and how many more genes need to be elucicated in this pathway. We have tried to address these issues by examining the genes expressed in the root epidermis with a position-specific manner and by examining the root phenotype of the root-specific activation tagging lines.
We have made more than 300 transgenic Arabidopsis plants overexpressing the position-specific genes and more than 4,000 root-specific activation tagging lines. We examined the molecular mechanism of the WER and the CPC in detail by misexpressing the WER gene using the GAL4-Upstream Activation Sequence (UAS) trans-activation system. We find that WER overexpression in the Arabidopsis root tip is sufficient to cause epidermal cells to adopt the non-hair cell fate through direct induction of GLABRA2 (GL2) gene expression. We also show that GLABRA3 (GL3) and ENHANCER OF GLABRA3 (EGL3), two closely related bHLH proteins, are required for the action of the overexpressed WER, and that WER interacts with these bHLHs in plant cells. Furthermore, we find that CPC suppresses the WER overexpression phenotype quantitatively. We also examined the effect of misexpressed CPC on cell fate specification and CPC localization in the root epidermis.
We show that CPC is able to move readily within the root epidermis when its expression level is high, and that CPC can induce the hair-cell fate in a cell autonomous manner. We provide evidence that CPC is capable of moving from the stele tissue in the center of the root to the outermost epidermal layer, where it can induce the hair cell fate. In addition, we show that CPC protein accumulates primarily in the nuclei of H-position cells in the early meristematic region, and this localization requires the H-cell-expressed ENHANCER OF GLABRA3 (EGL3) bHLH transcription factor. These results show that WER acts together with GL3/EGL3 to induce GL2 expression, and that WER and CPC compete with one another to define cell fates in the Arabidopsis root epidermis. In addition, these results
suggest that cell-cell movement of CPC occurs readily within the meristematic region of the root and that EGL3 preferentially traps the CPC protein in the H position cells of the epidermis.
