The materials baiting Rosellinia necatrix to diagnose white root rot can be applied by the chopped twigs such as mulberry, persimmon, and pear, but mulberry twigs showing a clear contrast between fresh tissue and the white mycelia developed on twig could be recommendable to the growers. Furthermore, observing the unique hyphal swelling of R. necatrix under the light microscope can be effective tool to diagnose white root rot in a real time. For five fungi commonly infestating the bark tissue in root such as Trichoderma sp., Botryosphaeria dothidea, Helicobasidium mompa, R. necatrix, and Phomopsis sp., the
unique PCR bands to distinguish R. necatrix from the others can be produced by the primers combinations such as R2-R8, R10-R7, and R2-R7. Commercial electric nose (e-nose MobileⓇ) and detection dogs were used to make sure whether these can detect the infection of white root rot (Rosellinia necatrix) in pear orchards. The quality of odors was not different between the infected plots and the non-infected ones but the intensity in the infected plots was significantly lower than in the control. For the plot of the infected rootlets exposed to the air in plastic basket partly open, the dogs succeeded to detect them at the rate of 88.9 percent after five day’s special training, and reached 100 percent success after 10 day’s training. But the dogs failed to detect the infected rootlets for the plot of potting with soil and infected rootlets, and for the plot of burying the infected rootlets into pear orchards 15cm in depth. Practically, there seemed to be impossible for detection dogs to be used as early detector of white root rot in pear orchards. To control white root rot in pear orchards, the control measures such as hot water irrigation(approximately 55℃), mixing soil with isoprothiolane, trunk injection with phosphorousacid and acibenzola-S-methyl, and their combinations were applied. Of treatments, hot water irrigation and it’s combinations were effective and stable to suppressing the revival of the pathogen in the infected soil in 10 months. Successive monitoring to the survival of the pathogen should be carried out to reach the complete control.
This study was performed to monitor the seasonal occurrence of Pseudococcus comstocki (Kuwana) by using pheromone traps and to test a mating disruption technique to control this pest in pear orchards. But we made the trap for P. comstocki forecasting which was formed ‘⋀’ and modified sticky plate of moths trap. We compared the attractiveness of rubber septa loaded with 1.5 and 3.0 mg of these pheromone, 2,6-dimethyl-1,5-heptadien-3-yl acetate of P. comstocki and 2-isopropyliden-5-methyl-4-hexen-1-yl butyrate of P. kraunhiae. In Y-tube test, P. comstocki moved to the pheromone bait but P. kraunhiae not attracted. A total of 1,021 and 1,431 males were caught in traps baited with 1.5 mg and 3 mg of the pheromone, respectively. The numbers of males caught were not affected by trap color, although yellow traps were more attractive than white traps. In 2012, P. comstocki males were captured between June and October, with peaks in late June, early August, and late September. In 2013, the males were trapped between June and October, with peaks in middle June, late July, and late September. In 2014, the occurrence peak chimed in with former years, but the captured males were low in late September to early October. In pheromone mating disruption tests, catches were reduced by 17.7, 65.3, and 62.9% in orchards treated with 450, 900, and 1,350 mg per 10a of the pheromone, respectively. At harvest, 3.4, 2.9, and 4.8% of fruits in orchards treated with 450, 900, and 1,350 mg per 10a were damaged by P. comstocki, while 9.5% were damaged in the control orchard. Although the control effect was shown in the treatments, the P. comstocki was captured many numbers in late September to early October. So, the 630mg of pheromone were use in early May and 270mg treated in early August in 2014. This treatment was shown 1.2% fruit damage rate and over 89% control value.
