1. 포도의 꽃 발달 단계 규정

1. 포도의 꽃 발달 단계 규정
‘Campbell Early’와 ‘Tamnara’의 두 품종의 포도에서 꽃의 구조와 발달에 따른 특징들을 조사하였다. 포도 꽃의 발달 단계 규정은 애기장대에서 소포자(microspore) 형성 시기부터 성숙한 수배우체(male gametophyte)의 출현 시기인 꽃 발달 9단계부터 13단계까지만 한정하여 규정하였다. 포도에서 4분포자(tetrad)가 나타나는 시기를 꽃 발달 9단계로, 단세포성 소포자 (unicellular microspore) 출현시기부터 중기 2세포성 화분(bicellular microspore)으로 발달하는 시기를 꽃 발달 10에서 11단계라고 규정하였다. 또한 성숙한 3세포성 화분(pollen)으로 발달하는 시기를 꽃 발달 12단계라고 하였다. 화관(floral cap)이 탈락하고 개화가 일어나는 시기는 꽃 발달 13단계로 정하였다. 꽃 발달 9단계부터 12단계까지 꽃받침 길이는 거의 변하지 않았지만 화관의 길이는 계속 증가했다. 따라서 포도 꽃봉오리 외관으로도 꽃 발달 단계를 판별할 수 있었는데, ‘꽃받침 길이에 대한 화관 길이의 비율’이 2.0, 3.0, 4.5, 그리고 6.5에 따라 각각 꽃 발달 9 단계, 10단계, 11단계, 그리고 12단계였다.
2. 포도의 꽃 발달 단계에 따른 배우체 발달 단계 규정
‘Campbell Early’와 ‘Tamnara’의 두 품종에서 ‘꽃받침 길이에 대한 화관 길이의 비율’을 이용하여 꽃 발달 9∼13단계인 꽃봉오리에서 암 수배우체 발달을 확인하였다. 애기장대의 경우 대포자 또는 암배우체 발달을 FG1부터 FG7 단계로 나눈다. 애기장대에서는 꽃 발달 11단계에서야 대포자(megaspore)의 FG1 단계를 확인할 수 있는 반면 포도에서는 꽃 발달 9단계부터 배낭 안에 한 개의 핵을 갖는 FG1 단계 대포자를 확인할 수 있었고, 수배우체는 4분포자 단계였다. 꽃 발달 10단계에 암배우체는 2개의 핵을 갖는 FG2 단계와 두 핵 사이 액포가 발달한 FG3 단계였고, 이 때 수배우체는 단세포성 소포자 단계였다. 꽃 발달 11단계에 암배우체는 4개의 핵과 2개의 액포가 존재하는 FG4 단계였고, 수배우체는 초기와 중기의 이세포성 소포자 단계였다. 꽃 발달 12단계에 암배우체는 8개의 핵 상태인 FG5 단계였고, 수배우체는 말기 이세포성 소포자 또는 삼세포성 소포자 단계였다. 개화시기인 꽃 발달 13단계의 암배우체는 배낭 안에 2개의 극핵, 1개의 알세포, 2개의 조세포를 갖는 FG6 단계였고 수배우체는 1개의 영양핵과 2개의 생식핵을 갖는 성숙한 화분 상태였다. 포도의 성숙한 암배우체에서는 애기장대에서처럼 2개의 극핵이 합쳐진 구조나 charazal end의 반족세포(antipodal cells)는 관찰할 수 없었다.
3. 유핵 및 무핵 포도의 종자발달에 대한 해부학적 비교
종자의 초기 발달을 비교하기 위해 사용한 유핵 포도는 ‘Campbell Early’, ‘Tamnara’ ‘Hokkou’, 그리고 ‘Cheongsu’의 4품종을 사용하였고 무핵 포도는 ‘Himrod’, ‘Tompson SDS’, ‘Princess SDS’의 3 품종을 사용하였다. 유 무핵 포도에서 적절한 종자 관찰 시기를 결정하기 위해 종자 경화시기와 종자 크기를 측정한 결과 30일 또는 35일 부터 유핵 포도 종자의 경화가 시작되었으며, 같은 시기에 유핵과 종자와 무핵과 종자 간에 크기가 뚜렷하게 차이 났다. 따라서 포도의 종자 발달 관찰은 수정 후 30 일이 적당하다고 판단하였다. 유핵 포도 종자는 수정 후 14일에는 구형 배(globular embryo) 상태였고 수정 후 30일에는 심장형 배단계(Heart embryo stage) 다음인 선형 배 단계(Linear embryo stage)였다. 반면 무핵과 유핵이 혼재하는 ‘Cheongsu’의 경우, 비교적 큰 종자의 배도 배 발달 이전(Pre-Embryo) 단계였다. 같은 시기에 무핵 포도인 ‘Himrod’의 배 발달 단계는 구형 배 발달 단계였다. 무핵포도의 경우 유핵 포도에 비해 비정상적으로 늦은 배 발달 단계를 보였으며 배유(endosperm)의 세포도 거의 남아있지 않았다.
4. 포도의 꽃 또는 종자 발달 단계 대표유전자 후보 선발
꽃 발달 단계의 지표 유전자를 탐색하기 위해 꽃 발달 패턴이 시기적으로 차이가 나는 ‘Tamnara’와 ‘Himrod’ 품종을 대상으로 꽃 발달 9∼13단계에서 발달단계를 구분할 수 있는 지표 유전자를 탐색하였다. 이 때 사용한 두 포도 품종의 발현유전자 특성 조사는 원예원으로 부터 제공받은 RNA 염기서열 자료를 분석하여 수행하였다. 애기장대에서 배우체 발달에 관여하는 유전자 정보를 이용하여 서열 유사도가 높은 69개의 포도 유전자를 확인하였다. 이중, 15개의 유전자는 두 포도 품종에서 특정 꽃 발달 단계를 표지할 수 있는 유전자 표지후보로 선발하였다. 또한 초기 종자발달에 대한 지표 유전자를 탐색하기 위해 애기장대에서 구형 전(pre-globular) 배, 구형 배, 심장형 배, 선형 배 단계에 특이적으로 발현되는 유전자 정보를 이용하여 포도에서 대응하는 36개 유전자를 선발하였다.
5. 유핵과 무핵 포도 구분 SNPs 마커 후보 조사
유핵 포도와 무핵 포도를 구분할 수 있는 후보 SNPs을 발굴하기 위해서 원예원에서 생산한 10개 유핵 품종과 4개 무핵 품종에 대한 유전체 재분석 서열을 분석하였다. 그 결과 유·무핵 구별이 가능한 1,229개 SNPs을 발굴하였고, 이들 중 1,046개는 포도 유전체 내 위치를 확인할 수 있었다. 19개 포도 염색체 중 7번과 12번 염색체에서 특히 많은 수의 SNPs 이 집중하여 분포하였다. 7번 염색체의 11개 SNPs 구간에 대해 14개 유핵 품종과 5개 무핵품종을 대상으로 염기서열 분석을 수행한 결과 5개의 가용한 SNPs 좌위가 검증되었고, 이들 중 1개의 SNPs을 대상으로 HRM(high resolution melting)을 수행하였다. 40개 무핵 품종과 8개 유핵 품종을 대상으로 조사한 결과, 분석 양상이 크게 3패턴으로 나뉘어서 한 개의 SNPs 만으로는 무핵과 유핵을 구분할 수 없었다. 향후 다른 SNPs 과의 조합을 이용하여 유 무핵 구분 마커로 개발할 수 있을 것으로 기대한다.

The main purpose of table grape breeding has recently been the selection of new seedless cultivars bearing desirable traits. For the effective breeding of seedless cultivars, basic histological, physiological, genetic, and molecular biological studies are required for berry development.
Here we identify the basic data for the development of gametophytes and seeds that can be provided in the breeding of seedless grapes. We report the discrimination of floral development in 9 to 13 stages and the anatomical characteristics of gametophyte development in each floral development stage of the grapes. The flower cap-to-calyx length ratio was 2.0, 3.0, 4.5, and 6.5 at floral stages 9, 10, 11, and 12, respectively. The flower cap-to-calyx length ratio was consistent throughout the stages for ‘Campbell Early’ (Vitis labruscana) and ‘Tamnara’ (V. spp.) of the two seeded grape cultivars, and for ‘Himrod’ (V. spp.) of the seedless cultivar. The floral development stage standards will be useful in determining the optimum sampling time for microspores culture needed to generate doubled haploid lines and the setting of an appropriate method for the application of gibberellins needed to induce parthenocarpic fruit development in ‘Tamnara’ grapes. We show differences in early seed development using four seeded cultivars and three seedless cultivars. The seed coats of the seeded grapes started hardening and had bigger seeds than the seedless cultivars after 30 days of fertilization (30 DAF). At that time, seeded cultivars have linear staged embryos and a large number of endosperm, while seedless cultivars have globular or zygote staged embryos and only a few remnant endosperm cells. To search marker genes for floral development stages and the early seed development stage, we analyzed the RNA sequencing data of ‘Tamnara’ and ‘Himrod’ provided by the NIHHS (National Institute of Horticultural and Herbal Science). We identified 15 gametophyte development-related gene candidates and 36 embryo development-related gene candidates. These candidates have the potential to act as diagnostic markers for a specific flower or seed development stage. We also searched the SNPs candidates to sort between seeded and seedless grapes using genome resequencing data of ten seedless cultivars and four seeded cultivars provided by the NIHHS. A total of 1,046 SNPs were detected as marker candidates for classification between seeded and seedless grapes. In the future, a set of SNP markers that consists of several SNPs selected from the candidates might be used for classification between seeded and seedless cultivars. SNP markers have the potential to improve the efficiency and precision of conventional plant breeding via marker-assisted selection (MAS).
