1. 연구과제의 개요

Ⅱ. 연구내용 및 결과
1. 연구과제의 개요
o 포도상 구균은 그람 양성의 구균으로 농가진, 피부감염증, 식중독, 폐렴, 패혈증, 뇌수막염 등의 광범위한 감염질환의 원인균으로 알려져 있다(4). 특히 황색 포도상구균 (Staphylococcus aureus)은 각종 감염증의 주된 원인균으로 대부분 90% 이상 β-lactamase를 생성하여 페니실린 등의 항생제에 내성을 가지며 최근 분리되는 황색포도상구균들은 거의 모두 메티실린에 내성을 나타내는 MRSA (methicillin resistant Staphylococcus aureus)들이다. MRSA는 1961년 처음 보고된(5) 이래 점차 높은 비율로 분리되고 있으며 국내에서도 MRSA의 분리빈도가 증가하고 있다(7, 10, 11). MRSA는 세팔로스포린을 포함한 대부분의 β-lactam계열의 항생제뿐만 아니라 다른 항생제에도 동시에 내성을 갖고 있어 그 문제는 더욱 심각하다(1, 8).
o MRSA는 왜 중요한가?
MRSA는 병원성일 뿐만 아니라 병원 획득 감염의 주원인이 된다. 현재 MRSA의 유일한 치료제로 glycopeptide 계열의 반코마이신이 사용되고 있으나 그 사용이 아주 제한적이며 반코마이신 등에 대한 내성 균주의 출현으로 새로운 항생제 개발이 요구되고 있다(2).
o 항생제 내성 문제 해결을 위한 해양미생물 자원
특정 항생제에 내성을 가진 병원균을 제어하기 위해서는 새로운 구조나 작용 메카니즘을 가진 새로운 항생제의 개발이 필요하다. 지금까지의 항생물질의 검색을 위해서 이루어진 연구는 방선균과 같은 토양미생물을 대상으로 주로 이루어져 왔고 해양미생물을 대상으로 한 연구는 거의 전무한 실정이다(3, 6). 해양미생물은 토양미생물과 달리 그 서식처가 해양인 관계로 저산소, 저온도, 고염분, 고압력 등의 특수 서식환경에서 생존하므로 지금까지 알려지지 않았던 생리활성 물질들을 많이 생산하고 있다. 따라서, MRSA에 대하여 뛰어난 항균력을 가진 해양미생물을 분리하게 된다면 그 분리균주는 기존의 토양미생물에서 분리된 항생물질들과는 전혀 다른 새로운 구조나 작용 메카니즘을 가진 새로운 항생물질을 생산하는 균주일 가능성이 높다. 그리고, 이러한 분리균주가 생산하는 항생 물질의 구조와 항균작용을 밝힐 수 있게 된다면 MRSA균에 의한 항생제 내성 문제를 해결할수 있는 신물질의 검색 및 신물질 합성에도 연결 될 수 있을 것이다. 따라서 다양한 해양미생물 자원으로부터 새로운 항생물질 탐색을 위한 연구의 필요성이 절실하다.

Isolation and structural identification of anti-MRSA compound In the process of solvent extraction, the anti-MRSA activity was detected in the ethyl acetate extract (48 g).The ethyl acetate extract was concentrated and the residual aqueous suspension was subjected to ODS vacuum flash chromatography with aqueous MeOH followed by methylene chloride. The active fraction against MRSA is observed in 80% MeOH fractions. Then, the active fraction was subjected to silica gel chromatography with a stepwise gradient mixture of hexane /EtOAc as eluant followed by MeOH. After then, active fractions are observed in hexane/EtOAc (8:2) fraction (70 ㎎), EtOAc 100% fraction (540 ㎎) and 100% MeOH fraction(360 ㎎). The final purification was achieved by reversed-phase HPLC as described in Materials and Methods. It was obtained 2 pure compounds, compound 2 (1.7 ㎎) and compound 9 (1.3 ㎎), exhibiting anti-MRSA activity from hexane/EtOAc (8:2) fraction. The compound 2 was then subjected to structural analysis. LC–MS spectrum of anti-MRSA compound indicated that molecular weight of compound 2 is 281.48. The molecular formula of the compound 2 was established as C13H35NO by LC–MS spectrometry and 13C NMR spectroscopic analyses. The 1H NMR data, 13C NMR spectrum, COSY, and HMBC analyses of the compound led to reveal the structure of fatty acid moisty. The identity was confirmed by the comparison with literature data of the chemical shifts. The compound was first isolated from the culture filtrate of Streptomyces kasugaensis and marine actinomycete. However, our study is the first report on originated from a marine eubacteria. In addition, we found that the previous study reported slightly different results on antibacterial activities against MRSA.Therefore, we here corrected the anti-MRSA activity of the compound and also reported the antibacterial activity against major pathogenic bacteria. In order to quantitatively evaluate the antibacterial activity of compound 2 against MRSA and other pathogenic bacteria, MIC values of the compound 2 were determined by the two fold dilution method as described in Materials and Methods. Among the 19 strains tested in this study, all MRSA strains including two standard and 10 clinical isolates only contains mecA gene encoding a penicillin-binding protein 2a (PBP2a), which plays a key role in acquiring a drug-resistance against β-lactam antibiotics (Foster,2004). As shown in Table 2, this compound showed an antibacterial activity against MRSA strains with MICs ranging from 4 to 256 μg/ml. The MICs of this compound against MRSA strains were less than those of β-lactams (ampicillin, penicillin and oxacillin), which were in a range of 64–512 μg/ml. Also, it has been previously reported that the MICs of (−)-epigallocatechin, (−)-EGCg, (+)-gallocatechin and (−)-gallocatechin from green tea (Camellia sinensis) against MRSA was 64 μg/ml (Stapleton et al., 2004). Thus, the anti-MRSA activity of 1-acetyl-beta-carboline isolated from Pseudomonas sp. YJ-1 was superior or equal to those of catechins originated from green tea.
