사람의 유전형, 식이 및 환경에 의해 다양성에 차이를 보이는 장내미생물총은 식품, 의약품 등의 대사반응에 차이를 나타내므로, 약물대사기반연구사업단은 한국인의 장내미생물총을 분석하고, 그 대사활성을 분석하고, 식품, 의약품 등의 대사체를 분석하고, 그 화합물들의 독성을 평가하고, 무균동물을 이용하여 이의 대사반응에 장내세균총의 역할을 분석하여, in vitro에서 독성평가등에 활용할 수 있는 fecalase 또는 이를 활용한 intestinal microbial enzyme mix를 제조하며, 이를 결과들을 데이터베이스화하는 사업

● 장내미생물 배양기술 확립 및 대사체 분석법 개발: arbutin, puerarin, baicalin 등을 수행함
● 장내미생물 대사체의 면역독성 평가: 마우스 비장세포 배양에서 B- 및 T-cell mitogenicity 평가
● 장내 미생물을 이용한 세포독성 및 세포사멸 평가기술 개발: 장내세균효소복합체(fecalase)와 장내미생물 대사효소계(intestinal microbial enzyme mix)를 이용한 세포독성, ROS, Bcl-2 family, caspase-3, apoptosis 등의 독성 평가기술을 개발
● 장내 미생물을 이용한 유전독성 평가기술 개발: p53 유전자 발현 관련 독성평가기술, 8-oxoguanine glycosylase 발현을 이용한 유전독성 평가기법, Comet 시험 그리고 in vitro 소핵 생성시험 기술 개발

● 항생제 처치동물에서의 식품 또는 의약품의 동태변화에 대한 연구: 무분별하게 사용되고 있는 항생물질에 의하여 장내미생물총의 변화가 생길 때 나타날 수 있는 화학물질의 동태지표의 변화에 대한 연구를 1, 2차년도에 걸쳐 수행
● Hesperitin을 rat liver microsome을 사용하여 배양 후 생성된 대사체는 각각 demethylation 과 hydroxylation 대사체로 추정되는 대사체 확인하였음: 2-methoxyphenol group이 demethylation된 대사체로 확인되었음
● 항생체 처리 모델에서의 총 hesperetin 의 체내동태파라미터는 대조군의 AUC가 항생제 처리군보다 약 3배가량 높았음: Cmax는 정상군에서는 578 ng/mL, 항생제처리군에서는 198 ng/mL로 정상군에서 더 높게 나타났음
● Percision-cut small intestine을 이용한 독성평가 방법의 확립하기 위하여 rat 소장을 250 μm의 slice로 만들어 William E 배지에서 배양하였으며, 배양시간에 따른 Na+/K+ ATPase, NADPH oxidase, alkakine phosphatase의 활성을 비교하여 2 hr, 6 hr에서 geniposide와 genipin의 영향을 비교하였음
● 항생제 투여에 의한 germ free rat model의 대사변화를 위한 대사체 연구을 통해 Global metabolic profiling으로 내인성 대사체의 변화 확인
● 항생제 처리 후 rat 뇨 시료에서 cholesterol과 25-OH-cholesterol의 농도가 감소하였고, 담즙산 젓 deoxycholic acid의 농도도 감소함. 반면 담즙산 대사체인 5β-cholestane 3α,7α,12α-triol와 간 독성을 유발한다고 알려진 chenodeoxycholic acid의 농도 증가가 확인됨. 이와 관련하여 cholesterol 7α-hydroxylase (cholesterol->7α-hydroxycholesterol)의 활성은 증가하였고, 7α-dehydroxylase (cholic acid->deoxycholic acid)와 7α-dehydrogenase (chenodeoxycholic acid->lithocholic acid)의 활성은 감소하였음을 확인함
● Esculetin과 esculin의 혈장 젓 농도를 LC/MS/MS 방법으로 분석하고 분석법을 validation 하여 분석법을 확립하였음: Esculetin(40 mg/kg)의 경구 투여 후, 대조군과 비교시 항생제 처리모델에서 esculetin의 AUC에는 유의성 있는 차이는 없었음

● 장내 미생물의 특이 대사시스템이 약물의 독성 유발 기전 및 약동력학 특징에 미치는 영향을 확인하기 위하여, bacitracin, neomycin, streptomycin 등의 항생제를 투여하여 장내미생물을 제거한 pseudo germ free rat (antibiotics treated group, 항생제 처리군)을 완성하고, 장내 미생물에 의한 약물의 독성기전 및 약동력학 특징 규명을 위한 in vivo system으로 사용하고자 하였으며, 아세트아미노펜을 정상 쥐와 pseudo germ free 쥐 (antibiotics treated group, 항생제 처리군)에 경구 투여하고 모약물 및 대사체 약동력학적 변화를 관찰하여 장내 미생물이 아세트아미노펜의 대사 및 약동력학적 특징에 미치는 영향, 특히 독성 유발 대사체 변화에 미치는 영향을 확인하였음
● Rat 혈장 젓 Ginsenoside Re 및 그 대사체들을 분석하기 위하여 triple-quadrupole LC/MS/MS를 이용한 분석법을 개발하였음
● 항생제 처리군과 대조군에서 baicalin 체내동태 비교연구: 항생제 처리군과 대조군에 각각 baicalin을 50 mg/kg 용량으로 경구투여하고 혈장 내의 baicalin의 농도를 측정하였음: Baicalin을 경구투여하였을 때 AUC는 대조군이 항생제 처리군보다 1.4배 높았고 Tmax도 차이가 보였음
● 사람의 분변으로부터 170 여개 균주를 분리, 동정하고, 대사효소 활성 높은 균주 16 개을 선별하고, 한국인 fecalase를 대신할 수 있는 intestinal microbail enzyme로 4 mix와 7 mix를 제안함: 제조된 microbial enzyme mix는 fecalase와 유사한 대사활성을 보였음

● 사업단 홈페이지제작: 장내미생물과 관련된 정보를 모아서 활용도를 증가시킬 목적으로 약물대사사업단 홈페이지를 작성함. 홈페이지는 사업단 개요, 소개, 연구내용, 자료실, 정보교류 및 이용안내와 회원가입 등의 내용을 포함하고 있으며 DB 홈페이지는 연구사업단, 사업안내 및 소개, 통합 정보 검색, 용어 사전, 논문, e-book 자료실, 열린광장 등의 내용으로 구성하여 사업단 로고와 캐릭더 뿐만 아니라, 트위터에 연결되어 상호 의견교환도 가능토록 함

● 현재까지 5종의 신규 장내 미생물을 분리 및 동정, 계통분류학회지(IJSEM)를 통해 보고 및 등록하였으며, 3종 이상의 신규 미생물로 추정되는 장내 미생물의 특성을 규명
● 제안한 메타지놈의 순수-균질화 기법을 연구 기간 내 확립하였으며, 이를 토대로 메타지놈 DNA 칩을 개발하였음. 51개의 한국인 장내 메타지놈과 reference로서 2개의 토양 환경 메타지놈 그리고 E.coli의 genome을 이용하여 100장의 메타지놈 DNA칩을 개발하였음
● DNA 칩의 신호 증폭 기술을 개발하기 위해 68개의 미생물의 16S rRNA gene fragment를 이용한 DNA칩을 개발하였으며, 장내미생물 주요 그룹 (Bacteroides-Prevotella, Roseburia, Faecalibacterium 등)을 탐지할 수 있는 10개 이상의 프로브를 개발하였음. 또한, biotin-TSA기법으로 기존의 cy5 probe보다 10배 이상의 신호 증폭, linker probe와의 결합으로 30배 이상의 신호 증폭 기술을 개발하였음.
● 한국인 분변시료로부터 virus-like particle만을 분리 및 농축과정을 확립하였고, 농도 단위 계수법, 형태학적 관찰법 역시 확립하였음. Viral genomic DNA를 추출하고, 증폭하여 다량의 메타지놈 DNA를 확보하였으며, 대용량 sequencing 기술에 적용하여, 장내 박테리오파아지의 genomic DNA가 담고 있는 유전적 특성을 분석하였음. 바이러스의 분류학적 정의를 바탕으로 박테리오파지인 dsDNA Podo-, Sipho-, Myophages와 ssDNA microphages가 장내 바이러스의 우점군임.
● 대용량 염기서열 분석법을 통해 359명의 한국인 장내미생물 전체 균종의 조성을 파악함. 성별, 연령별, 식이, 비만(건강), 지역별에 따른 장내미생물의 특성을 정의하였으며, 그 변화를 조사하여 한국인의 장내미생물과 건강과의 관계를 파악함. 이는, 한국인 장내 미생물 표준화 및 독성대사 기반 연구의 기초자료로 활용될 것으로 기대함

● To investigate the role of metabolism by intestinal microflora in the biological action of the ingredients of foods, drugs, herbs, etc, a variety of in vitro and in vivo testing methods have been developed. In addition, the studies on intestinal microbiome and its metabolic activity for Korean were standardized.
● Methods to culture intestinal bacteria and to analyze the metabolites produced by bacteria were developed: Arbutin, puerarin, and baicalin.
● Immunotoxicity testings following metabolism by intestinal bacteria were developed using Band T-cell mitogenicity in spleen cells isolated from normal mice

● Establishment of an infra for maintenance and supply of germ-free animals: Experimental conditions for maintaining germ-free animals and testing microbial contamination were set. For stable supply of animals in toxicity testing, breedings were expanded to produce 300 heads in the facility.
● Production of gnotobiote: Whole human intestinal bacteria were fed to produce human intestinal microflora-containing gnotobiote. The animals were used to test the possible role of intestinal bacteria in drug- or toxicant-induced biological activity in vivo.
● Pharmacokinetic studies with several drugs or toxicants in germ-free animals and gnotobiotes: Acetaminophen and ginsenoside Re were already tested and acetaminophen at toxic dose, geniposide and baicalin were tested soon for comparing their pharmacokinetics in germ-free and gnotobiotic animals. Normal SPF animals were used as a control group.
● Pharmacokinetics studies with baicalin in antibiotic-treated animals: Antibiotic would not only reduce the absolute number of intestinal bacteria but also change the intestinal microflora, which eventually reduce metabolism of certain chemcials including baicalin. To study the role of metabolism by intestinal microflora in baicalin pharmacokinetics, antibiotic-treated models were tested in rats and mice.
● The present study was quite successful, particularly in the standardization of human fecal preparation for Korean and in disseminating the research outcomes to public media, although there were a limitation of budget and research period given. Further funding for this project will help establish the concrete infra structure for studying microbial metabolism and drug action.
● Pseudo-germ free rat model was induced after 3 consecutive oral administration of mixed solution of cefradoxil, erythromycin and oxytetracycline. Germ-free mouse model was also used. The rat model was orally received hesperidin, ginsenoside Re, baicalin, esculin, geniposide, or acetaminophen (ASAP). Ginsenoside Re or ASAP also was given to the germ-free mouse for the pharmacokinetics. The analytical methods of the drug concentrations in plasma and urine samples were developed by LC/MS/MS. The pharmacokinetic parameters, AUC and Cmax, were decreased in the antibiotics-treated groups compared to the control.
● The concentrations of various endogenous metabolites in bile and urine were investigated by application of global metabolic profiling technique. As the results, 5β-dihydrocortisol and 3-methylhistidine were significantly reduced in the antibiotics-treated rats and cholesterol and chenodeoxycholic acid were also reduced with the related 2 enzymes. This data suggests that effects of microbial metabolism on drugs may cause liver toxicity due to reported liver toxicity of chenodeoxycholic acid.
● ASAP treatment to antibiotics-treated rats or germ-free mouse was affected to the concentration of parent and its metabolites. AUCs in parent and its 6 metabolites in the germ-free mouse and parent and ASAP-glutathione metabolite in antibiotic-treated rats were significantly increased. This data should be considered carefully because ASAP is extensively used as analgesics.
● Small intestine precision-cut slice in rats in vitro was established and used for the testing the effects of geniposide concentrations and incubation time on NADPH oxidase and LDH activities
● About 500 microbial strains were separated from feces of healthy adults, and 21 kinds of enzyme activities were measured. From these strains, 100 strains were identified. 7 strains including BL-3 etc. among 100 strains were chosen and again 4 strains (BL-3, Gam-6, JY-6, A-44) with high enzyme activity by measurement of β-D-glucosamide was finally selected for preparation of coarse enzyme mixture. Microbial enzyme mixture was made from the combined mixture of the enzyme. The condition for the best stability was confirmed in freeze-drying. Using the enzyme mixture Ames test with metabolic activation system was tested in the presence of baicalin.
● For the purpose of extensive sharing of the information of this study, web homepage and microbial DB were constructed. This homepage is composed of the overview and introduction of this projects, study contents, data, information exchange, use guideline, and member affiliation. DB contains the project groups, introduction of the projects, integrated informational retrieval and search, dictionary for terminology, references, e-book, open discussion for opinion etc. In addition, information exchange was also available via Twetter.
● We had capability of culture-dependent method to cultivate unknown gut microbes by developing anaerobic incubation system, and this method will contribute to the understanding physiology of gut microbes and their ecological role in human intestine. Besides, higher than 50 species of gut microbes were isolated from the faeces obtain from Korean, and 5 strains of them were validated as novel species via the International Journal of Systematic and Evolutionary Microbiology and 3 more were on process for validation. These works will be used as biological materials to investigate gut microbial ecology.
● We developed the proposed community analysis technique, Genome Probing Microarray. Total 100 microarray chip were developed using fifty-one metagenomes from Korean subject faeces, soil environmental metagenomes and E. coli genomes. This technique will be used to analyze gut microbial community. Also, over 10 linker probes for main gut microbial groups, including Bacteroides-Prevotella, Roseburia and Faecalibacterium, were designed and developed, which will be applied to detect the variation of dominant group in intestinal microbiota by pathogenesis. To strengthen the signals of microarray, biotin-TSA technique was employed, and 30 times improved signal was established more than that of cy5 probe signalm which will be introduced to determine the pattern of gut microbial community dynamics.
● Viral-particles were purified and enriched from faeces, and enumeration and morphology of viruses were established. Extracted viral genomic DNAs were amplified using phi29 polymerase, and conducted to high-throughput 454 pyrosequencing. Most of the sequences (approximately 90%) were unidentified, phages of viral families Podo-, Siphi-, Myo- and Microviridae were predominating in identified viral sequences. However, the presence of much higher unknown sequences may reflect that investigation of gut viral assemblages is required for genetic novelty and their ecological role as microbial regulators should be determined.
● Using the faecal samples of 359 Korean subjects, Korean gut microbiota was extensively determined. By age, sex, diet, obesity and residence, relative abundance of gut microbial groups were analyzed. Deep metagenomic sequence analysis will be defined to figure out the characteristics of Korean gut microbiota compared with those of other countries.
