그린독성평가기술개발을 통해 독성 예측을 위한 기존 독성시험법의 시험 기간 단축, 비용 절감 및 시험에 필요한 동물 수를 대폭 감축함으로써 경제적으로 비용을 최소화하며 안정된 친환경/친인간 녹색사회의 건설을 촉진하고 의약품의 안전성 및 유효성 평가 분야의 차세대 첨단기술의 개발을 위해 수행되었음.

<1중단위 과제>
표적장기 독성평가를 위한 in vitro 분자스크리닝 시험법 개발 및 검증연구에서는 신장독성을 유발시키는 ER stress, 미토콘드리아 손상 및 erythrophagosis 등의 신장독성 유발 기전에 기초한 화학물질의 in vitro 스크리닝 battery system을 구축하여 검증하였음.
◦ Tier 1 스크리닝 방법에서는 다량의 샘플을 처리할 수 있고 스크리닝 속도가 빠르며 독성경로의 상위신호에서 예측할 수 있는 시험법을 구축하였으며, Tier 2 검색 시험법에서는 독성경로의 하위신호에서의 예측 평가, Tier 3에서는 독성경로 활성화에 따른 effector system을 확인하였음.
◦ 화학물질의 ER stress 유발은 Tier 1에서는 ER stress의 3가지 sensor의 활성화를 측정하기 위한 incell western assay를 실시하였고, Tier 2에서는 3가지 경로 활성화에 따른 유전자 발현 변화를 관찰하기 위하여 RT-PCR을 실시하였음.
◦ Tier 3에서는 ER stress 경로 활성화에 따른 apoptosis와 autophagy를 측정하였음. 이를 검증하기 위하여 ethylmercury를 이용하여 ER stress를 in vitro에서 관찰하였을 때 3가지 ER stress sensor가 활성화되었으며, Xbp1 splicing 및 관련 유전자 발현이 증가되었음. 또한 autophagy가 apoptosis 보다 강하게 나타났으며, ER stress 저해제인 4-phenylbutyric acid를 처리했을 때 autophagy가 저해되었음. Mouse에서 ethylmercury 투여에 의해 신장독성이 유발됨을 확인하였으며 이때 ER stress 유발 및 autophagy를 신장조직에서 확인하였음.
◦ ER stress 유발 독성물질을 평가하기 위한 in vitro battery system을 in vivo에서 검증하였음. 그러나 본 사업단에서 구축한 battery system을 이용하여 21종의 신장독성물질에 대해 검색하였을 때 그 정확도가 약 50%이었음. Mitochondria 손상에 따른 신장독성물질을 검색하기 위하여 Tier 1 시험법에서 MTT assay를 통한 세포독성, ATP level 및 ROS를 측정하였으며, Tier 2에서는 mitochondrial membrane potential을 관찰하였고, Tier 3에서는 mtDNA의 손상을 관찰하였음. 구축한 battery system을 이용하여 20종의 화학물질을 검색 시험하였을 때 정확도가 약 60%정도로 나타났음.
◦ Chemical-induced Anemia 분자스크리닝 검색 시험법에서는 신장독성물질 처리에 의해 적혈구 세포막에서 phosphatidyl serin(PS)이 유도되어 신장에서 erythrophagocytosis를 유도하고 이에 의하여 신장에 철이 축적되어 reactive oxygen species(ROS) 생성에 의해 oxidative stress를 유발하며, 이것이 kidney damage를 준다는 가설을 입증하였음.
◦ In vitro에서 직접적인 적혈구에 영향을 미치는 신장독성물질의 검색으로는 적혈구의 용혈, 적혈구막의 PS exposure 및 microvesicle 생성과 적혈구 형태 변화로 스크리닝하였음. Quinone류나 여러 중금속들 중 납, 수은 등이 영향을 미쳤으며, lipid 염증매개체나 염증관련 protein marker들 중에서 phosphatic acid와 lysophosphatic acid, 및 HMGB1이 적혈구에 변형을 유도하였음. 이들 중 최근 신장독성물지로 주목받고 있으며 직접적인 적혈구에서 PS potency를 증가시키는 Pb을 이용하여 in vivo 실험을 실시하였을 때 신장에 erythrophagocytosis, iron 축적 및 신상손상을 확인하여 가설인 독성물질이 RBC PS 노출을 유발하여 erythrophagocytosis를 통해 신장독성이 유발됨을 in vitro 및 in vivo 시험에서 최초로 확인하였음.
◦ 본 과제에서는 또한 가상세포를 이용한 신장 독성예측 시스템을 구축하였음. 여기에서는 다양한 독성물질 투여에 따른 세포 내 생리학적 현상을 모사하기 위하여 세포 신호전달/미토콘드리아/ER/세포막전달현상 등을 포함하는 통합적인 가상세포 모델을 구축하였으며, 이를 실험결과와 비교하였다. 이를 활용하여 여러 가지 다양한 신장 독성물질들의 투여 시 생리학적인 변화 예측 및 세포독성을 추정할 수 있는 소프트웨어를 개발하였음.
◦ 컴퓨터를 이용한 신장독성 예측 평가 모델로 QSAR 모델을 개발하였으며, 이를 위하여 신장독성 화합물의 실험 데이터를 수집 및 분석하였음. 본 결과를 활용하여 화합물 구조 기반의 단백질 활성화, 저해 정도를 예측할 수 있는 QSAR 모델을 개발하였다.
<2중단위 과제>
본 과제에서는 다야한 종류의 화학물질에 의한 신장독성 평가에 기존의 전통적인 독성 평가 기술에 따른 고비용ㆍ고에너지를 대체할 수 있는 최첨단 신장 독성 평가기술을 개발하는 데 목적이 있음. 또한 표적장기독성 예측 및 평가지표 개발을 위한 유전자발현, 단백 발현 산물 및 대사체 발현 산물을 연구하여 신약개발을 지원하고, 화학물질의 신속한 독성평가체계를 구축하는 것을 목적으로 함.
◦ 독성물질에 의하여 변화하는 신호를 유전자 발현 분석을 통해 분석하여 이를 통해 세포 내 신호전달지표 분자를 도출하고 네트워크 모델을 완성하였음. 이들 도출된 결과를 활용하여 신장독성을 예측할 수 있는 전사체학적 신규 지표인자를 발굴하였음.
◦ 표적장기의 세포모델에서 cisplatin cfl에 따른 microarray 자료 분석 (NCBI GEO)을 통하여 유전체 정보를 수집, 재분석하고 이를 사멸 생화학 신호 와 네트워킹.
◦ 규명된 전사인자 및 독성 유전체 기능을 세포 신호전달회로와 연계하여 상위세포신호망과 평가지표 도출 하였음.
◦ 동물모델 및 유전체 분석법을 활용하여 독성 네트워크 모델을 완성하고, 이들 자료를 GEO에 등재.
◦ 신장독성에서 변하는 miRNA 발현을 microarray 분석.
◦ 지표유전체의 혈중/뇨중 검출을 확인하여, 표적장기 독성을 예측 및 평가할 수 있는 시스템을 확립.
◦ 단백체를 이용한 신장독성 예측 평가 기술에서는 기 발표된 신장독성 in vivo 바이오마커들을 인간 신장세포주 human kidney proximal tubular epithelial cells (HK-2)를 이용하여 in vitro 바이오마커로서 가능성 있는 단백질들로서 KIM-1, calbindin, TIMP-1을 도출하였음.
◦ 또한 신규 신장독성 바이오마커를 찾기 위하여 프로테옴 분석을 수행하였음. Cisplatin, CdCl2, amphotericin B, cyclosporin A 및 HgCl2 등의 신장 독성 유발 약물들을 HK-2와 NRK-52E 세포에 처리하여 신장 독성을 유발시킨 후 변화된 세포 내 또는 배양액 내 프로테옴의 변화를 2-DE/MALDI-TOF-MS, SILAC/label-free quantification, SILAC/LC-MS 등의 단백체 분석 기술로 관찰하였음. 그 결과 in vitro 신장 독성 평가 지표 물질로 활용할 수 있는 단백체 산물 (annexin A5, PKM2, EF-1γ, SLC25A6, VDAC2, PRKCSH, Hsp70)을 도출하였음.
◦ 표적장기독성 예측 대사산물연구 (메타볼로믹스)에서는 이미 발표된 연구논문 및 관련 웹사이트 database를 통해 신장독성 유발물질에 따른 대사산물 정보를 수집, 종합 분석하여 대사체 산물에 대한 DB를 종합 분석 하였음.
◦ 사람유래 정상 신장 세포주주인 HK-2 세포에서 cisplatin 및 cyclosporin A를 처리한 후 신장세포의 사멸에 미치는 영향에 대한 기전연구를 수행하여 대사체 산물과 비교 검토하였음.
◦ In vivo 동물실험에서 cisplatin, cyclosporin A 및 D-galactosamine을 투여한 후 시간-의존적으로 변화하는 대사체 산물을 평가하였으며, 신규 신장독성 지표인자와 sensitivity를 상호 비교하였음.
◦ 신장독성 환자의 시료에서 신규 biomarker에 대한 검증 연구를 수행하였음.

◦ 첨단독성평가 기술 개발을 위하여 생체지표 및 독성물질 간의 생물학 네트워크를 생성, 효율적 생체 지표를 발굴할 수 있는 메타분석 기반 통합된 시스템 개발하였음.
◦ 효율적인 생체지표를 발굴을 위하여 우선 기존에 알려진 독성관련 생체지표에 대한 수집, 분석 및 해석과 메타분석을 통해 좀 더 신뢰성 있는 생체지표를 발굴. 또한 표적장기 관련 독성물질과 다양한 생화학적 오믹스 데이터와의 융합 및 분석에 대한 기반 연구 및 생물학 데이터의 마이닝 기법을 통해 얻어진 융합 데이터를 바탕으로 독성기전 관련 네트워크 모델의 기본 틀을 완성하고, 이를 통해 도출된 독성 생체지표물질을 평가할 수 있는 독성평가 시스템 기반 프레임워크를 개발 하였음.

The project of green toxicology & technology seeks to development of new applicable screening methods based on in vitro computational toxicity evaluation system. These include the simplicity, sensitivity of the biological responses, and the broad applicability of in vitro assay to various compounds.

These include the simplicity, sensitivity of the biological responses, and the broad applicability of in vitro assay to various compounds. Based on our experimental results, we therefore propose to development new in vitro screening methods and identify novel biomarkers for detection of target organ toxicity. This procedure will reduce the need of costly, demanding and potentially irrelevant animal studies.

◦ Development and validation studies of in vitro molecular-screening system for Target organ toxicity test", in vitro molecular screening test method systems based on the toxic mechanism such as ER stress, mitochondrial damage and erythrophagosis for the chemical induced renal toxicity were established and validated.
◦ Tier 1 Screening methods which can handle a large amount of samples and predict the toxic pathway on an upper layer signaling, were established.
◦ Tier 2 and Tier 3 Screening methods were checked the lower layer signaling of toxic pathway and the effector system according to the activation of toxic pathway, respectively.
◦ ER stress-induced chemicals were screened using the In-cell western assay to observe the activation the activation of the three ER stress sensor in the Tier 1. In Tier 2 assay, the changes of gene expression were measured by RT-PCR to identify the activation of the three ER stress pathways.
◦ Tier 3 assay were determined apoptosis and autophagy due to the activation of ER stress pathway. In order to verify this screening system, ethylmercury induced ER stress was tested. Etylmercury was activated three ER stress sensor in In-cell western assay, spliced Xbp1 mRNA and increased the ER stress related gene expression.
◦ It was induced auophage stronger than apoptosis and 4-phenylbutyric acid, an ER stress inhibitor, was inhibited ethylmercury induced autophagy.
◦ Ethylmercury was induced kidney toxicity in mouse and ER stress and autophagy were identified in the mouse kidney tissues.
◦ From these experiments, In vitro battery system for screening ER stress induced chemicals was vilified in vivo mouse model. However, when we screened 21 renal toxicants using establishing in vitro ER stress screening battery system, the accuracy was less than 50%.
◦ For screening Mitochondria damage due to kidney toxic substances, Cytotoxicity by MTT assay, cellular levels of ATP and ROS were measured in Tier 1 and the mitochondrial membrane potential and mtDNA damage was observed in Tier 2 and Tier 3, respectively.
◦ When tested 20 renal toxic chemicals using in vitro mitochondria dysfunction screening battery system, the accuracy was about 60% or so.
◦ Chemical-induced Anemia molecular screening system confirmed the hypothesis that renal toxicants allowed exposing the phosphatidyl serin (PS) on erythrocytes membrane; it induced erythrophagocytosis in the kidney, followed the accumulation of the iron, induced the oxidative stress by generating reactive oxygen species and resulted in the damage of kidney.
◦ In in vitro systems, kidney toxic substances affecting directly the red blood cells were screened by hemolysis of red blood cells, PS exposure on the membranes of RBC, microvesicle generation of erythrocyte and morphological changes of RBC.
◦ Quinones, heavy metals such as lead and mercury, lipid inflammatory mediators (phosphatic acid and lysophosphatic acid) and HMGB1, inflammation-related protein marker, were exhibited the erythrocyte deformation.
◦ In vivo animal experiments were carried out by selecting a lead which has been focused on the renal toxicity and has high potency the direct expose of PS on RBS membranes. Lead was induced the kidney damage and concomitantly identified the erythrophagocytosis and iron accumulation in kidney tissues.
◦ These results confirmed first time the hypothesis that renal toxicants allowed exposing the phosphatidyl serin (PS) on erythrocytes membrane induced renal toxicity through erythrophagocytosis, in vitro and in vivo experiments.

◦ In this sub-project, an integrative system of virtual cell was developed. To simulate cellular physiological phenomena, we implemented a systematic model of virtual cell that includes cellular signaling pathways, Mitochondria, ER, and membrane physiology and compared the computed results with experimental ones.
◦ Moreover, a computer software to evaluate the probability of renal cellular apoptosis by toxins was also developed.
◦ In addition, we gathered experimental data of renal toxin complexes to evaluate renal damage by toxins and developed a QSAR model to predict protein activation or inhibition level based on the complex structures.

◦ To development of novel biomarkers for acute kidney injury (AKI) is important to replace renal biopsies. Various serum and urinary biomarkers have been identified for detection of AKI. In this study, we compared the sensitivity of biomarkers using NMR-based metabolites with conventionally used biomarkers of AKI.
◦ Measurement of urinary indexes and blood biochemical parameters were performed. 3-Indoxyl sulfate and trigonelline levels were measured in the serum, urine, kidney, and liver. In the cisplatin-treated rats, urinary blood urea nitrogen (BUN) and creatinine levels were dramatically decreased and protein, glucose and LDH levels were significantly increased, which were thought to be caused by the dysfunction of proximal tubule injury.
◦ 3-Indoxyl sulfate was significantly reduced in urine of Sprague-Dawley rats treated with cisplatin and cyclosporin A, whereas it's levels were significantly elevated in the serum and kidney.
◦ These results suggest that serum 3-indoxyl sulfate and trigonelline may be used as an early biomarker for detecting AKI. It may be possible for measurement of serum and urinary 3-indoxyl sulfate level to replace renal biopsies in evaluation of drug-induced renal toxicity.
