실크단백질 복합체를 이용한 인체조직공학용 스케폴드를 개발하기 위하여 시판되고 있는 인공시멘트, 흡수성 플레이트 및 조직공학용 스케폴드의 형태와 특성을 조사하였으며, 실크피브로인 농도, 분자량, 동결온도, 포로겐 등에 따른 실크 지지체의 다공성 등의 특성 변화를 전자현미경으로 확인하고 결정구조 및 생분해성 등을 측정하였다. Hap, TCP 등 무기염 또는 유기산을 이용하여 인공뼈 소재용 실크스케폴드 및 실크분말의 특성을 분석하고, 치주뼈 형성 촉진을 위한 무기염 복합 실크 소재 및 조직공학용 실크 스케폴드를 제조하여 공극율, 밀도 및 실크 소재의 형태 안정성을 확인하였다.
실크단백질을 주재로 한 인체 적용 소재를 개발하기 위하여 실크 용액의 제조 조건 등을 변화시켜 기계적 강도, 구조 특성 분석 및 동물실험을 통하여 실크 인공고막 소재를 개발하였으며, 일반 실크 생체막에 비해 인장 강도가 약 40배 가량 높고 시판 인공뇌경막에 비해 2.5배정도 강한 특성을 가지는 투명하고 유연한 두꺼운 실크 생체막 (500 ㎛이상)을 제조하여 형태, 구조, 생분해성, 공극의 크기 및 형태 등 특성을 분석하였다.
뼈 형성촉진인자 BMP, DICAM 및 누에 Dpp 등 신소재 생산기술 개발 및 효능을 검증하고, BMP 생산 세포주 및 이를 이용한 뼈 세포분화 유도 기술 개발, BMP 생산 세포주를 이용한 효율적 세포분화 유도체계 개발, 아가로스 및 실크를 소재로 한 3차원 세포배양 기술 및 지지체 개발, 아가로스를 이용한 다공성 지지체 및 LSF 함유 3차원 지지체 개발, 인간 줄기세포주를 이용한 3차원 세포배양 및 분화유도 기술을 개발하였다.
실크 나노섬유를 이용한 생체 이식 및 의료용 소재를 개발하기 위하여 생체막 특성 분석과 인공 생체막 설계를 위한 실크 나노섬유 집합체 제조 조건을 확립하고 나노섬유 막의 생분해성을 조절하고, 실크/하이드록시아파타이트 유무기 복합나노섬유의 제조 및 성능을 평가하였으며, 3차원 실크 피브로인 나노섬유 지지체의 공극크기 및 공극도 조절 기술을 이용한 골 재생용 지지체 개발 및 3차원 실크 피브로인 나노섬유 지지체의 표면개질에 의한 기능화 소재를 개발하였다.
탄소나노튜브 함유 실크단백질의 생체적용 소재를 개발하기 위하여 탄소나노튜브 표면의 극성기 도입을 통한 탄소나노튜브 분산 수용액 제조 및 인공고막용 탄소나노튜브 함유 실크단백질 필름의 제조 및 특성 분석하고, 박테리아 셀룰로오스 나노크리스탈 제조 공정을 확립 및 실크단백질/셀룰로오스 나노크리스탈 복합체 개발 및 탄소노나튜브와 셀룰로오스 나노크리스탈을 이용한 고강도 실크단백질 플레이트 제조 및 특성 분석하였다.

To develop tissue engineering scaffold using silk fibroin and its complex, commercial goods were examined. Preparation conditions such as concentration, porogen etc. for silk scaffolds were tested in the point of morphology, crystalline structure, and biodegradability. Inorganic components including nHap and/or TCP were used to fabricate silk scaffold.
Silk membranes were prepared and tested to evaluate the feasilbility of artificial eardrum and dura mater. To facilitate bone formation using silk fibroin scaffold, BMP and DICAM gene and protein were tested using cell line and tested the function.
In the 1st cooperative subject(Title: Development of implanted biomedical materials using silk nanofibers), the main purpose is to develop the tissue engineering scaffold for bone regeneration using a 2-dimensional(2D) and 3-dimensional(3D) silk nanofibrous scaffold. Fabrication methods, structural characterization, physical/mechanical property measurement of scaffolds were performed with proper biological evaluationsin this subject. In order to develop the silk scaffolds for high performance and functionality, highly porous nanofibrous structure of real 3D scaffold was fabricated using aqueous silk fibroin solution and NaCl porogen through novel electrospinning method in methanol/ethanol coagulation bath. Biodegradability control, pore size control, surface modification with plasma treatment, immobilization with cell adhesion peptide, and other functionalized techniques were carried out for improving the performance of the scaffolds. In the case of silk protein, hybridization with other biopolymers is specially important due to overcome the drawbacks of silk as well as to improve the properties. Here, the silk/hydroxyapatite composite nanofiber scaffold was evaluated to find out its feasibility on specific bone regeneration and silk blend nanofibers of PCL or Keratose were also fabricated for improving physical/mechanical property of the scaffolds. It is expected that high performance 3D scaffold of silk nanofiber can be developed mainly for the bone regeneration through more detailed biological evaluations(in vitro and in vivo test) and this silk nanofibrous scaffold can also expand their application fields into nerve conduit, artificial skin, DDS etc. in a future.
Lots of polymeric materials with biocompatibility and biodegradability are great interested as a tissue engineering material for regeneration and repair of diseased tissues. Silk fibroin produced by the silkworm, Bombyx mori is a fibrous biopolymer that has been used for thousands of years as one of the most important materials in the textile industry and as a medical sutures. Recently, considerable effort has been directed towards its use as a biotechnological material on account of its excellent biodegradability and biocompatibility. In order to be used as a biotechnological material in biomedical applications, it is essential to regenerate silk fibroin in to its proper form of similarities to native extra cellular matrix. However, the crystallinity of silk fibroin decreases during the regeneration process, resulting in a deterioration of the mechanical properties of the regenerated silk compared to natural cocoon fiber. In this study, carbon nanotubes and cellulose nanocrystals with the high mechanical strength and aspect ratio were used as organic reinforcing filler which lead to improve mechanical properties for regenerated silk fibroin. Various processes and methods were developed and investigaed for silk fibroin nanocomposite with carbon nanotube and bacterial cellulose nanocrystals.
