최근 들어 기존의 “의약품 등의 독성평가 시험법”의 개선 필요성이 인식되어 ICH (국제조화협력회의)를 중심으로 독성시험 가이드라인의 변경작업이 이루어지고 있다. 또한, 배 · 태자발생독성시험에서 발생이상에 대한 평가는 판정에 객관성이 요구되나 국내에는 아직까지 표준화된 평가기준이 없다. 따라서 본 연구에서는 유전독성시험과 배 · 태자발생독성시험의 선진화를 위해 국내 GLP 시험기관에 유전독성 시험의 동물대체시험법인 in vitro, in vivo Comet assay 의 평가기반을 마련하고, 표준화된 배 · 태자 발생이상 판정기준 통일안을 확립하고자 하였다. 유전독성분야에서는 국내의 in vivo comet assay 기반 구축을 위해 현재 국제검증연구에서 사용하는 시험법 (version 14)을 국내 3곳의 GLP 기관에게 보급하였으며 확립된 시험법을 바탕으로 in vivo comet assay를 수행, 검증연구를 진행하였다. 또한 아직까지도 문제점이 제기되고 있는 in vitro comet assay 시험법 개선을 위한 근거자료 제시를 위해 기존의 시험법 (version 6.2) 과 본 연구진이 새롭게 제안하는 시험법과의 비교분석을 위해 in vitro comet assay를 수행하였다. 생식 · 발생독성분야에서는 발생이상의 판정기준을 마련하기 위해 GLP 기관과 대학 등 3개 기관이 공동으로 협력연구를 수행하였다. 국내 ·외 시험기관 간 발생이상 평가기준을 조사하여 국내 시험기관에서 사용하는 발생이상 평가기준의 현황과 문제점을 파악하였다. 발생이상 판정기준 마련을 위해 시험 대상물질 6종을 선정하여 배 · 태자발생독성시험을 수행하였고, 참여기관간 점검과 선진외국의 판정기준을 반영하여 발생이상 판정 기준의 초안을 만들었다. 발생이상 판정기준의 초안을 관련 전문가들과의 자문회의를 통해 국내에 적용할 수 있는 참여기관간 판정기준 통일안을 도출 하였다. 총1,720종의 발생이상소견에 대한 판정기준 통일안을 만들었고, 주요 발생 이상에 대 한 사진자료 244종을 확보하였다. 본 연구에서 도출된 판정기준 통일안과 사진자료는 배 · 태자발생이상 자료집으로 개발하여 향후 자료집으로 발간할 수 있도록 정리 하였다. 결론적으로 본 연구결과는 국내 GLP 시험기관의 유전독성과 발생독성의 평가능력과 규제기관의 위해성평가 능력을 국제 수준으로 향상시키는 계기가 될 것이며, 이를 통해 국내 독성 시험기관의 국제화와 독성시험자료의 신뢰성 확보, 국내 타 연구기관에 정보제공 및 교육, 국민보건 향상, 신약개발산업의 지원 등의 성과를 얻을 수 있을 것으로 기대된다. Recently with the recognition of the necessity to improve existing “Guidelines for Toxicity Studies of Drugs" toxicity studies guidelines are being revised mainly led by ICH. In addition, objectivity is required to evaluate developmental abnormalities in embryo-fetal development study, but there are no standardized evaluation criteria in Korea yet. Thus, in order to advance genetic toxicity tests and developmental toxicity tests, this study purposed to lay the base of Comet assay, a genetic toxicity test method alternative to animal tests, at domestic GLP laboratories in Korea, and to establish standard criteria for determining embryo-fetal developmental abnormalities. We educated in vivo comet assay method which is using on international validation study to GLPs for construction in vivo comet assay foundation in Korea. Based on this method, we conducted in vivo comet assay using 2 kind of test chemicals. After collecting data, we peformed inter-institute validation study via statistical analysis. Regarding with in vitro comet assay, we conducted comparative in vitro comet assay on TK6 and HepG2 cell line for having sufficient evidence to revise in vitro comet assay guideline. In order to establish criteria for determining developmental abnormalities, we conducted a collaborative study jointly with three institutions. We compared developmental abnormality evaluation criteria at domestic and foreign laboratories, and surveyed the current state and problems of existing evaluation criteria. In addition, we selected 6 substances to be tested and conducted developmental toxicity tests for them. The results were reviewed by the participating institutions, and then developmental abnormality evaluation criteria were drafted based on evaluation criteria in developed countries. Lastly, domestically applicable standard abnormality evaluation criteria were derived through meetings with specialists in relevant areas. Standard evaluation criteria were prepared for a total of 1,720 types of developmental abnormalities, and 244 photographs showing major developmental abnormalities were collected. The standard evaluation criteria and photographs were organized so that they could be compiled into a reference book on embryo-fetal developmental abnormalities. In conclusion, the results of this study is expected to enhance domestic GLP laboratories' abilities to evaluate genetic toxicity and developmental toxicity and regulatory agencies' abilities to conduct risk assessment. Furthermore, it may contribute to the internationalization of domestic toxicity test laboratories, the reliability of toxicity test data, information provision and education for other research institutes in Korea, people's health, and support to the new drug development industry.
