Ⅳ. 연구개발 결과
○ 인삼 쓴맛 물질 규명
- 진세노사이드 Rb1, Rg1, Rg3, Re, Rc의 쓴맛에 있어서 쓴맛의 강도가 높게 나타 났으며, 서로 간 유의적인 차이가 없음을 확인하였음. 이로 볼 때 진세노사이드는 인삼의 쓴맛에 영향을 주는 하나의 성분으로 확인할 수 있었음.
- 인삼의 주요 페놀의 쓴맛의 관능평가 결과 페놀 추출물 시료는 2.38~4.13점으로 쓴맛의 강도가 낮게 나타났으며, 유의적으로 큰 차이가 없었음. 전자혀를 이용하여 페놀 종류
Ⅳ. 연구개발 결과
○ 인삼 쓴맛 물질 규명
- 진세노사이드 Rb1, Rg1, Rg3, Re, Rc의 쓴맛에 있어서 쓴맛의 강도가 높게 나타 났으며, 서로 간 유의적인 차이가 없음을 확인하였음. 이로 볼 때 진세노사이드는 인삼의 쓴맛에 영향을 주는 하나의 성분으로 확인할 수 있었음.
- 인삼의 주요 페놀의 쓴맛의 관능평가 결과 페놀 추출물 시료는 2.38~4.13점으로 쓴맛의 강도가 낮게 나타났으며, 유의적으로 큰 차이가 없었음. 전자혀를 이용하여 페놀 종류별 쓴맛을 측정한 결과 7.8점으로 caffeic acid의 쓴맛의 강도가 높은 것을 확인할 수 있었으며, 다른 페놀 시료는 2.4~3.1점으로 쓴맛의 강도가 낮게 나타났음. 관능평가 결과와 비교하였을 때 caffeic acid 시료를 제외한 나머지 페놀 시료는 쓴 맛을 인지하는데 있어서 낮게 측정되어 페놀은 인삼의 쓴맛에 영향을 주지 않는 성분인 것으로 사료됨.
- 인삼의 전체 폴리아세틸계 Panaxynol, panaxydol의 전자혀를 이용하여 쓴맛을 측정한 결과 각각 2.9, 4.0으로 쓴맛 강도 값이 낮게 나타나 인삼에 쓴맛에 있어 크게 영향을 주는 성분이 아닌 것으로 확인할 수 있었음.
- 전자혀를 이용하여 알칼로이드의 쓴맛을 측정한 결과 알칼로이드 분획물은 6.9를 나타내었으며, harman, norharmane은 각각 8.0, 7.5의 값을 나타내었음. 이로 볼 때 인삼의 성분 중 하나인 알칼로이드는 인삼의 쓴맛에 영향을 주는 하나의 성분으로 확인할 수 있었음.
- 전자혀를 이용하여 알칼로이드 물질과 폴리아세틸렌 물질 추출물에 대해 쓴맛을 측정한 결과 알칼로이드 시료군이 폴리아세틸렌 시료군보다 쓴맛의 강도 값이 약 두배 정도 높게 측정되었으며, 알칼로이드 샘플 중 harman의 쓴맛 강도의 값이 8.0으로 가장 높게 나타났음. 이와 같은 결과로 살펴볼 때 알칼로이드는 폴리아세틸렌 보다 인삼의 쓴맛에 영향을 주는 물질인 것으로 판단할 수 있음.
- 전자혀를 이용하여 알칼로이드 물질과 진세노사이드 물질에 대해 쓴맛을 측정한 결과 알칼로이드 시료군 보다 진세노사이드 시료군의 쓴맛의 강도 값이 높게 나타났으며, 이 중 진세노사이드 중 Rb1의 쓴맛의 강도가 8.2점으로 가장 높은 것을 확인할 수 있었음. 이로 살펴볼 때 인삼 진세노사이드는 인삼의 쓴맛에 있어 가장 많이 영향을 주는 물질로 판단되어짐.
○ 고기능성 신규홍삼 제조기술 개발
- 고온고압처리를 통하여 원형삼 형태의 고기능성 신규 홍삼을 개발해 보고자 하였음. 인삼의 원형유지를 위해 전처리로 1차 건조공정을 적용하였으며 50℃에서 30~36시간 건조하여 수분함량을 20~30% 수준으로 조절하는 것이 적절하였음. 외형적 품질 및 성분특성을 고려할 때 140℃, 3 ㎏/㎠에서 10~30분 정도 처리하였을 때 페놀성 화합물, 말톨 및 일부 비극성의 활성 진세노사이드 함량을 높이면서도 홍삼과 유사한 외형을 유지할 수 있는 신규 홍삼 제조가 가능한 것으로 판단되었음.
최종적으로 2차 건조방법을 열풍건조와 감압건조로 달리하여 신규홍삼을 제조해 본 결과 감압건조시 건조시간이 단축되고 색과 외형 역시 기존홍삼과 유사하게 나타났음. 또한 미세구조에 있어서도 신규홍삼의 경우 기존홍삼에 비해 대체로 표면 구조가 거칠고 용매의 침투가 용의한 굴곡과 홈을 다량 가진 구조를 나타내어 효율적인 성분 용출이 가능할 것으로 판단되었음.
- 신규홍삼의 성분표준을 구하기 위해 9차례 반복제조 후 외형 및 성분특성을 비교해 본 결과 전체적으로 유사한 외형 특성을 나타내었으며 신규 홍삼(n=6)의 경우 38.4~52.0%(mean: 43.2%)의 총당 함량, 3.12~6.47%(mean: 5.17%)의 산성당 함량, 1.11~3.22%(mean 2.44%)의 총 페놀함량을 나타내었으며 특히 폴리페놀 화합물 중에서 홍삼 특이성분으로 알려진 maltol의 경우 4.66~8.72 mg%(mean: 6.17 mg%), 진세노사이드 Rg3의 경우 1.47~2.48 mg/g (mean: 2.48 mg/g) 함량 수준을 나타내어 기존 홍삼에 비해 높은 maltol 및 진세노사이드 Rg3 함량을 나타내었음. 또한 신규홍삼의 경우 건조인삼 및 시판 홍삼에 비해 각각 22배, 6배 정도 높은 DPPH 소거능을 나타내었으며 ABTS 라디컬 소거능을 역시 각각 4.3배, 4.7배 높은 활성을 나타내어 항산화 및 이와 관련된 항피로 소재로서의 활용성이 높은 것으로 판단되었음.
- 항산화성이 높은 천연소재와의 혼합 추출물 제조를 통해 기능성 및 관능적 품질 특성을 향상시키기 위해 항산화성 활성 및 항피로 효능이 높은 것으로 매실, 오미자, 복분자, 블루베리, 솔잎, 인진쑥와 신규홍삼의 혼합 추출물 제조결과 복분자, 솔잎, 인진쑥과의 혼합추출물 제조시에 폴리페놀 함량 및 항산화력이 증대되었으며 특히 솔잎 혼합 추출물의 경우 비교적 관능적 특성도 우수하였음. 폴리페놀 함량 및 항산화력 증진을 근거로 적정 솔잎 혼합비율은 20% 이상이 적절하였음. 이 경우 총 폴리페놀의 함량은 홍삼의 3.5배, 신규홍삼의 1.9배였으며 DPPH radical scavenging 활성에 있어 5 mg/ml의 농도에서 홍삼의 4.7배, 신규홍삼의 2.6배 증가하는 것으로 나타났음.
- 세포모델을 이용한 신규 홍삼의 산화적 손상 억제효능을 C2C12 muscle cell과 지방세포에서 구명하였다. 신규 제조 홍삼의 산화적 스트레스에 대한 효능을 평가 하기 위해 C2C12 muscle cell에 H2O2를 처리하였고 이에 따른 세포 morphology 및 항산화 관련 유전자를 조사하였음. H2O2를 처리 시 세포의 morphology 및 항산화 관련 유전자 발현이 상당히 감소하는 것으로 나타났지만 신규 제조 홍삼 처리 시 Cu/Zn SOD 및 Mn SOD의 발현량이 증가하는 것으로 나타났고 또한 MRF4의 발현량이 증가하는 것으로 나타났음. 또한 Morphology 변화에서도 대조군과 같은 morphology를 나타내는 것을 확인하였음. 3T3-L1 지방세포의 경우 분화과정에서 과량의 지방을 축적하며 이에 따라 산화적 스트레스 또한 높아지는 것으로 나타났으므로 이러한 지방 세포 분화 시 산화적 스트레스에 대한 신규 제조 홍삼의 억제 효과를 평가하였음. C2C12 세포 모델에서와 마찬가지로 항산화 관련 유전자인 Cu/Zn SOD 및 catalase의 발현량을 증가시키는 것으로 나타났고 이에 따라 지방세포 내 ROS 축적량을 감소시키는 것으로 나타났음.
- 동물모델에서 신규 홍삼의 산화적 스트레스 억제효과를 확인하기 위해 고강도 운동을 실시하여 동물모델 내 산화적 스트레스를 유도하였고 100 mg/kg 농도의 신규 제조 홍삼을 경구투여 하고 현상학적, 혈액학적 및 조직학적 분석을 통하여 산화적 스트레스에 대한 신규 홍삼의 효능을 평가하였음. 고강도 운동으로 인하여 혈액 내 GOT 및 GPT의 효소 활성이 크게 증가되었고 간의 MDA 함량 또한 증가되었지만 신규 제조 홍삼을 섭취한 군에서는 상당히 낮아지는 것을 확인하였음. 근육 내 glycogen함량의 경우 신규 제조 홍삼을 섭취한 군에서 운동군에 비해 상당히 높은 함량을 가지는 것으로 나타났음. 또한, 근육 내 효소 활성을 평가 시 항산화 mechanism의 항상성 유지에 중요한 역할을 하는 G6PDH의 활성이 높았다. 하지만 신규 제조 홍삼의 섭취는 근육 내 LDH의 효소활성을 높이지만 SDH의 경우 그 활성에 영향을 주지 않는 것으로 나타났음. 근육 내 항산화 및 근육 생성 관련 유전자 분석한 결과 신규 제조 홍삼 섭취군에서 상당히 증가된 것으로 나타났음.
- 신규 홍삼의 에너지 생성 및 운동능력 향상에 기여하는지를 평가하기 위해 주요 대사과정 중 하나인 β-oxidation을 평가하였음. β-oxidation은 지방산을 에너지원으로 사용하기 때문에 C2C12 myotubes 내 과량의 지방 축적을 유도한 후 본 연구에 사용하였음. 신규 제조 홍삼 추출물을 세포 처리 시 주요 조절 인자인 AMPK의 phosphorylation이 증가하였고 이에 따라 ACC의 불활성화가 증가하는 것으로 나타났음. Triglyceride로부터 free fatty acid를 생성하는 lipolytic enzymes의 단백질 발현량 평가 시 FoxO1의 핵 내 translocation에 의해 ATGL의 발현량이 증가하였고 LPL의 발현량이 신규 제조 홍삼에 의해 증가되는 것으로 나타났음. 미토콘드리아 내 유리지방산 이동에 관여하는 CPT-1의 발현량의 경우 신규 제조 홍삼 추출물 처리에 의해 상당히 증가되었고, Oil Red O staining 시 근육세포 내 지방함량이 상당히 감소되어 신규 홍삼 추출물은 AMPK/ACC/ATGL/LPL/CPT-1에 의한 β-oxidation을 증진시키는 것으로 나타났음.
- 강제수영모델을 활용하여 신규홍삼의 지구력향상효능을 살펴보았음. 우선 신규홍삼 분말을 1.25~5.00 g/Kg․d 농도로 투여했을 때 체중 증가에는 큰 영향을 미치지 않았으며 수영시간의 경우 대조군에 농도 의존적으로 증가하는 경향을 나타내어 2.50 g/Kg․d 투여시에 53.4%, 5.00 g/Kg․d 투여시에 66.2% 증가하였으며 기존 홍삼 투여시와 비교 시에도 약 14.7% 정도 수영시간이 길어지는 경향을 나타내었음.
또한 신규 홍삼투여는 강제수영으로 인한 혈중 BUN 함량 및 lactic acid 함량을 농도 의존적으로 감소시키는 효과를 나타내었으며 간의 glycogen 함량을 농도 의존적으로 증가시키는 것으로 나타났음.
○ 인삼제품의 소화·흡수 연구
- 개별 진세노사이드의 in vitro digestion을 살펴본 결과 진세노사이드는 위와 소장에서 대부분 대사되었는데, Protopanaxadiol (PPD) 계열인 Rb1은 Rg3를 중간대사체로 하여 compound K (CK)까지 대사되었음. 결과 중 특이적인 사항으로는 Rb1이 대사되는 동안 중간 대사체는 Rg3의 형태로만 나타나고 다른 중간 대사체인 Rd와 F2는 발견되지 않았음. 또한 최종 대사산물이 CK로만 존재하고 Rh2의 형태로 존재하지 않는 것으로 미루어 보아 in vitro digestion 과정이 진세노사이드 Rb1의 3번 및 20번 위치에 있는 glucose의 1-6결합은 분해할 수 있으나, 3번 위치의 1-2 결합은 분해할 수 없는 것으로 사료되었음. Protopanaxatriol (PPT)계열인 Re는 Rg2를 통해 Rh1으로 대사가 되었음.
- 인삼차 및 인삼농축액의 in vitro digestion 결과를 살펴보면 위소화 단계를 거치며 대사체 진세노사이드인 Rg3, CK, Rg2, Rh1이 증가하는 것을 확인할 수 있었음. 이는 단일 진세노사이드인 Rb1, Re의 in vitro digestion 결과와 일치하는 것이었음. 특이한 점은 PPT 계열의 경우 Rg1, Re의 함량이 소화에 따라 감소하며 그의 대사체인 Rg2, Rh1이 증가하는 반면, PPD 계열의 경우 Rb1, Rb2, Rc의 함량감소 없이 이들의 대사체인 Rg3, CK가 증가한 것임. 이는 아마도 차와 농축액내에 존재하고 있는 Rb1, Rb2, Rc의 malonyl 형태들이 대사되어 Rb1, Rb2, Rc로 전환되기 때문으로 사료되었음. 또 다른 인삼제품인 백삼분말의 경우 소화액으로 용출되어 나오는 진세노사이드들의 경우 대사체 진세노사이드인 Rg3, Rh2등으로의 전환이 확인되었지만, 용출되지 않은 백삼분말내의 진세노사이드들의 경우에는 Rg3로 소량 전환되는 것을 확인할 수 있었음. 이는 소화액내로 용출되어 생체접근성(bioaccessibility)을 확보한 진세노사이드들의 경우 용이하게 대사체로 전환되는 반면, 생체접근성을 확보하지 못한 진세노사이드들의 경우 대사체로의 전환이 용이하지 못하다는 사실을 증명하는 결과임.
- 진세노사이드 Rb1의 만복(Fed) 및 공복(Fast)시의 in vitro 소화형태를 비교해 보기 위해 위 소화 단계의 pH를 1.3, 2, 3, 4, 5, 6으로 설정하여 실험을 진행해 본 결과, 위 소화 단계의 pH가 1.3에서 2.0으로 증가하였을 경우 대사체 진세노사이드인 Rg3 및 CK로의 전환이 줄어드는 것을 확인할 수 있었으며, pH가 3.0 이상으로 증가할 경우 전환이 이루어지지 않았음. 인삼제품인 인삼농축액의 소화에서도 진세노사이드 Rb1과 동일한 결과를 얻을 수 있었음. 이는 위 소화 단계의 pH가 소화에 의한 진세노사이드 전환에 중요한 역할을 한다는 것을 시사해 주는 결과로 소화에 의한 진세노사이드의 전환은 만복 시 보다 공복 시에 더욱 활발하게 진행될 수 있을 것으로 사료됨.
- Caco-2 세포를 이용한 개별 진세노사이드(Rb1, Rb3, Rc, Rd, Re, Rf, Rg1, Rg2, Rh2)의 흡수실험 결과 진세노사이드의 투과율은 개별 진세노사이드의 분자량과 비례하였는데, 분자량이 작을수록 더욱 빠른 투과율(R2=0.8842)을 보였음. 이는 진세노사이드의 대사가 진행될수록 장내흡수가 촉진된다는 이전의 연구결과와 동일한 경향을 보이는 것이었음.
○ 국내산 인삼제품의 품질특성 표준화 연구
- 백삼의 외형적 품질 특성 및 주요성분 함량에 대한 평가와 통계분석을 통해 표준품질특성을 도출해 보고자 하였음. 시료는 시판중인 백삼제품중 곡삼(curved ginseng) 4년근 30, 50편급(piece grade), 피부직삼(straight ginseng) 15, 25편급을 선정하여 2006년부터 2010년까지 5년에 걸쳐 일정량씩 수집하였으며 weight, length, diameter and surface color 등의 외형적 품질 특성(Morphological characteristics)을 측정한 후 기초통계 분석한 후 최대 및 최소값으로 품질특성치의 범위를 나타낸 결과, 곡삼 50과 30편급의 경우 각각 4.9~6.1 cm와 5.6~7.3 cm의 길이, 0.8~1.8 cm와 0.6~1.4 cm의 직경, 5.5~7.2 g과 9.4~11.7 g의 중량을 나타내었으며 5년근(5-year-old) 피부직삼 25와 15편급의 경우 10.5~14.7 cm와 12.0~16.3 cm의 길이, 1.4~1.9 cm와 1.7~2.5cm의 직경, 10.1~16.0 g와 16.8~23.6 g의 중량을 나타내었음. 표면색도(surface color)의 경우 동일한 종류의 백삼제품에는 편급간 차이가 거의 없었으며 백삼종류별 보면 직삼은 밝기(brightness(L))는 낮은 반면 적색도(redness(a))와 황색도(yellowness(b))는 다소 높은 것으로 나타났음.
- 주요 백삼제품인 곡삼 4년근과 피부직삼 5년의 편급별 이화학적 성분 특성을 분석한 후 통계분석하여 표준 성분 함량을 구명해 보고자 하였다. 평균값을 기준으로 가용성 총당 함량은 4년근 30과 50편급의 경우 33.31%와 33.81%, 직삼 5년근 15와 25편의 경우 각각 36.35%와 36.94%이었으며 산성 다당체 함량은 30편급 5.19%, 50편급 5.35%이었으며 피부직삼 15편은 4.33%, 25편은 4.93%이었음. 곡삼 4년근 30편급의 총페놀화합물 함량의 경우 곡삼은 편급에 상관없이 0.37%, 피부직삼은 15편급이 0.45%, 25편급이 0.42%의 총페놀화합물 함량을 나타내었음. 인삼 대표 활성성분인 조사포닌함량의 경우 모든 시료에서 기준규격인 2%이상의 함량을 나타내었음.
주요 ginsenoside인 Rg1, Rf, Rb1 함량의 경우 곡삼 4년근 30편급의 경우 2.72, 0.79, 3.14 mg/g, 50편급은 2.37, 0.74, 2.97 mg/g이었으며 피부직삼 5년근의 경우 15편급은 3.51, 1.09, 4.79 mg/g, 25편급은 3.02, 1.00, 3.89 mg/g이었음. 전체적으로 산성다당체, 총페놀화합물, 조사포닌과 주요 진세노사이드 분석결과, 변화량 계수값이 20% 이상으로 나타내 평균값을 활용하여 성분함량을 표준화하기 어려웠으며 백삼의 종류의 다른 성분 함량 차이는 있었으나 동일 백삼제품의 편급간 성분함량차이는 거의 없었음.
- 저년근(4년근) 홍삼제품의 대, 소 크기별 이화학적 성분 특성을 분석한 후 통계 분석하여 표준 성분 함량을 구명해 보고자 하였음. 평균값을 기준으로 대편과 소편의 경우 각각 36.5%와 35.5%, 산성 다당체 함량은 5.2%, 50편급 5.4%이었으며 총페놀화합물 함량의 경우 크기에 상관없이 0.40% 수준이었음. 조사포닌 함량의 경우 모든 시료에서 기준규격인 2%이상의 함량을 나타내었으며 대편이 4.65%, 소편이 4.23%로 오히려 대편이 다소 높은 조사포닌 함량을 나타내었음. 주요 ginsenoside인 Rg1, Rf, Rb1 함량의 경우 대편은 각각 2.72, 0.70, 3.85 mg/g, 소편은 2.35, 0.64, 3.95 mg/g이었음. 저년근 홍삼의 경우 현재 실험을 진행중에 있으며 추가적으로 2~3년정도 자료를 확보한 후 최종적으로 성분함량에 대한 표준품질 특성안을 구명할 예정임.
○ NIR을 이용한 인삼의 품질 특성 구명
- 인삼의 픔질괸리 방안의 일환으로 NIR 적용 기술을 확립하기 위하여 시중에서 시판중인 4년근 곡삼, 5년근 직삼, 5년근 홍삼을 구입하여 분말화하여 NIR(perten DA 7200, VELP Co.) 원시 data를 획득한 후 PLS 기법을 이용하여 회귀선을 작성함.
- 수분은 대편 인삼의 경우 결정계수가 0.8463, 소편의 경우 0.8918로 비교적 높은 상관관계를 보였으며, 사포닌과 개별 진세노사이드인 Rb1과 Rg1의 결정계수를 살펴보면 각각 0.91114, 0.9279, 0.9992로 상당히 높은 결과를 보이고 있으나, 소편의 경우 0.7988, 0.9994, 0,7151로 다소 낮게 나타났음.
- 인삼 추출액의 유효성분인 사포닌과 개별 진세노사이드 함량을 NIR을 이용하여 신속하게 측정하기 위한 기술을 개발하고자 시판 홍삼 파우치에 대한 NIR 분석과 HPLC 분석을 통하여 예측값과 실측값간의 상과관계를 조사하였음.
- 인삼 추출액(파우치)의 조사포닌 함량은 0.54 mg/mL∼1.47 mg/mL의 범위였으며, NIR의 검량곡선의 R2 값은 0.6974였으며, 미지시료에 대한 예측값과 실제 측정값간의 상과계수는 0.7864였음.
- 개별 진세노사이드인 Rb1과 Rg1의 예측 검량선을 구한 결과 Rb1은 0.8761, Rg1은 0.9105으로 상대적으로 높은 값을 나타내었으며, 미지 시료에 대한 예측값과 실제 측정값간의 상과계수는 0.8664였음.
○ 인삼잎 및 열매발효 미용소재의 개발
- 인삼씨 추출물은 ginsenoside 함량 및 페놀성 성분 함량이 매우 낮았으나 세포독성 없이 뛰어난 피부색소 억제작용을 나타내었음.
- 인삼 잎 역시 우수한 피부미백활성과 ABTS 소거활성을 나타내었으며 물 추출물의 활성이 더 우수하였고 이의 활성성분은 페놀성 성분인 esculetin으로 판단됨.
- 인삼씨의 초임계 추출물 높은 미백활성을 나타내지 않았으며 인삼시료의 발효소재는 KCTC5033 균주로 발효할 때 다른 균주 발효물에 비하여 우수한 미백활성을 나타내었음.
○ 신규 진세노사이드 분리기술과 함량 및 생리활성 규명
- 인삼 ethanol extract는 n-hexane, CH2Cl2, EtOAc and n-BuOH(181.0g)의 극성별 유기용매를 통해 각각의 분획물을 얻은 후, chromatography를 통해 물질을 분리 정제하며 이 과정을 순수하게 분리하기 위해 반복 진행하였음. n-BuOH 분획물을 MPLC를 통하여 fractions 1-6을 분리하였으며, sub fraction 5에 대하여 Lichroprep을 통하여 물질을 분리 정제하였음.

- 인삼분야 연구논문 정보는 NDSL(KISTI 운영 국가과학기술정보)에서 2011년 이후 신규논문을 458건 업데이트 하였으며 현재 총 3,238건의 국내외 인삼관련 학술논문을 서비스 하고 있음. 인삼분야 국내외 특허는 2011년 이후 신규특허 357건을 업데이트 완료하였으며 현재 6,455건을 인삼정보센터에서 서비스 하고 있음. 연구사업보고서는 총 399건의 국내외 연구사업결과 보고서를 수집하여 인삼정보센터에서 서비스하고 있음. 인삼정보센터에 업로드된 논문 DB, 특허 DB는 서지사항을 기준으로 매핑하여 원문정보를 한국과학기술정보연구원의 NDSL및 KPRIS(한국특허정보원)과 링크하여 원문보기도 가능하도록 개선함.

- The results of sensory evaluation on the bitterness of major phenolic compounds in ginseng showed that the bitterness was low in phenol extract samples at 2.38~4.13 and was not significantly different among samples. The bitterness by phenol types as measured by electronic tongue was 7.8, and the intensity of bitterness of caffeic acid was high; that of other phenolic samples was low as 2.4~3.1. Compared to the results of the sensory evaluation, all phenolic compound samples except the caffeic acid sample measured low in terms of detecting the bitterness; thus, phenolic compounds are considered not to affect the bitterness of ginseng.

○ Development of new high-functionality red ginseng manufacturing technology
- The study was conducted to develop new high-functionality red ginseng products with the original ginseng shape through high-temperature, high-pressure treatment. The preliminary drying process as a pretreatment was applied to maintain the original shape of ginseng, wherein samples were dried at 50℃ for 30~36 hours to control the proper moisture content to 20~30%. By considering the appearance quality and ingredient characteristics, manufacturing new red ginseng that can maintain the appearance similar to that of red ginseng was possible while increasing the contents of phenolic compounds, maltol, and some non-polar active ginsenosides when treated at 140℃, 3 ㎏/㎠ for 10~30 minutes. Finally, hot air drying and vacuum drying were used as secondary drying method in manufacturing new red ginseng; in vacuum drying, the drying time was shortened, and the color and appearance were also similar to those of the normal red ginseng. For microstructure, the new red ginseng had structures with relatively rough surfaces as well as many curves and grooves for easier solvent penetration compared to the normal red ginseng, suggesting possibly efficient component elution.
- To figure out the standardized content of major component for new red ginseng, the manufacturing process was repeated for 9 times, and the appearance and physico-chemical characteristics were compared; overall, the new red ginseng showed similar appearance. In case of new red ginseng (n=6), total sugar content was 38.4~52.0% (mean: 43.2%), acidic sugar content was 3.12~6.47% (mean: 5.17%), and total phenolic compounds were 1.11~3.22% (mean 2.44%). In particular, maltol was 4.66~8.72 mg% (mean: 6.17 mg%), and ginsenoside Rg3 was 1.47~2.48 mg/g (mean: 2.48 mg/g); thus showing higher maltol and ginsenoside Rg3 contents compared to the normal red ginseng. Furthermore, the new red ginseng showed 22 times and 6 times higher DPPH scavenging activity compared to dried ginseng and normal red ginseng, respectively, and 4.3 times and 4.7 times higher ABTS radical scavenging activity, respectively, suggesting its higher usability as antioxidant and related anti-fatigue ingredient.
- To improve functional and sensory quality characteristics through the manufacture of extract mixture with natural materials with high anti-oxidant activity, the extract mixture of new red ginseng was manufactured using Maesil (Prunus mume), Omija (Scisandra chinensis fruit), Bokbunja (Rubus coreanus Miguel), blueberry, pine needle, and Mugwort (Artemisia capillaris), which are all known to have higher anti-oxidant activity and anti-fatigue efficacy. Polyphenol content and anti-oxidant activity increased when manufacturing the extract mixture with Bokbunja, pine needle, and Mugwort. In particular, the pine needle mixture showed better sensory characteristics. The proper ratio of pine needles was above 20% based on the polyphenol content and anti-oxidation improvement. In this case, total polyphenol content was 3.5 times higher than red ginseng and 1.9 times higher than new red ginseng only. For DPPH radical scavenging activity, it was 4.7 times higher than red ginseng and 2.6 times higher than new red ginseng only at a concentration of 5 mg/ml.
- C2C12 muscle cells were exposed to H2O2, which is known for inducing oxidative stress in the cells, and then treated with new red ginseng. In morphological cell image analysis, New red ginseng protected C2C12 muscle cell damage against oxidative stress induced by H2O2. In addition, new red ginseng enhanced the mRNA expression related to antioxidant enzymes such as Cu/Zn SOD and Mn SOD as well as MRF4 which is associated with myogenesis of C2C12 muscle cell.
- It has been reported that 3T3-L1 fibroblasts increase the ROS production when they were differentiated into adipocytes. For this reason, 3T3-L1 cells were treated with new red ginseng during the differentiation. At day 8, new red ginseng remarkably decreased the ROS production when compared to CON group. In addition, the new red ginseng promoted the mRNA expression of antioxidant enzymes such as catalase and Cu/Zn SOD
- The efficacy of new red ginseng on oxidative stress was investigated in in vivo model. The oxidative stress in mice was induced by high intensity exercise and the mice were supplemented with new red ginseng at the concentration of 100 mg/kg. Although the GOT and GPT activities as well as hepatic MDA level was increased by high intensity exercise, the group supplemented with new red ginseng (E+NGE) had decreased activities of GOT and GPT and had lower hepatic MDA level than E group. In addition, E+NGE group had higher glycogen contents when compared to E group. In the case of analysis of enzyme activities in gastrocnemius tissues, G6PDH and LDH activities were increased by supplementation with new red ginseng, but SDH activity did not be modulated by new red ginseng. In molecular level, mRNA expression levels of Cu/Zn SOD and MRF4 were significantly increased by new red ginseng when compared to the other groups.

To assess β-oxidation modulation, C2C12 muscle cells were used after lipid was accumulated by palmitate. New red ginseng extract increased the phosphorylation of AMPK and ACC which are the major elements of β-oxidation. In addition, new red ginseng upregulated the LPL protein level and also increased the protein level of ATGL by enhancing FoxO1 translocation into nuleus. In the case of CPT-1, the mRNA expression was increased by new red ginseng via AMPK signaling pathway. As a result, the accumulated lipid in C2C12 myotubes was hydrolyzed into free fatty acid by new red ginseng and β-oxidation was increased via AMPK/ACC/FoxO1/ATGL/LPL/CPT-1
- The effect of new red ginseng product on improving endurance was studied using a forced swimming test model. When the new red ginseng powder was administered at a concentration of 1.25~5.00 g/Kg․d, the body weight was not affected, and the swimming time was increased in a dose-dependent manner compared to the control group with 53.4% at 2.50 g/Kg․ d and 66.2% at 5.00 g/Kg․d and 14.7% longer swimming time compared to the normal red ginseng administration. Moreover, the administration of new red ginseng reduced the forced swim-induced BUN content and lactic acid content in the blood in a dose-dependent manner and also increased the glycogen content in the liver in a dose-dependent manner.
○ Study on the digestion/absorption of ginseng products
- The results of in vitro digestion of individual ginsenoside showed that ginsenosides were mostly digested in the stomach and the small intestine. Rb1, kind of the Protopanaxadiol (PPD) family, was metabolized through Rg3 as an intermediate metabolite to compound K (CK). A specific finding among the results was that intermediate metabolites during the metabolism of Rb1 were only observed in the form of Rg3. Other intermediate metabolites such as Rd and F2 were not found. The final metabolite was presented only as CK and not in the form of Rh2, suggesting that the in vitro digestion process can degrade the glucose 1-6 binding at positions 3 and 20 of ginsenoside Rb1 but not the 1-2 binding at position 3. Re, kind of the Protopanaxatriol (PPT) family, was metabolized through Rg2 to Rh1.
- The results of in vitro digestion of ginseng tea and ginseng extract showed that metabolite ginsenosides Rg3, CK, Rg2, and Rh1 increased during gastric and intestinal digestion. This result was consistent with the in vitro digestion result of single ginsenoside Rb1 and Re. The specific finding among the results was that, in case of PPT family, the amount of Rg1 and Re was decreased by digestion, and their metabolites Rg2 and Rh1 were increased. In the case of the PPD family, the amount of Rb1, Rb2, and Rc was not changed, and their metabolites Rg3 and CK were increased. The malonyl forms of Rb1, Rb2, and Rc present in tea and extracts were assumed to have metabolized and transformed into Rb1, Rb2, and Rc. In the case of another ginseng product, white ginseng powder, the conversion into metabolite ginsenosides such as Rg3 and Rh2 was found if ginsenosides were eluted into the digestive fluid. However, the conversion to only a small amount of Rg3 was found if ginsenosides were not eluted into digestive fluid from white ginseng powder. These results confirmed that ginsenosides eluted into the digestive fluid and which had bioaccessibility could be easily converted into metabolites. However, the ginsenosides lacking bioaccessibility could not be easily converted into their metabolites.
- To compare the in vitro digestion of ginsenoside Rb1 in fed and fast situations, the experiment was performed by setting the pH of the gastric digestion process as 1.3, 2, 3, 4, 5, and 6. When the pH was increased from 1.3 to 2.0, conversion to metabolite ginsenoside Rg3 and CK was decreased. When pH was increased above 3.0, conversion was not observed. The same results were obtained in the digestion of ginseng extract as in the case of ginsenoside Rb1. This suggests that the pH in the gastric digestion process plays an important role in ginsenoside conversion during digestion. Therefore, the conversion of ginsenosides by digestion may be more active in fast situation than in fed situation.
- The results of absorption experiment in individual ginsenoside (Rb1, Rb3, Rc, Rd, Re, Rf, Rg1, Rg2, and Rh2) using Caco-2 cells showed that the transmissivity of ginsenosides was proportional to the molecular weight of the individual ginsenoside, which was faster transmissivity (R2=0.8842) as the molecular weight became smaller. This was consistent with the previous results, i.e., intestinal absorption was promoted as the metabolism of ginsenosides proceeded.
○ Establishment of qualities standards draft for domestic ginseng products.
- To figure out the morphological properties and chemical components contents of commercial Korean white ginseng products, we collected 30 and 50 pieces of curved ginseng(4 years old), 15 and 25 pieces of straight ginseng(5 years old) with various sizes and different processing methods, every year from 2006 to 2010. Morphological properties (weight, length, length, rootlet length, diameter and surface color) and the contents of water, acidic polysaccharide, total polyphenolic compound and ginsenosides were examined and statistical analysis was conducted.
- In the basis on max and min values, the morphological properties for curved ginseng of 50 and 30 pieces were as follows: diameter of 0.8~1.8 cm and 0.6~1.4 cm; length of 4.9~6.1 cm and 5.6~7.3 cm, weight of 5.5~7.2 g and 9.3~11.7 g ; surface brightness (L) of 69.50~83.80 and 66.42~80.89, respectively. In the case of 5-year-old straight white ginseng of 25 and 15 pieces, the morphological properties were as follows: diameter of 1.4~1.9 cm and 1.7~2.5, length of 10.5~14.7 cm and 12.0~16.3 cm, weight of 10.1~16.0 g and 16.8~23.6 g, surface brightness (L) of 57.96~74.35 and 59.82~74.35, respectively. There was less than 10% deviation in terms of length, weight, and surface brightness. These properties were applicable to standard parameters for white ginseng products.
On the other hand, properties such as diameter, headlength and surface redness showed impermissible deviation, higher than 20% of CV.
- The content of major components such as moisture, total sugar, acidic polysaccharides, total phenolic compounds, and saponin were analyzed to examine the standard quality characteristics. The moisture content of all dried ginseng samples was under 15% which complies with the reference defined in the domestic laws and regulations. Based on the mean values, the total water-soluble sugar content was 33.31 and 33.81%, respectively, in Curved ginseng (CG) 30 and 50 piece-grade, and 36.35 and 36.94%, respectively, in straight ginseng (SG) 15 and 25 piece-grade. The acidic polysaccharides content was 5.19 and 5.35%, respectively, in CG 30 and 50 piece-grade, and 4.33 and 4.93%, respectively, in SG 15 and 25 piece-grade. The total phenolic compound content was 0.37% in CG regardless of the piece-grade, and 0.45 and 0.42%, respectively, in SG 15 and 25 piece-grade. The crude saponin content, which is the representing active component of ginseng, was over 2% in all samples. The content of major ginsenosides, Rg1, Rf, and Rb1, was 2.72, 0.79, and 3.14 mg/g, respectively in CG 30 piece-grade; 2.37, 0.74, and 2.97 mg/g, respectively in CG 50 piece-grade; 3.51, 1.09, and 4.79 mg/g, respectively, in SG 15 piece-grade; 3.02, 1.00, and 3.89 mg/g, respectively, in SG 25piece-grade. Inoverall, the analytical results of acidic polysaccharides, total phenolic compounds, crude saponin, and major ginsenosides showed that the coefficients of variation was over 20% and that it was difficult to standardize the component contents using the mean value. There were differences in component content between the types of DG, not in the same types of DG. The results of this study are thought useful as basic data for selection of standard samples and standardization of component content for the verification of efficacy of DG in the future.
- The content of major components of domestic 4 year-old red ginseng products were analyzed to examine the standard quality characteristics. The moisture content of all dried ginseng samples was under 15% which complies with the reference defined in the domestic laws and regulations. Based on the mean values, the total water-soluble sugar content was 36.5 and 35.5% and the acidic polysaccharides content was 5.2 and 5.4%, respectively in large and small grade. The total phenolic compound content was approximately 0.4% in regardless of size. The crude saponin content, which is the representing active component of ginseng, was over 2% in all samples. The content of major ginsenosides, Rg1, Rf, and Rb1, was 2.72, 0.70, and 3.85 mg/g of large grade, 2.35, 0.64, and 3.95 mg/g of small grade, respectively. In overall, additional analysis next 2~3 years shall be necessary to figure out standard quality drafts for young(4-year-old) red ginseng products.
○ Investigation on the quality characteristics of ginseng using NIR
- To establish the NIR application technology as part of ginseng quality management, 4 year-root curved ginseng, 5 year-root straight ginseng, and 5 year-root red ginseng were purchased in the market and pulverized to obtain NIR (Perten DA 7200, VELP Co.) raw data. The regression line was then plotted using the PLS technique.
- For moisture, coefficient of determination was 0.8463 for big size ginseng pieces and 0.8918 for small size ginseng pieces, showing relatively high correlation; coefficient of determination of saponin and individual ginsenoside Rb1 and Rg1 was considerably high with 0.9114, 0.9279, and 0.9992, respectively, but slightly low in small size ginseng pieces with 0.7988, 0.9994, and 0,7151, respectively.
- To develop technology for the quick measurement of active ingredients in the ginseng extract, saponin, and individual ginsenoside contents using NIR, the correlation between the predictive value and conventional measurement was examined through NIR analysis and HPLC analysis of red ginseng pouch products in the market.
- The crude saponin content of ginseng extract (pouch) was in the range of 0.54 mg/mL∼1.47 mg/mL, and the R2 value of the calibration curve for NIR was 0.6974. The correlation coefficient between the predictive value and the conventional measurement of the unknown sample was 0.7864.

○ Separation and quantification of new ginsenosides and verification of their physiological activities
- Each fraction of ginseng ethanol extract was obtained through organic solvent by polarity such as n-hexane, CH2Cl2, EtOAc, and n-BuOH(181.0g), and then separated and purified by chromatography; the process was repeated for pure separation. The n-BuOH fraction was separated by MPLC into fractions 1-6; for the sub fraction 5, the substances were separated and purified through Lichroprep.

Currently, a total of 3,238 cases of domestic and foreign ginseng-related scientific papers are in service. For domestic and foreign patents in the ginseng research area, 357 cases of new patent have been updated since 2011. Up to now, 6,455 cases have been in service at the ginseng information center. For research project reports, a total of 399 domestic and foreign research project reports have been collected and are in service at the ginseng information center. The thesis DB and patent DB uploaded at the ginseng information center have been mapped on the basis of bibliographical matters, and the information on original articles is linked to KISTI's NDSL and KIPRIS to enable browsing the original papers.
- The currently operating ginseng information center is operated as consigned management by KISTI. To transfer the operating body to KFRI, the operation environment and systems were examined, and the adequacy of homepage operation and functions was checked.
