□ 연구의 목적 및 내용
본 연구는 생체친화적 환경에서 가교결합의 형성과 외부 자극에 의해 분해가 가능한 유기고분자 소재를 개발하여 살아있는 세포를 인공껍질로 캡슐화한다. 고분자로 세포를 캡슐화하는 과정에서 실시간으로 만들어지는 가교겹합을 이용하여 단단한 껍질을 형성하고, 생체친화적인 외부 자극으로 캡슐을 원하는 시점에 분해하고자 한다. 나아가 인공껍질로 세포를 보호하고 세포의 분열을 제어할 수 있음을 보여주고자 한다. 또한, 외부환경 변화에 취약한 동물세포를 자극 감응성 고분자 물질로 캡슐화하여, 자연에 존재하는 미생물의 보

□ 연구결과
단단한 물질로 단일세포를 캡슐화하는 현재의 연구에서 한 단계 더 나아가, 외부자극에서 의해서 분해되거나 단단하게 변형되는 인공껍질로 세포를 캡슐화하는데 성공하였다. 1) 이황화결합을 포함한 고분자를 합성하였고, 이를 이용하여 실시간으로 가교결합이 일어날 뿐만 아니라 원하는 시점에 외부 자극으로 분해할 수 있는 새로운 형태의 다층박막적층법을 개발하였다. 생체친화적인 환경에서 가교결합과 분해를 모두 일으킬 수 있다는 점에서 기존의 방법과는 차별화될 뿐만 아니라, 생물체에 넓게 응용할 수 있다. 2)이 방법을 살아있는 세포에 적용하여, 살아있는 세포를 캡슐화하는데 성공하였다. 단단한 캡슐의 형성하기 위해서 무기물에 의존하던 기존의 방식에서 벗어나, 유기고분자도 충분히 단단한 캡슐을 만들어 세포를 보호할 수 있음을 보여주었다. 또한, 원하는 시점에 세포의 생존력에 영향을 주지 않고 캡슐 분해할 수 있음을 보여줌으로써, 자연 상에 존재하는 포자와 더욱 유사한 형태의 인공포자를 구현하였다. 3) 더 나아가, 연약한 세포막을 가지고 있기 때문에 환경 변화에 민감한 동물세포를 타닌산으로 캡슐화하는데 성공하였다. 외부의 독성물질로부터 동물세포를 보호할 수 있음을 보여주었고, 원하는 시점에 외부 자극을 가하여 동물세포의 분열을 제어할 수 있었다.
□ 연구결과의 활용계획
세포 캡슐화 연구는 화학적 관점에서 세포의 기능을 향상시켜 유용하게 사용할 수 있다는 점에서 학문적으로 새로운 접근법이라고 볼 수 있다. 수동적/집단적 관찰 대상으로서의 세포에서, 개별적/능동적 제어 대상으로서의 세포로 발상 및 개념의 전환점을 제시한다. 이러한 발상의 전환은 “단일세포 생물학(single-cell biology)”이라는 새로운 분야를 개척하는 시작점이 될 것이다. 응용적 측면에서 있어서, 인공껍질 속의 세포는 세포이식 및 세포 치료 분야에 직접 이용될 수 있을 뿐만 아니라. 의/약학적 응용성을 가진 바이오센서, 바이오칩 등의 구현을 위한 원천 기술이 된다. 실제로 현 연구를 통해서 개발된 분해성 고분자 박막은 특허로 출원되어서 실용화 가능성을 타진하고 있다. 세포 캡슐화는 의학, 약학, 바이오산업 분야에 새로운 방법론을 제시함으로써, 관련 분야의 발전을 크게 가속화시킬 것이다.

It was developed a simple and biocompatible coating technique that cross-links the layers through covalent bonds under physiological conditions without requirement of any post-treatments or additives. By utilizing the dynamic nature of the disulfide bond or bio-inspired reactions, stable films can be spontaneously formed during the process, further functionalized by biocompatible reactions, and degraded in response to chemical cues. In addition, we demonstrated that the method is applicable to plate surfaces, spherical nanoparticles, and even living cells. Especially, encapsulation of living cells with degradable polymer was beneficial to imitate natural spore which can endure in unfavorable conditions and restart their life.

It was succeed that the living cells were encapsulated within artificial shells degradable by stimuli-response.
1) It was developed cytocompatible method for assembling and disassembling cross-linked LbL films in a programmed manner, based on the in situ thiol–exchange reaction.
2) The living cells were encapsulated by applying the method while maintaining the viability of the cells. It was demonstrated that the organic polymer, as well as inorganic materials can be used for artificial shells because the mechanical strength can be improved by cross-linking. In addition, it was possible to degrade the artificial shell by external stimuli, which resembles the natural spore.
3) Furthermore, mammalian cells were successfully encapsulated by tannic acids and the cells were protected by artificial shells and cell division was also controlled by degrading the shells. Considering the fragileness of the mammalian cells, it is one step forward to the biomedical application of living cells.
□ Expected Contribution
Cell encapsulation is a new approach to controlling the biological behavior of living cell and enhancing function of the cells by chemical methods.
Development of the concept of artificial spores and concomitant cytocompatible approaches to formation of artificial materials onto single cells hold great promise for the understanding of fundamental cell processes at the single-cell level, as well as for the development of many cell-based applications. Demonstrations of enhanced viability, control of cell division, protection against foreign aggression, and chemical functionalization, are the basis for the realization of artificial spores, and for further developments involving chemical manipulation and control of cellular metabolism at the single-cell level.
