Purpose&contents Primary cilia are ubiquitous cellular appendages that play a role in Hedgehog, Wnt, and PDGF signaling. Defects in the structure or function of primary cilia cause various human diseases including cancer, polycystic kidney, and genetic disorders affecting the brain, the retina and the digit. Therefore it is critical to understand that regulatory mechanisms governing ciliogenesis and ciliary functions. Expected Contribution Recent research results have suggested that because of the far-reaching impact of primary cilia on cell signaling, we are unable to understand cell signaling at systems levels without deeply understanding the mechanisms of primary cilia. The ciliogenesis regulators identified in this study may serve as a regulatory hub in cell signaling network. Therefore, the ciliogenesis regulators will be utilized as targets of drug development for diseases that are caused by misregulation of cellular signaling, such as cancer and psoriasis.
