연구의 목적 및 내용
본 연구는 다양한 생화학 분자에 대한 높은 선택성(specificity)과 민감도(sensitivity)를 가진 포유동물의 후각조직과 수용액상에서 저전압 구동이 가능한 용액공정 트랜지스터 어레이가 결합된 초고감도 생화학 분자 센서용 생체조직-전자소자 인터페이스 개발을 목적으로 한다.
연구결과
본 연구는 살아있는 세포와 전자 디바이스간의 상호작용을 통해, 고감도 및 고선택성을 갖는 체세포 기반의 미래형 바이오센서 개발을 목표로 전해질 수용액에 장기간 구동가능한 소자의 개발과, 세포-소자 인터페이스 기반 연구로 나누어 진행하였다. 후각세포조직 인터페이스를 구현하기 위해서, 수용액 구동이 가능한 저전압 고성능 금속산화물 기반 디바이스 구현을 시작으로, 전기화학적 실험법을 통해 상호 계면 안정성과, 장기적 구동 안정성을 확보하였다. 나아가, 전해질의 pH에 따라 선택적인 구동전압의 변화를 발생시키는 산화물 반도체 기반 pH센서를 개발하고, 그 선택성을 이온농도, 이온종, pH의 변화를 통해 확인 하였다. 추가적으로, 현재 생체전자소자 인터페이스 연구에서 가장 활발히 연구되고 있는, 티오핀 기반 유기반도체(PEDOT:PSS)의 용액매개결정화법을 통하여, 장기간 구동이 가능한 고성능 전기화학 디바이스 구현에 성공하였으며, 열적, 전기적 피로에도 강한 내성을 가짐을 확인 하였다.
세포-소자 인터페이스의 관점에서도, 세포-반도체 소재, 조직-반도체 소재간의 독성을 평가하여, 앞서 개발한 소재가 세포의 생장에 영향을 끼치지 않는 것을 확인 하였다.
실시간 세포신호측정 기반 기술을 구축하기 위해, 유기물 반도체 기반 마이크로 전극 어레이(microelectrode arrays; MEA) 제작 기술을 개발하여 소자 표면에 직접적으로 배양된 세포의 신호를 효율적으로 읽을 수 있음을 확인하였다.
연구결과의 활용계획
본 연구 과제를 통해 성공적으로 개발된 여러 전해질 구동 전자소자와 세포-소자 인터페이스 구축과 더불어, 향후 후속연구를 통해 후각조직-전자소자 인터페이스의 개발을 실현시킬 것이다. 이러한 감각세포와 전자소자 인터페이스의 도입은 고감도 생화학 분자검출 및 세포 신호 측정의 새로운 분석을 가능케하여, 극미량의 생화학 분자 병원체 마커, 바이러스, 박테리아 등의 검출을 통한 질병 진단에 활용 될 것이다. 더불어, 마약과 같은 금지약물의 탐지, 지하자원 발굴 등 비생명과학의 여러 분야에서도 광범위한 이용이 가능할 것으로 예상한다. 궁극적으로, 2차원 전기신호 맵핑이 가능하고, 인체의 감각기와 매우 유사한 전자소자 구현을 바탕으로, 감각기관(시각, 촉각, 미각, 청각) 장애를 보조할 수 있는 감각보조기의 개발도 실현 될 것으로 보인다.

The aim of project is developing the bioelectronic interface device arrays for ultra-sensitive biochemical molecular sensor, which combines the mammalian olfactory tissue with high specificity and sensitivity for various biochemical molecules and a solution process transistor capable of ultra low-voltage operation on aqueous solution.
Result
This study was conducted to develop the next-generation cell-based biosensor with extreme sensitivity and selectivity through the development of electronic device for long-term operation in aqueous electrolyte and the investigation of cell-device interface between living cells and electronic devices. In order to interface electronic device with the olfactory tissue, we developed high-performance metal oxide-based device capable of a low-voltage operation in aqueous solution, and then, the electrochemical stability and long-term operational stability of metal-oxide semiconductor were confirmed through the electrochemical experiment. Furthermore, we developed the metal-oxide transistor based pH sensor which shows the changes in the threshold voltage according to the pH of the electrolyte, and its pH selectivity was confirmed by investigating the threshold voltage changes depending on ion concentration, ion species and pH. In addition, through the solution-mediated crystallization of PEDOT:
PSS, we developed a high-performance electrochemical transistor that was operated for a long time in aqueous electrolyte without performance degradation and showed a strong resistance to thermal and electrocal stresses caused by autoclaving and repetitive voltage application.
In terms of the cell-device interface, we confirmed that the metal-oxide material and PEDOT;PSS did not affect the cell viability and growth by evaluating their biological toxicities. In order to establish the generic techniques on real-time cell signal measurement, we developed PEDOT:PSS based microelectrode arrays (MEA), and it was shown that the arrays efficiently read the electrical signals of cardiomyocytes directly cultured on the device surface.
Expected Contribution
In addition to the successful development of various electrolyte-gated transistors and understanding of interfaces between cells and electronics through this research project, further research will realize the development of olfactory tissue-transistor array interfaces. The development of sensory tissue-transistor array interfaces will afford high sensitivity biomolecule detection and bioelectric signal measurement, and will be applied to diagnostic sensors for detecting trace amounts of pathogen, viruses, bacteria, and etc. Moreover, we can also expected the transistor-based sensors to be widely incorporated in various sensors besides biomedical application, including the chemical detection platforms for prohibited drugs such as methamphetamine and the exploration equipment of underground resources.
