연구의 목적 및 내용
본 연구가 추구하는 궁극적인 목표는 식물의 표피세포를 덮고 있는 큐티클 (왁스, 큐틴)의 생성 과정 및 생리학적인 역할을 이해하는 데 있다. 특히 외부 환경스트레스에 대한 식물 큐티클의 역할을 규명함으로써 농작물의 양적 질적 수준을 향상시키며, 큐틴 및 왁스 유래 biofuel의 산업화에 기여하고자 한다. 본 과제의 최종 연구 목표는 큐티클 대사에 관여된 유전자들이 외부 환경스트레스에 어떻게 관여하고 있는지? 이들 유전자의 발현을 조절하고 있는 전사조절유전자는 존재하는지? 존재 한다면 어떤 조절 유전자에
연구의 목적 및 내용
본 연구가 추구하는 궁극적인 목표는 식물의 표피세포를 덮고 있는 큐티클 (왁스, 큐틴)의 생성 과정 및 생리학적인 역할을 이해하는 데 있다. 특히 외부 환경스트레스에 대한 식물 큐티클의 역할을 규명함으로써 농작물의 양적 질적 수준을 향상시키며, 큐틴 및 왁스 유래 biofuel의 산업화에 기여하고자 한다. 본 과제의 최종 연구 목표는 큐티클 대사에 관여된 유전자들이 외부 환경스트레스에 어떻게 관여하고 있는지? 이들 유전자의 발현을 조절하고 있는 전사조절유전자는 존재하는지? 존재 한다면 어떤 조절 유전자에 의해 발현이 조절 되는지? 에 대한 의문점을 해결하는 것이며, 이러한 연구는 궁극적으로 다양한 외부 환경스트레스에 대한 큐티클 역할에 대한 이해와 폭을 넓게 할 수 있을 것이다.
연구결과
다양한 환경스트레스에 대한 애기장대 식물체의 전사체 분석 결과들을 바탕으로 애기장대 큐티클 대사 관련 유전자들의 microarray를 분석한 결과, 암(dark)과 저산소 스트레스 하에서 큐티클 대사 관련 유전자들의 전사체 함량이 감소됨을 확인하였고, 이들 유전자들의 발현 조절에 관여하는 전사조절인자를 발굴하였다. 조금 더 자세히 살펴보면, 암 스트레스 하에서 발현이 유도되는 AP2/ERF type 전사 조절인자 Decreased Wax Biosynthesis(DEWAX) 유전자를 발굴하였고, DEWAX 유전자의 돌연변이체와 과다발현체의 화학적 분석 결과, 잎과 줄기에서 큐티클 왁스의 함량이 각각 증가 및 감소됨을 확인하였고, 담배원형질체와 reporter 시스템을 이용한 transient gene expression 및 Chromatin immunoprecipitation 분석을 수행한 결과, DEWAX는 wax 대사 관련 유전자들의 promoter에 직접적으로 결합하여 그들의 전사를 억제하는 전사조절 억제자임을 밝혔다.
특히 낮밤 주기로 조절되는 DEWAX는 LACS2, ACLA2, ECR 유전자들의 발현을 조절함으로써, 애기장대 식물체는 낮보다 밤에는 상대적으로 적은 왁스 함량이 축적됨으로써 탄소동화 산물의 효율적 사용 가능성을 밝혔다. 더불어 DEWAX는 애기장대의 줄기보다는 잎에 월등히 많은 발현 양을 보이므로 이는 줄기에 비해 10배 이상 적은 wax 함량을 갖는 잎에서도 wax 생합성 전사조절 억제자로써의 기능을 제시하였다. 또한, 저산소 스트레스 하에서 발현이 증가되는 AP2/ERF type 전사 조절인자AtERF106는 효모를 이용한 yeast-one hybrid 실험에서 전사활성 억제자임을 확인하였고, AtERF106 과다발현체의 잎과 줄기에서 큐티클 왁스의 축적이 현저히 감소되었고, 더불어 LACS2, BCCP2, ACLA2 유전자를 포함하여 큐티클 대사 관련 유전자들의 전사체 발현양이 현저히 감소됨을 확인하였다. 반면 aterf106 돌연변이체에서는 왁스 함량의 증가가 확인되었다. AtERF106은 target 유전자들의 프로모터에 직접적으로 결합하여 전사 활성을 억제함을 확인하였고, 저산소 스트레스 하에서 왁스층을 얇게 형성함으로써 외부 산소의 공급을 원활하게 할 가능성을 제시하였다. 그 외 고염 스트레스에 의해서 왁스 함량이 증가하므로 고염에 의해 발현이 증가되는 AP2/ERF type 전사 조절인자 WRI4를 클로닝하였고, 분자적 수준에서의 WRI4의 특성 규명이 완료되었다 그러나, wri4 돌연변이체 분석을 통하여 WRI4가 고염 스트레스에 의해 증가되는 왁스의 함량 증가에는 관여되지 않음을 확인하였다.
연구결과의 활용계획
해당분야 학문발전 및 연구 실적에의 기여 : 식물 세포 외벽의 큐티클층이 식물세포에서 중요한 생리적인 기능을 포함하는 분자적 생합성 기작에 대한 폭넓은 이해가 이루어질 것이며, 그에 따른 연구 논문 발표 및 특허의 출원이 가능할 것이다. 산업 발전에의 기여 등 국가 경제력 증대로의 활용: 큐티클 대사를 조절하는 전사조절인자를 규명함으로써 이를 응용한 환경스트레스 내성 작물의 개발 및 기술이전에 활용될 것이다. 연구수행 과정을 통한 고급 연구 인력 양성 효과: 생화학, 분자생물학, 분자 유전학, 식물분자생물학, 식물생리학 등 관련분야에서 능력 있는 박사가 배출되었다. 이렇게 양성된 고급 인력은 국가 경쟁력 향상에 기여할 것이다.

The purpose of this research are to elucidate expression profiling of genes involved in cuticle (cutin and wax) biosynthesis under environmental stress conditions, to analyze cutin and wax composition and amounts, and finally to identify transcriptional regulatory factors controlling cutin and wax biosynthesis. Specific aims of this research is to understand the roles of cuticle layer (wax, cutin) during in plant responses against various environmental stresses and to develop environmental stress-tolerant transgenic crops.
Result
Base on the Arabidopsis microarray database, the expression patterns of genes involved in cuticle metabolism was extensively analyzed under normal or environmental stress conditions including drought, salt, dark, and hypoxia. In particular, we observed that the transcript levels of genes involved in cuticular wax biosynthesis and transport were reduced by dark and hypoxia treatments, but were elevated by drought and salt stresses. To understand the transcriptional regulatory mechanisms underlying cuticular wax biosynthesis in Arabidopsis under various environmental stress conditions, several candidate genes encoding transcription factors involved in cuticular wax biosynthesis were isolated and functionally characterized in this study. To investigate the role of a dark-inducible AP2/ERF type transcription factor, Decreased Wax Biosynthesis (DEWAX) in planta, we isolated T-DNA-inserted dewax mutant and generated transgenic plants overexpressing DEWAX under the control of CaMV 35S promoter (DEWAX-OX).
Total cuticular wax amounts increased in leaves and stems of dewax mutant, but decreased in the same organs of DEWAX-OX plants. Transient transactivation assay in tobacco protoplasts and chromatin immunoprecipitation analysis showed that DEWAX represses the expression of LACS2, ACLA2 and ECR genes by direct binding to their promoters. A diurnally controlled DEWAX negatively regulates cuticular wax biosynthesis during daily light and dark cycles. We suggest that DEWAX might be involved in the organ-specific cuticular wax biosynthesis based on the significant differences on the transcript levels of DEWAX between stems and leaves. In addition, an AP2/ERF type transcription factor, AtERF106 that was induced by hypoxia was characterized as a transcriptional repressor that controls cuticular wax biosynthesis under hypoxic stress conditions. Overexpression of AtERF106 caused a decrease in total wax loads in Arabidopsis stems and leaves.
The levels of LACS2, BCCP2, and ACLA2 transcripts were significantly reduced in AtERF106 OX plants. By contrast, total wax loads increased in leaves of aterf106.
Furthermore, the expression of LACS2, BCCP2, and KCS12 was down-regulated by directly binding of AtERF106 to their promoters. These results provided that the AtERF106 is a novel transcriptional repressor involved in cuticular wax biosynthesis under hypoxic stress. Finally, we characterized an salt-stress inducible WRI4 encoding an AP2/ERF-type transcription factor, which is a transcriptional activator in cuticular wax biosynthesis. However, we observed that WRI4 was not involved in the up-regulation of cuticular wax biosynthesis under salt stress condition.
