본 연구에서는 생체 인지질과 구조적 유사성을 갖는 AB2형 믹토암 고분자가 수성 환경 하에서 항암제 또는 유전자와 안정한 폴리머좀, 폴리플렉스등과 같은 다양한 형태의 나노베시클을 형성하며, 기존의 블록공중합체 미셀과 지질기반 제형과 같은 고전적인 전달시스템보다 효과적으로 세포 소기관을 표적하여 약물 및 유전자를 전달할 수 있는 시스템을 개발하고자 하였다.
세부적으로는:
1) 다양한 친수성, 소수성 고분자블록들의 선택적 조합(맞춤형 합성)을 통해 생체인지질 모방형 AB2형 믹토암 고분자 및 나노베시클 구조를 설계하고, 환경
본 연구에서는 생체 인지질과 구조적 유사성을 갖는 AB2형 믹토암 고분자가 수성 환경 하에서 항암제 또는 유전자와 안정한 폴리머좀, 폴리플렉스등과 같은 다양한 형태의 나노베시클을 형성하며, 기존의 블록공중합체 미셀과 지질기반 제형과 같은 고전적인 전달시스템보다 효과적으로 세포 소기관을 표적하여 약물 및 유전자를 전달할 수 있는 시스템을 개발하고자 하였다.
세부적으로는:
1) 다양한 친수성, 소수성 고분자블록들의 선택적 조합(맞춤형 합성)을 통해 생체인지질 모방형 AB2형 믹토암 고분자 및 나노베시클 구조를 설계하고, 환경반응성, 생기능성, 표적화능를 부여하였음.
2) 나노구조 분석 및 제어를 통해 구현된 다양한 형태학적 특성의 자기조립 나노베시클들의 전달체적특성, 세포유입, 세포소기관 표적화, 약물방출거동 간의 상관관계를 규명하였음.
3) 다양한 화학약물과 생물약물을 효과적으로 세포소기관으로 전달하고, 약물맞춤형 방출 및 전달특성의 구현을 위한 믹토암 고분자 기반 나노베시클의 구조 및 기능성의 최적화하였음.
-1차년도 연구에서는 맞춤형 합성을 통해 얻어진 다양한 믹토암 고분자의 체계적인 화학적 구조 변환을 통해 고분자의 일차구조와 나노베시클 구조와의 상관관계를 규명하였음. 합성된 구성블록, 믹토암 고분자 및 나노베시클의 물리화학적 특성과 세포독성을 평가하고 세포소기관 표적화 나노베시클의 맞춤형 설계를 위한 기초로 이용함.
-2차년도 연구에서는 단백질, 펩타이드 및 유전자를 위한 생기능성 블록을 포함하는 믹토암고분자와 특정 세포소기관을 표적할 수 있는 블록을 갖는 믹토암 고분자를 제조하여, 이를 이용한 생기능성 및 표적화 나노베시클의 제조 및 특성평가를 수행하였음.
-3차년도 연구에서는 다양한 약물에 대하여 생체반응성 분해와 같은 생기능성을 가지면서, 미토콘드리아, 핵, 세포질, 퍼록시좀 등 다양한 세포 내 소기관을 효과적으로 표적할 수 있는 나노약물베시클을 제조하고 그 성능을 평가하였음.

This study investigated the formation of stable nanovesicles, which have various morphologies (e.g., polymersome, polyplex) and load various therapeutics (e.g., chemicals, genes), from AB2-type miktoarm polymers with structurally mimicking phospholipids and the development of the more effective subcellular targeting delivery systems compared to the conventional delivery systems (e.g., block copolymer-based micelles and lipid-based vesicle).
The study focused on
1) the synthesis of phospholipid-mimic AB2 miktoarm polymers using customized combionations of various polymer blocks and the design of their nanovesicular structures with stimuli-responsiveness, biofunctionality, and targetability
2) the indentification of the correlation between various morphological characteristics of the self-assembled nanovesicles through analysis and control of nanostructures and various delivery traits such as drug delivery, cellular uptake, subcellular targeting, and drug release.
3) the optimization of the miktoarm polymer-based nanovesicular structure and functionality for realizing customized drug delivery and release of various chemical and biological therapeutics at target subcellular organelles.
- In the first year, the study investigated the relationship between the nanovesicular structures and the polymeric architectures through systemical variation of chemical structures of customized miktoarm polymers. Physicochemical characteristics of synthesized block components, miktoarm polymers, and nanovesicles were evaluated and their results were ultilized for customized designs of subcellular targeting nanovesicles.
- In the second year, the study synthesized and characterized the miktoarm polymers having the biofunctional block components either for delivering proteins, peptides, and genes or for targeting subcellular organelles.
