본 연구는 한국 연근해에 존재하는 다양한 수괴의 기원을 추적하고 해양생태계와 수 환경의 연간 및 계절변화를 관찰하기 위하여 미소유전체 분석을 활용하여 수괴 분포 범위를 구명하고자 하였다. 플랑크톤 군집의 장단기적 변화 양상 모니터링을 위해서 대한해협, 마산만, 낙동강 하구역에서 시료를 채집하였고 메타게놈 분석을 실시하였다.

대한해협은 동해의 울릉분지로 진입하는 주요 해류가 지나는 길목이며, 마산만은 반폐쇄성 내만으로 육상기원 오염원에 의한 수환경 오염이 심각한 곳이다. 낙동강 하구역은 한국 연안의 가장 큰 담수 유입원이며 마산만을 지나는 남해 해류와 더불어 대한해협에 영향을 주는 것으로 알려져 있다.
본 연구진은 다년간에 걸쳐 대한해협과 연근해에 대한 해수시료를 확보하고, 이화학적 특성과 생물학적 정보를 획득하여 각 수괴에 대한 DB를 구축하고자 하였다. 또한 수괴 특이적인 생물학적 지표종을 발굴하여 수괴 변화 양상에 따른 생물학적 변화를 정밀하게 모니터링 하고자 하였다.
물리화학적 분석 결과, 2013년 7월의 대한해협의 서수도는 대마난류 표층수의 특징이 나타났으며, 표층수는 연안에서 수심 0~10 m, 외해역에서 수심 0~50 m 구간에서 발견되었다. 대마난류 중층수로 추정되는 수괴는 연안 약 10~50 m, 외해역에서 수심 약 50~140 m에서 나타났다. 저층수는 비교적 용존산소가 풍부했으며, 수심이 깊어질수록 증가하였다. 영양염은 밀도가 낮은 표층에서 매우 적은 농도로 존재하였으며 밀도가 큰 저층으로 갈수록 증가하는 양상이 나타났다. 2014년 11월 대한해협 저층냉수는 존재하지 않는 것으로 분석되었고, 대마난류 표층수는 약한 담수 유입에 의한 저온고염으로 관측되었다. 수심 50m 이상에서 나타나는 고염저용존산소 수괴는 염분으로 볼 때 쿠로시오에서 기원한 해수이나 용존산소가 낮은 것으로 보아서 그 기원이 대기와 접촉하지 않는 유광대 하부의 아표층수로 판단된다. 대한해협 서수도의 고온기에는 동중국해 표층수, 쿠로시오 아표층수, 대한해협저층냉수의 세종류 수괴가 존재하고, 겨울에는 동중국해 표층수와 심층 기원의 쿠로시오 아표층수만 존재하는 것으로 판단된다.
메타게놈 분석 결과, 대한해협의 U07정점에서 총 374개의 clone에 대한 119개의 phylotype을 발굴하였고, 와편모조강과 Syndiniales가 수심에 따라 다른 비율로 나타나는 경향성을 확인하였다. KS1108 정점에서는 총 548개의 clone을 분석하여 338개의 phylotype을 발굴하였고, 표층에서는 확인되는 절지동물문이 저층에서는 관찰되지 않으며 와편모조강과 자포동물문이 높은 비율로 나타나는 특징을 보였다. 대한해협은 수심에 따른 와편모조강 내의 종다양성이 증가하는 특징을 보였다. KS110816는 표층에 야광충목이 우점하여 나타났으며 엽록소 최대층에서는 Gymnodiniales, 저층에서는 Syndiniales가 우점하였다. 쿠로시오 표층에서 기원 된 수괴는 아표층수 기원의 수괴와는 다르게 섬모충문, 절지동물문, 사족충류, Choanoflagellida, Telonemida 속의 생물이, 쿠로시오 아표층수 기원의 수괴에서는 방사극충강, 다냥류, Sticholonchida가 특이적으로 나타났다.
마산만의 생물종 분석 결과, 표층에서는 퇴적물 내에 혹은 표면에 부착하여 서식하는 환형동물문이 저층에 비해 높은 비율로 나타났다. 와편모조강은 대한해협과 달리 표층에 더 많이 출현하였다. 마산만 저층에서는 주로 표층에서 발견되는 야광충목과 플랑크톤 섭식활동을 하는 절지동물문도 높은 비율로 나타났다. 프로테오박테리아는 표층과 저층 모두에서 높은 비율로 나타났으며, 남조류, 규조류, 녹조식물문은 저층에서 발견되지 않았다.
낙동강 하구역에서는 담수, 기수, 해수의 특징을 가지는 시료를 대상으로 총 456개의 clone을 통해 122 phylotype을 발굴하였고, 그 중에서 전 세계적으로 보고된 바 없는 7개의 phylotype을 발굴하였다. 해수에서는 와편모조강이 가장 많이 나타났으며, 기수에서는 와편모조강 및 광염성 규조류와 해양성 생물들이 확인 되었다. 담수에서는 와편모조강이 나타나지 않았으며, 담수의 유입에 의한 영향을 잘 반영하는 시누라조류, 황녹조류, Moina 속과 같은 생물종 조성이 확인되었다.
각 생물종 정보는 비슷한 환경에서의 유사성이 높게 나타났으나, 물리화학적 특성으로는 구분되지 않는 수괴에 대한 생물학적 특성이 나타나기도 하였다. 또한 기존에 보고되고 있는 서식지 정보를 바탕으로 해양환경 변화에 잠재적으로 지표종의 역할을 할 수 있는 생물에 대한 정보를 확인하였다.
본 연구를 통해 축적된 DB는 수괴의 특성과 생물군 군집을 확인하고 그 연관성을 파악하는 하는 데 중요한 기초자료가 될 것으로 판단된다. 특히 잠재적인 지표종에 대한 정보는 정밀한 수환경 변화 및 해양생태계 변화 양상을 확인할 수 있을 것으로 기대된다. 또한 한국 연근해의 생물종 다양성을 확인할 수 있는 기본 레퍼런스 자료를 제공하며, 전 세계에 보고된 바 없는 생물종 추적 및 발굴에 중요한 역할을 할 수 있을 것으로 기대된다.

This study traced the origins of the various water masses that exist in Korean coastal water and investigated the distribution range using genomic analysis to observe the seasonal and annual changes of environmental and marine ecosystems. For the short- and long-term monitoring of the changes of plankton populations, the samples collected in the Nakdong River Estuary, the Masan bay and the Korea Strait and the metagenomic analyses were conducted.
The Korea Strait is an entrance for a major ocean current passing to the Ulleung Basin of the East Sea. And the Masan bay is a place contaminated by a number of serious terrestrial sources. The Nakdong River Estuary is the largest freshwater inflow sources of the Koea Strait and has been known to affect it.
We obtained water samples from the Korea Strait and coastal waters for about over the years, and to acquire the physicochemical properties and biological information to build a database for each water mass. Using the water mass specific biological indicator species, we precisely monitored the biological changes in the water mass.
From the results of physical and chemical analysis, the Western Channel of the Korea Strait on July 2013 showed the characteristics of Tsushima surface water. The surface water was found at depths 0 ~ 50 m in the offshore and in 0 ~ 10 m depth of coastal region. The water mass was estimated to be the Tsushima intermediate water appeared about 10 ~ 50 m depth and 50 ~ 140 m depth. Bottom water was relatively oxygen-rich following the depth. Nutrients were present at very low concentrations in the low-density surface layer showed a tendency to increase with the depth.
The Korea Straits bottom cold water did not exist on November 2014 and the surface water of Tsushima current was observed at low temperatures with high salt due to weak fresh water inflow. The water masses with high salt and low oxygen appears more than 50 m in depth was originated from the Kuroshio current.
From the meta-genomic analysis, 119 phylotypes (a total of 374 clones) were isolated from U07 in the Korea Strait. Dinoflagellates and Syndiniales showed the different ratios depending on water depth. The position of KS1108 revealed 338 phylotypes (a total of 548 clones). Dinoflagellates in the Korea Strait increased the diversity of species in the depths. Noctiluca species were dominant in KS110816. In the chlorophyll maximum layer, Gymnodiniales showed the high biodiversity. Species of ciliates, arthropoda, cercozoa, Choanoflagellida, and Telonemida were presented in the water masses originated from Kuroshio current.
The species community analysis for the samples of Masan bay, Annelida were dominantly found from the surface. In low depth of Masan bay, arthropod and Noctiluca were found at a high rate. Proteobacteria was shown at a higher rate in both the surface and the blue-green algae, diatoms, green algae were not found in the bottom. In the Nakdong River Estuary freshwater, a total of 456 clones(122 phylotypes) with the characteristics of seawater, freshwater and brackis hwater. Specially, seven phylotypes have not been reported from the world wide.
The biological characteristics of the water masses were more sensitive to the physical and chemical properties. Moreover, the biological information can serve as a potential indicator species in the marine environment change based on the habitat information being reported. The database accumulated in this study is expected to be an important basis to determine the characteristics of the water masses and biota communities and identify their relevance. In particular, the information on potential indicator species are expected to be able to determine the precise number of changes of climate change and marine ecosystems.
