- 광생물반응기(Photo bioreactor; PBR)를 이용하여 회식분 배양과 연속식 배양을 통해 유기성 폐수에서 성장이 우수한 미세조류를 선별하였으며, 미세조류 바이오매스의 생산을 목적으로 최대 생산성을 위한 배양 인자를 도출하였음. 또한 옥외에서 mini raceway pond와 pilot scale raceway pond를 이용하여 유기성 폐수의 처리와 미세조류 바이오매스 생산이 우수한 옥외배양 조건을 검토하였고, 부착 매질을 이용한 부착 미세조류 배양 시스템을 검토하였음.

- 미세조류의 군집 분석과 PSB(Phyco-sphere bacteria)를 분석하기 위한 목적으로 분자유전학적 기법을 이용하여 미세조류와 박테리아의 모니터링 시스템을 구축하였으며, 미세조류와 박테리아의 상호작용 연구를 위해 axenic 미세조류를 유도하였고 관련 박테리아를 분리하였음. Axenic과 분리한 박테리아의 혼합배양을 통해 미세조류와 박테리아의 성장이 함께 촉진되는 상리공생을 확인하였고, PGPR인 Rhizobium 속 박테리아의, cell free extract에서 미세조류 성장 촉진능이 높은 것으로 확인되었음.
- 친환경적 미세조류 수확방법 확립을 위해 박테리아로부터 생산되는 미생물응집제의 생산조건과 응집조건을 최적화하였다. 또한 미세조류 박테리아의 상호작용에 의한 자가응집 효과를 확인하였고, 미세조류 응집에 관여하는 박테리아를 분석, 분리 및 동정하였음.

Photobioreactor(PBR) was used for the selection of microalgae highly growing in wastewater and the investigation of growth factors. Chlorella vulgaris effectively utilized organic wastewater as their growth supporting nutrients in batch and continuous cullture, respectively. The factors highly affecting the microalgal growth were agitaion, LD cycle, CO2 concentration, temperature and photosynthetic active radiation(PAR). Optimal conditions of microalgal biomass production were 100 rpm, continuous light, 3% CO2, 25℃, 13:1 NP ratio and 500 μmol/m2/s·PAR. Chemostat culture of C. vulgaris showed the highest volumetric biomass productivity of 1.013/g/L/d in optimal condition and dilution rate of 0.75 d-1. CO2 and PAR was key growth factor among the analyzed cultivation factors.
The microalgae was cultured in outdoor mini raceway pond for municipal wastewater treatment and biomass production. Fed batch raceway pond has depths and water capacity of approximately 30 cm and 60 L, respectively. The raceway was introduced CO2 by mixed of a paddle wheel to give a mean horizontal water velocity of approximately 0.3m/s. Cultivating microalgae for municipal wastewater treatment to generate low nitrogen (2.36 ppm), low NH3-N (0.58 ppm), low phosphorus (0.88 ppm), and low COD (27.72 ppm) effluents could also reduce nutrient costs for microalgal cultivation. Also, the nitrogen, NH3-N, phosphorus, and COD showed very high removal efficiencies (94.69%, 96.74%, 80.44% and 74.27% for nitrogen, NH3-N, phosphorus, and COD, respectively). The outdoor mini raceway pond culture showed good settleable property and could totally settle within 3 min. The mini raceway pond culture showed relatively high concentration of microalgal biomass(1.26 g/L) and lipid content of microalgae was significantly maintained from 21.97% to 18.88%. From these results, it was believed that the outdoor mini raceway pond system could be used for removing the nitrogen, phosphorus, and COD from municipal wastewater effectively as well as for the production of microalgal biomass.
Comparison of diverse culture conditions revealed that phosphate addition of 0.3 ppm was superior to all other treatments such as longer retention time, CO2 gas supply, and bicarbonate supply. Phosphate addition led to the highest biomass and lipid productivity as well as top removal rate for nitrogen and phosphorus. Adjusting optimal N:P ratio was most influential in determining lipid productivity.
Periphytic microalgae were cultivated using mesh-type substrate, to get higher biomass even in cold seasons. Biomass productivity of periphytic microalgae was about 7 times higher than that of suspended microalgae, even under 10℃. Removal rates for nitrogen and phosphorus were also enhanced by periphytic microalgae. In addition, it was much easier to harvest periphytic microalgae, compared with suspended microalgae.
High efficiency DNA extraction method and new primer were developed for monitoring of microalgae and the associated bacteria. DGGE and pyrosequencing were performed for analysis of phyco sphere bacteria(PSB). Pyrosequencing analysis showed that the xenic culture broth included Flavobacterium sp.(37%), Hyphomonas sp.(21%), Sphingomonas sp.(20%) and Rhizobium sp.(16%) as phycosphere bacteria.
The axenic cultures of C. vugaris was developed from ogamic waste water by a combination of ultrasonication, FACS and micropicking. We performed co-cultivation of axenic C. vulgaris with each isolated bacteria. All isolated bacterial strains enhanced the growth rate of microalgal cells(41~115%). These results show that mutualism between C. vugaris and its associated bacteria under phototrophic conditions.
To harvest of microalgae with bio-friendly and economic methods by using microbial flocculants, Bacillus sp. A8 which producing extracellular polysaccharide was cultivated. And then this bioflocculant was inoculated in the C. vulgaris culture by chainging pH and flocculant concentration. In pH 5, flocculating activity with cationic ions and low concentrations of bioflocculant A8 (1 ppm) was more than 30% compared with only cationic ions were innoculated. Besides, xenic culture which contains microalgae-associated bacteria showed much higher flocculating activity of 94% than almost 0% of axenic culture in pH 11. This implies bacteria can affect the flocculation, from the basis of these results, flocculating activity of C. vulgaris culture was investigated with microalgae-associated bacteria to obtain more enhanced flocculating activity. Two kinds of xenic culture broth, bacterial cell-existing broth and bacterial cell-free broth were used. From this result, both bacterial cell and bacterial secreted metabolites can be effective for the flocculation of C. vulgaris. To identify the bacterial community involved in xenic culture, DGGE was performed and Flavobacterium sp., Terrimonas sp., Sphingobacterium sp., Rhizobium sp., Hyphomonas sp. was identified as C. vulgaris associated bacteria. Among them, Flavobacterium sp., Terrimonas sp., Sphingobacterium sp. may play a key role in the flocculation.
Funnel shaking method used chloroform was high efficiency extraction method among various lipid extraction method. Chloroform/Methanol(2:1) was best extraction solvent through TLC analysis for TG, DG, MG and FFA. Analysis of spots on TLC plate were performed through Gene tool image analysis system. Conversion of FAME form total lipid is an important step in the overall process of biodiesel production. Quantitative analysis of converted FAME is also important. For accurate quantitative analysis, we derived standard curve about 14 fatty acid standard.
Conventional lipid extraction methods require completely dried algae biomass and this precondition worsens low economic feasibility of biodiesel, because considerable expenses are consumed in drying, lipid extraction, and transesterification. A variety of methods were compared to solve this problem. By using diverse catalysts and optimizing conditions, higher amounts of FAME were obtained just by one-step process, directly from wet biomass.
