1) 국내외 독성발현경로 및 오믹스 기술 연구 동향 조사

• AOP는 위해성 평가 등의 규제 결정과 관련이 있는 직접적인 분자 수준의 초기 현상(Direct Molecular Initiating Event)과 각 생물학적 단계에서의 악영향(Adverse Effect at a Biological Level of Organization) 간의 연관 경로와 관련이 있는 기존의 지식을 전달(Portray)하는 개념적인 구조(Conceptual Construct) 임
• 생태위해성 평가 활용을 위한 AOP는 급성/만성 어류 독성, 어류 생식 독성 등이 연구 중
• 인체위해성 평가 활용을 위한 AOP는 피부 자극성, 미토콘드리아 독성 등이 연구 중
• 미토콘드리아 독성의 경우 전체 개체에 대한 AOP가 아닌 세포수준에서의 AOP임

2) 탄소나노튜브 관련 유전체, 단백체 및 생물정보학을 이용한 AOP 분석
① 시험관 내(in vitro) 모델에서의 독성 연구
■ 기능기와 크기가 다른 MWCNTs 독성 비교: 기능기(Pristine- MWCNTs, OH-MWCNTs, COOH-MWCNTs), 크기 (OD 크기, 튜브 길이), 세포주(폐세포(Beas-2B), 간세포(HepG2))에 따른 독성 비교
• 세포주에 따른 독성 민감도를 관찰하였을 때 폐세포(Beas-2B)가 간세포(HepG2) 에 비하여 MWCNTs 노출에 민감하게 반응하는 것을 확인
• 기능기에 의한 독성의 차이는 확연하게 나타나지 않았으나 같은 기능기를 가졌을 경우(OH-MWCNTs) 크기에 따른 독성영향으로 크기가 작을수록 독성이 증가하는 것을 확인
• 이러한 현상은 작은 크기의 MWCNTs가 세포 내로의 유입(Uptake)이 증가하는 것과 상관관계가 있으며, 세포 내로 유입이 증가할수록 독성영향도 증가하는 것으로 생각됨
• 세포독성, 산화적 스트레스, DNA 손상 등의 종말점(Endpoint)을 조사한 결과 대체적으로 Pristine-MWCNTs의 독성이 강함
• 소포체(ER) 스트레스는 MWCNTs 노출에 의하여 발생하는 것으로 관찰되었으며, OH-SS-MWCNTs의 영향이 큰 것으로 나타남
• 후성유전학(Epigenetic) 영향을 확인한 결과 간세포(HepG2)에서는 기능기에 따른 DNA 메틸화(Methylation)의 차이를 확인할 수 없었으나, 폐세포(Beas-2B) 의 경우 MWCNTs 에 의하여 저메틸화 (Hypomethylation)가 진행되었으며, Pristine-MWCNTs에 비하여 기능기를 가진 MWCNTs에서 저메틸화가 유도됨
■ 기능기가 다른 MWCNTs 독성 비교: 기능기 (Pristine-MWCNTs, NH₂-MWCNTs, O+-MWNTs, COOH-MWNTs) 에 따른 독성 비교
• 기능기에 따른 세포 내 유입의 차이를 관찰한 결과 COOH-MWCNTs가 세포 내에서 많이 관찰됨
• 세포독성, 산화적 스트레스, DNA 손상, ER 스트레스, 세포사 (Apoptosis) 등의 종말점을 조사한 결과 COOH-기능기를 가진 MWCNTs가 가장 독성이 강한 것으로 관찰
• 후성유전학 영향을 확인한 결과 MWCNTs에 의하여 저메틸화가 진행되어 유전자 발현의 증가를 유도하는 것으로 나타남
• COOH-MWCNTs가 가장 독성 영향이 강한 것은 쉽게 유입되어 세포 내에서 세포의 정상적 기능과 관련된 메커니즘에 영향을 주었기 때문으로 사료됨
• 독성메커니즘을 연구하기 위하여 COOH-MWCNTs 노출 후 PCR 어레이를 실행한 결과, 주요 독성메커니즘은 세포사/괴사(Apoptosis/Necrosis), 저산소증(Hypoxia), ER 스트레스, DNA 손상인 것을 확인
② 생체 내(in vivo) 마우스 모델에서의 독성 연구
• 기능기가 다른 MWCNTs에 노출시킨 마우스의 혈액에서 산화적 스트레스 마커(Marker)로 알려진 GSH/GSSG 비율을 측정한 결과, COOH-MWCNTs 노출에 의하여 GSH/GSSG 비율이 유의하게 감소
• MWCNTs 가 마우스 혈액에서 DNA 손상을 유도하며, COOH- MWCNTs가 가장 크게 영향을 미침
③ 생체 내(in vivo) C. elegans 모델에서의 독성 연구
■ 단백체학의 결과와 이를 이용한 경로 분석에서 내세포작용과 산화적 스트레스, ER 스트레스가 C. elegans에서의 MWCNTs에 의한 유입과 주요 독성메커니즘으로 나타남
■ C. elegans 돌연변이를 이용한 기능유전체학 결과는 MWCNTs에 의한 독성메커니즘인 산화적 스트레스와 ER 스트레스의 관련성을 확인함
독성발현경로(AOP) 연구는 OECD 등 국제기구에서 독성유전체 등 분자 수준의 메커니즘 데이터의 위해성 평가에의 활용을 위해 활발한 연구가 진행되고 있음. 국내의 독성 유전체 연구도 AOP의 개념과 결합되어 화학물질 관리에 활용하려는 적극적인 노력이 필요하며, 이를 위해서 국내에 AOP 개념의 도입과 적극적인 확산이 필요함.
탄소나노튜브 관련 유전체, 단백체 및 생물정보학을 이용한 독성발현경로를 인 체세포, 마우스, 꼬마선충 모델에서 연구한 결과 탄소나노튜브의 기능기가 독성 에 영향을 미치며 산화적 스트레스, 소포체 스트레스 등이 독성 작용 기전인 것 으로 드러남. 보다 정교한 독성 작용 기전은 추가적인 연구를 통해 밝혀질 수 있을 것임.

In this study, we first reviewed the concept and development of Adverse Outcome Pathway(AOP) in regulatory context, secondly investigated toxicity of muti-wall carbon nanotube(MWCNT) in various irt vitro and in vivo systems, and finally we tried to apply AOP concept to nano toxicity evaluation. AOP methodology is one approach to provide a framework for information from toxicogenomics, bioinformatics, systems biology and computational toxicology to be collected and rationalized. An AOP is a representation of existing knowledge concerning the linkage(s) between a molecular initiating events(MIE) and an adverse outcome at the individual or population level. To implement a predictive strategy for risk assessment, results from in vitro toxicity assays focused on cellular responses to molecular initiating events will need to be extrapolated to effects on organisms and ultimately to populations. This can be achieved by developing the AOP which links an MIE with adverse effects. As they are intended to be used by the regulatory agencies, it is important to standardize the way in which AOPs will be developed and documented. For developing an AOP, a consistency across several levels of biological organization, including anchoring to the apical effects, is likely to be required. The concept of AOP should be implemented to toxicogenomics research for chemical toxicity/safety prediction.
Due to attractive structural, mechanical, electrical and optical properties of carbon nanotubes(CNTs), CNTs have found application as sensors and electronic devices, as well as in catalysis, wastewater treatment and drug delivery. However, the commercial application of CNTs is often limited by their low solubility and dispersibility in both organic and inorganic solutions. To overcome this difficulty, the material surface is functionalized by introducing hydrophilic chemical groups, which lead to higher dispersibility while maintaining the unique properties of pristine CNTs. Functionalized CNTs are therefore widely used as additives, catalysts, sensors, absorbents, intracellular carriers, electrodes, and imaging agents. The prevalence of CNTs in industrial and biomedical settings may lead to unintentional release into the environment, requiring a thorough hazard and risk assessment. Thus, the potential hazard of CNTs in human and environmental health has recently been investigated in numerous studies. Most of these studies have focused on respiratory exposure pathways or in vitro assays with pulmonary cells, due to the similarity of the structure of CNTs to that of asbestos. CNTs toxicity studies emphasize oxidative stress, inflammation, and fibrosis as potential mechanisms of toxicity. However, although functionalized CNTs are being applied to various fields, close investigation of the toxicity of CNTs depending on their functionalization remains limited. In this study, the toxicity of functionalized MWCNTs in human bronchial epithelial cell(Beas2-B), hepatoma cell(HepG-2), the nematode, Caenorhabditis elegans and ICR-mice was investigated using a comprehensive systems toxicology approach. This systems-based integrated approach is particularly valuable for investigating the toxicity of new chemicals, whose mode of action is not fully understood or incompletely characterized, such as with nanomaterials, since it provides a comprehensive insight into the mechanism of toxicity. In the present study we combined this approach with functional genomics tools based on C elegans mutants, which enhance and complement the predictive power of toxicogenomic-proteomics and pathway analysis. Thus, balancing the information of molecular assays and bioinformatics analysis with meaningful toxic outcomes, we were able to link genes to their functional outcomes. Overall results from different test systems suggest that surface functionalization affects toxicity of MWCNTs and oxidative stress and endoplasmic reticulum stress are being involved in MWCNTs toxicity. The integrated systems toxicology approach applied in this study provided a comprehensive insight into the toxicity of chemicals such as MWCNTs. Based on it, further mechanistic study is needed to complete our understanding of the toxicity of functionalized MWCNTs.
