실리콘 기반 나노 신경소자에서 세포 및 조직을 배양하고, 세포 내외부의 생물질 상호작용을 전기생리학적 혹은 분자생물학으로 분석하여 신경 조직분석용 신경소자칩 개발 연구의 목표 하에 1, 2 단계의 연구 수행내용은 다음과 같음.
➢ 1단계 연구 수행내용
1) 나노와이어 신경소자 상에서 배양방법 확립 연구
2) 초기 나노선 소자 상에 신경세포를 직접 배양하여 전기신호분석

추후 뇌 질환치료, 신경 회로망, 뇌유전체 기능 연구 등에 활용될 예정임.

The results of the study of "Development of Nanobio Neural Network Processor" are briefly summarized.
We studied and reported on the compatibility of various nanowires with hippocampal neurons and the structural study of the neuron–nanowire interface. Si, Ge, SiGe, and GaN nanowires are compatible with hippocampal neurons due to their native oxide, but ZnO nanowires are toxic to neuron due to a release of Zn ion. The interfaces of fixed Si nanowire and hippocampal neuron, cross-sectional samples, were prepared by focused ion beam and observed by transmission electron microscopy. The results showed that the processes of neuron were adhered well on the nanowire without cleft.
In this research project, we also present a novel semiconductor nanowire field-effect transistor-based sensing device with an intrinsic amplification mechanism. In this novel device, a nanowire field-effect transistor is integrated with an orthogonal sensing nanowire that amplifies any detected signal. The device operating and intrinsic amplification mechanism has been proposed and experimentally validated. Fabricated exclusively using the top–down processes, the novel device prototypes have demonstrated an appreciable improvement in photodetection against the cofabricated generic nanowire field-effect transistor sensors.
Regarding basic studies about biomolecules and cellular interaction, hypoxia-inducible factor (HIF)-1 has been established as a master regulator of vascular responses to hypoxia and ischemia by driving transcriptional activation of angiogenic factors. Oxygen- and 2-oxoglutarate (2-OG)-dependent, iron(II) containing HIF-specific prolyl-4-hydroxylases (PHDs) and factor inhibiting HIF-1α (FIH-1) catalyze the hydroxylation of the specific proline and asparagine residues of HIF-1α, thereby controlling the level of HIF-1α and ultimately the HIF response. Here, we unveil a new action of hinokitiol, an iron chelator found in natural plants, on stabilization of HIF-1α in cell cultures in a dosedependent manner. In vitro PHD2 and FIH activity assays based on fluorescence polarization reveal that such HIF-1α stabilization is likely medicated by inhibitory effects of hinokitiol on prolyl and asparaginyl hydroxylation of HIF-1α. In addition, the inhibition of PHD2 by hinokitiol is reversed by the addition of 2-OG and iron(II), suggesting that the underlying inhibitory mechanism involves displacement of 2-OG and a chelate formation with iron(II) at the enzyme active site by hinokitiol. Furthermore, hinokitiol treated cells show increased transcription of vascular endothelial growth factor, providing the therapeutic potential in the treatment of ischemic diseases.
We make a vertical nanoprobe with a thickness of about < 100 nm in top using a silicon nanowire and measured intracellular signal of GH3 cell, which has spontaneous action potential. Proper density of nanowires was founded by a culturing hippocampal neuron on a nanowires-grown substrate with various nanowire densities. The vertical nanoprobe was formed by the sequential deposition of passivation layer and electrode layer. Partial etching makes a top-exposed nanoelectrode, making an effective structure to record an intracellular signal. Using an as-prepared nanoprobe, we measured a voltage change along a time, which is similar with a patchclamp data of one. These outcomes show a feasibility to monitor and stimulate an intracellular signal for a long-term in a large area.
Serotonin (5-HT) receptors of type 6 (5-HT6R) play important roles in mood, psychosis, and eating disorder. Recently, a growing number of studies support the use of 5-HT6R-targeting compounds as promising drug candidates for treating cognitive dysfunction associated with Alzheimer‘s disease. However, the mechanistic linkage between 5-HT6R and such functions remains poorly understood. By using yeast two-hybrid, GST pull-down, and co-immunoprecipitation assays, here we show that human 5-HT6R interacts with the light chain 1 (LC1) subunit of MAP1B protein (MAP1B-LC1), a classical microtubule-associated protein highly expressed in brain. Functionally, we have found that expression of MAP1B-LC1 regulates serotonin signaling in a receptor subtype-specific manner, specifically controlling the activities of 5-HT6R, but not those of 5-HT4R and 5-HT7R. In addition, we have demonstrated that MAP1B-LC1 increases the surface expression of 5-HT6R and decreases its endocytosis, suggesting that MAP1B-LC1 is involved in the desensitization and trafficking of 5-HT6R via a direct interaction. Together, we suggest that signal transduction pathways downstream of 5-HT6R are regulated by MAP1B, which might play a role in 5-HT6R-mediated signaling in the brain.
