연구의 목적 및 내용
골격형성에 중요한 역할을 하는 비타민D는 최근 다양한 조직에서 비타민D 관련대사의 주요 효소와 특이수용체가 발견되면서 골격형성 외 다른 역할과 기전 규명이 요구되고 있음.
비만과 대사질환이 있는 사람들에게서 미토콘드리아의 수와 기능이 저하되고 있음이 보고됨.
본 연구는 비타민D가 미토콘드리아 기능 향상을 통하여 지방조직의 무게를 감소시키고 근육 내 에너지 소비를 증가시켜 비만억제를 야기하는지 분석하고자 함.

연구결과
활성형 비타민D, 1,25(OH)2D는 분화지방세포 내 지방축적을 억제하고 지방분해를 증가시킴. 지방세포에서 비타민D 투여로 지방축적, 지방산 산화, 열발생 관련 인자의 유전자 발현을 변화됨. 근육세포에서 지방산과 활성형 비타민D, 1,25(OH)2D를 함께 투여하면 지방산으로 증가된 근육세포 내 지방축적이 감소하고, 지방산으로 인하여 감소된 미토콘드리아의 양과 숫자가 증가한 것을 확인함. 근육세포의 미토콘드리아 생합성, 미토콘드리아 기능 관련 유전자가 활성형 비타민D인 1,25(OH)2D 투여로 변화됨. 활성형 비타민D,1,25(OH)2D를 처리한 지방과 근육세포 모두 에너지 항상성, 미토콘드리아의 생합성과 기능에 관여하는 SIRT1 유전자 발현과 활성이 증가하는 것이 관찰됨. 뿐만 아니라, 필수 보조효소인 NAD/NADH 비율이 활성형 비타민D을 투여한 지방과 근육세포에서 모두 증가하였고, 또 다른 중요 에너지 항상성, 미토콘드리아의 생합성과 기능 관여인자인 AMPK는 활성형 비타민D으로 근육세포에서 증가됨.
고지방식이로 비만이 유도된 쥐에서의 비타민D 결핍은 체충과 지방조직 무게 증가와 근육조직에서의 지방 축적을 야기함. 지방조직에서 비타민D 결핍은 지방세포의 크기 뿐 아니라 염증 증가, 지방축적, 지방산 산화, 염증, 열발생 관련 인자의 유전자 발현 변화를 유발됨.
비타민D의 결핍한 군의 근육조직은 지방축적 증가와 함께 미토콘드리아의 양과 숫자의 감소를 보였고, 미토콘드리아 생합성과 기능, 열발생 관련 유전자 발현 감소를 보였음.
에너지 항상성과 미토콘드리아의 생합성과 기능에 관여하는 SIRT1과 AMPK 활성은 지방과 근육조직에서 모두 비타민D 결핍시 감소됨. 따라서, 본 연구결과를 통하여 비타민D가 근육의 미토콘드리아의 생합성과 지방과 근육조직의 SIRT1과 AMPK 활성을 통한 미토콘드리아 기능에 관여함으로써 비만에 긍정적인 효과를 지님을 규명함.
연구결과의 활용계획
비타민D의 항비만효과 기전을 지방과 근육조직에서 규명한 본 연구결과로, 1) 비타민D가 미토콘드리아 대사와 기능 조절에 미치는 효과를 규명하여 생물학적/영양학적 지식 수준을 높일 수 있음. 2) 항비만연구 및 대사질환 억제를 위한 건강기능성 식품 및 의약품 개발의 새로운 기전과 활성마커를 제시가능함.

Vitamin D classical function is to induce bone formation. Recently enzymes and receptor for vitamin D have been found in many tissues, which function needs to be investigated. Mitochondria number and function were decreased in obesity and type 2 diabetic patients. The purpose this study is to investigate the favorable effects of vitamin D on obesity by regulating fat accumulation in adipose tissue and fat oxidation in muscle tissue via the improvement of mitochondria biogenesis and function.
Result
Active vitamin D metabolite, 1,25(OH)2D inhibited lipid accumulation and basal and isoproterenol-stimulated lipolysis in diffferentiated adipocytes. Gene levels involved in adipogenesis, fatty acid oxidation, and thermogenesis were modulated by 1,25(OH)2D. In addition, 1,25(OH)2D-treated C2C12 differentiated mytubes showed a significant decrease in FFA-increased TG accumulation. 1,25(OH)2D significantly increased FFA-decreased mitochondrial DNA content (mtDNA) and mitochondria size as observed by electron microscopic analysis, Gene expression related to mitochondria biogenesis and function were upregulated by 1,25(OH)2D in C2C12 muscle cells. In both 1,25(OH)2D-treated adipocytes and muscle cells, significant increments in SIRT1 gene expression and activity were observed. A ratio of NAD to NADH in adipocytes and muscle cells and AMPK activity in muscle cells were increased by 1,25(OH)2D treatment.
Significant increments in body weight, adipose tissue weight, and muscle TG levels were noted in obese rats fed with a high fat diet containing insufficient vitamin D levels. Vitamin D inadequacy increased adipocyte cell size and adipose tissue inflammation with modification in gene expression involved in adipogenesis, fatty acid oxidation, thermogenesis, and inflammation. In skeletal muscle, vitamin D insufficiency significantly decreased mitochondria size and mitochondrial DNA content (mtDNA) as well as gene expression related to mitochondrial biogenesis and function. Moreover, adipose and muscle SIRT1 and AMPK activities were significantly decreased in vitamin D insuffient obese rats.
Taken together, these results suggested at least partial involvement of muscle mitochondria biogenesis and adipose and muscle SIRT1 and AMPK activation in the vitamin D–mediated beneficial effects on obesity.
Expected Contribution
Based on results to demonstrate favorable effects of vitamin D on obesity via mitchondria biogenesis and function in adipose and muscle tissue, 1) it is possible to increase bilogical and nutritional knowledge. 2) These studies suggest new mechanisms and markers to develop new functional foods and therapeutic drugs for obesity and obesity-associated metabolic disease.
