To date, solid candidates for the ambient temperature sensor in plants have not been found among conventional genes. I have hypothesized that “small RNAs function as the ambient temperature sensor(s) in plants”. To test this hypothesis and to determine the signaling pathway mediated by the sensor, I will perform following experiments:
1. Ambient temperature-responsive small RNA profiling
2. Identification of putative target genes of small RNAs
3. Expression analysis of putative target genes of small RNAs
4. Isolation of thermomediator genes
5. Regulation of response to small RNA
6. Analysis of diversity of the ambient temperature signaling pathway Developement

○ The identity of a protein(s) that degrades SVP and FLM, thermal mediators in the ambient temperature pathway, has been revealed and its regulation mechanism has been elucidated (published in Science)
○ Structural determinants of miR156 and miR172, ambient temperature-responsive miRNAs, have been determined and the role of the base-stability of the lower stem of pri-miR156a on the miR156 processing has been investigated via NMR (published in Nucleic Acids Research, Journal of Experimental Botany)
○ Genetic pathway that involves the ambient temperature-responsive miRNAs and their role in the regulation of floweirng time has been elucidated (published in Nucleic Acids Research)

Expected Contribution
○ Scientifically, the identification of the temperature sensor in plants would be a major breakthrough in modern biology and open a new avenue for studies of biological protection against global warming and perturbations in ecosystems.
○ The expected results would be used to reduce greenhouse gas (GHG) emissions in Korea, which is expected to face GHG emission cuts by 2013, when the first stage of the 1997 Kyoto Protocol expires.
