연구의 목적 및 내용
진핵세포에서, 세포 소기관의 위치 유지 및 이동에 중요하게 작용하는 것이 미세소관이다. 미세소관의 다이나믹한 구조 변화가 세포의 형태를 유지하는데 중요한 역할을 한다.

미세소관의 역동성에는 미세소관에 결합하는 각종 단백질의 역할이 중요한데, 이 중 미세소관 절단효소는 세포 분열, 형태발생, 섬모의 발생 뿐 아니라 물질 이동에도 상당히 영향을 주는 것으로 밝혀졌다. 하지만 아직까지 정확한 미세소관 절단 기작은 밝혀진 바가 전혀 없다. 본 연구에서는, 미세소관 절단효소 중 카타닌의 활성 기작을 밝히기 위해 구조생물학적, 생화학적 연구를 수행하였다.
연구결과
선행연구결과와 Sequence alignment, Secondary structure prediction의 결과들을 종합하여, p60과 p80 복합체를 정제하기 위해서 p60 N-말단의 MIT 도메인 부분과 p80 C-말단의 Con80 도메인 부분을 클로닝하여 한 대장균에서 두 단백질을 동시발현(co-expression) 시키는 시스템을 이용하였다.
카타닌 p60과 p80을 동시발현 시킨 대장균을 키운 후, Ni-친화성 크로마토그래피, TEV 프로테아제를 이용한 His-tag 제거, 사이즈-배제 크로마토그래피를 거쳐 카타닌 p60-p80 단백질 복합체를 >95%의 고순도로 대량 확보하는데 성공하였다.
로봇을 이용한 단백질 결정 스크리닝을 통해 초기 결정 조건을 찾았고, 단백질 농도변화, salt 농도변화, 부가적인 스크리닝 용액 첨가로 고해상도 X-선 회절 데이터 수집이 가능한 결정화 조건을 최적화 할 수 있었다. 이와 같은 방법으로 셀레노메티오닌-치환 단백질 결정도 확보하였다.
가속기를 이용하여 카타닌 p60(녹색)-p80(파란색) 결합부위 복합체 결정의 x-ray 회절 데이터 수집하였다. 그 후, 데이터 분석, SAD 방법을 이용한 phasing, 아미노산 피팅, 구조 최적화 및 분석을 통해 3차원 구조를 규명하였다.
카타닌 복합체의 구조 규명을 통해 카타닌 p60과-p80의 결합부위의 상호작용을 분석하였다. 생화학적 연구를 통해 규명된 구조가 유효하다는 것을 증명할 수 있었다.
연구결과의 활용계획
본 연구의 구조적, 생화학적 결과를 바탕으로 후속연구가 계속된다면 미세소관의 역동성 조절에 대한 기초적 이해뿐만 아니라, 역동성 조절 인자의 변이로 발병하는 각종 신경학적 질환 등에 대한 치료제 개발에도 매우 중요한 자료로 활용될 수 있을 것이다.

In eukaryotic cells, microtubules play critical roles in maintaining the structure of the cell and cell migration. The microtubule can dynamically switch between growing and shrinking phases that important for functions of microtubule. Microtubule-associated proteins carry out a wide range of functions in microtubule dynamics.
Miicrotubule-severing enzymes contribute to cellular activities including mitosis and meiosis, morphogenesis, cilia biogenesis and disassembly, and migration. However, the molecular mechanisms of microtubule severing enzymes are still not understood.
In this study, I report the results of structural and biochemical studies of katanin, the microtubule severing protein, to determine the molecular mechanisms of microtubule severing enzymes.
Result
To determine the molecular mechanisms of katanin p60-p80 complex, structural and biochemical studies were carried out in this study.
Each cloned katanin p60 and p80 genes was successfully co-expressed in Escherichia coli system, and over 95% pure katanin p60-p80 protein complex was obtained by affinity chromatography, TEV protease cleavage, and size exclusion chromatography.
The katanin p60-p80 complex was crystallized with various conditions of protein concentration, salt, and additional precipitants. Then, X-ray diffraction data of katanin p60-p80 complex were collected at the Pohang Accelerator Laboratory, and the data were processed and refined. Finally, Katanin p60-p80 complex structure was determined by Se-SAD method. The interaction between p60 and p80 was analyzed, and the structure of katanin p60-p80 was validated by biochemical studies.
Expected Contribution
Structural and biochemical results in this study could contribute to understand the molecular mechanisms of microtubule severing enzymes academically, and further studies could be used for development medicines of many neurological diseases.
