연구의 목적 및 내용
본 연구에서는 트랜스퍼 몰딩 기술을 이용하여 프로그래머블 약물전달 가능한 심혈관 질병 치료용 stretchable 마이크로니들 메쉬를 연구 및 개발하였음. 특히, 혈관과의 유사한 기계적 특성을 갖는 수술용 메쉬 위에 마이크로니들 구조를 형성하여, 혈관의 지속적인 수축 및 팽창에 영향을 주지 않으며, 혈관으로의 높은 약물전달 효율을 갖는 프로그래머블 약물전달 장치를 개발하였음. 또한, 본 마이크로니들 메쉬를 이용한 동물실험을 통해 그 치료효과 및 개발된 약물전달 장치의 혈관으로의 안정성을 확인하였음.
연구의 목적 및 내용
본 연구에서는 트랜스퍼 몰딩 기술을 이용하여 프로그래머블 약물전달 가능한 심혈관 질병 치료용 stretchable 마이크로니들 메쉬를 연구 및 개발하였음. 특히, 혈관과의 유사한 기계적 특성을 갖는 수술용 메쉬 위에 마이크로니들 구조를 형성하여, 혈관의 지속적인 수축 및 팽창에 영향을 주지 않으며, 혈관으로의 높은 약물전달 효율을 갖는 프로그래머블 약물전달 장치를 개발하였음. 또한, 본 마이크로니들 메쉬를 이용한 동물실험을 통해 그 치료효과 및 개발된 약물전달 장치의 혈관으로의 안정성을 확인하였음.
연구결과
본 연구에서는 트랜스퍼 몰딩 기반 혼합 마이크로니들 메쉬 제작, 프로그래머블 약물전달이 가능한 stretchable 마이크로니들 제작 및 약물전달 특성 연구, stretchable 생분해성 기판 제작 및 혈관과의 기계적 유사성 연구, 마이크로니들 메쉬의 혈관조직으로의 약물전달 특성 연구 및 심혈관 질환 치료효과 입증의 목표를 갖고 연구를 수행하였음.
■ 트랜스퍼 몰딩 기반 혼합 마이크로니들 메쉬 제작에서는 잘 늘어나고 유연한 수술용 메쉬 위에 마이크로니들을 제작할 수 있는 트랜스퍼 몰딩 기술을 개발, 이종 재료의 기판에 자유로이 생분해성 마이크로 니들을 제작할 수 있는 원천기술을 개발하였음.
■ 프로그래머블 약물전달이 가능한 stretchable 마이크로니들 제작 및 약물전달 특성 연구로 혈관의 협착 및 폐색을 방지할 수 있도록, 두 가지 이상의 약물의 시간차 전달 기술을 개발하여 약물분포 및 시간에 따른 치료효과의 정량적 분석을 진행하였음. 이에 따라, 본 연구에서 개발된 프로그래머블 마이크로 니들의 차별적 우수성을 보였음.
■ Stretchable 생분해성 기판 제작 및 혈관과의 기계적 특성 연구를 통해 실크 피브로인 기반의 생체 적합성 메쉬 기판을 제작하여 그 기계적 특성을 연구하였고, 생분해성 수술용 메쉬를 이용하여 마이크로니들 메쉬를 제작, 혈관과의 기계적 유사성을 분석하여 마이크로니들 메쉬의 최적의 디자인 및 혈관으로의 안정성을 확보하였음.
■ 마이크로니들 메쉬의 혈관조직으로의 약물전달 특성 연구 및 심혈관 질환 치료효과 입증을 위해 토끼 동물모델을 이용하여 ex vivo 형태의 동물 실험을 진행, 마이크로니들의 혈관으로의 조직투과 결과를 통해 혈관외벽으로의 약물전달 가능성을 확인하였음. 또한, in vivo 형태의 동물실험을 통해 마이크로니들 메쉬의 혈관신생내막과다형성 억제 및 혈관조직으로의 장착 안정성을 확인하였음.
연구결과의 활용계획
본 연구에서 개발한 프로그래머블 약물전달 가능한 심혈관 질환용 마이크로니들 메쉬는 마이크로니들의 혈관질환 적용에의 기초 원천 연구로, 본 연구에서는 기술 개발 및 유효성 검증이 진행됨. 그에 따라, 본 연구는 심혈관 질환치료의 새로운 방향이 될 수 있을 것이라 판단됨. 특히, 본 연구에서 검증된 마이크로니들 메쉬의 혈관으로의 높은 약물전달 효율 및 우수한 치료효과를 바탕으로 향후 임상진입 및 상용화 기술로의 높은 가능성을 가짐이 판단되며, 본 연구의 개발 결과를 바탕으로 국내외 특허 출원 및 기업 대상으로의 기술이전을 적극적으로 시도하고자 함.

We developed “stretchable & programmable” microneedle mesh for the treatment of vascular diseases using a transfer molding method. With an array of heterogeneous microneedles fabricated on flexible surgical mesh which has similar mechanical properties with blood vessel, we investigated highly efficient drug delivery devices without affecting contraction and expansion of blood vessels. After both ex vivo and in vivo animal studies using a balloon injury model, we demonstrated feasibility, high efficacy and safety of our microneedle mesh to treat vascular disease.
Result
In this study, we investigated microneedle mesh with the aims: 1) fabrication of stretchable microneedle mesh using a transfer molding method, 2) development of heterogeneous microneedle and its drug delivery profile, 3) fabrication of stretchable substrate with the study of mechanical properties between biodegradable mesh and blood vessel, and 4)efficacy of microneedle meshes to vascular disease”
■ Fabrication of stretchable microneedle mesh using a transfer molding method: We developed a transfer molding method to fabricate multiple array of biodegradable microneedles on flexible and stretchable surgical meshes.
■ Development of heterogeneous microneedle and its drug delivery profile: In order to prevent both stenosis and occlusion of blood vessel, we investigated the programmable drug delivery technology which can deliver of multiple drugs in sequence.
■ Fabrication of a stretchable substrate with the study of mechanical properties between biodegradable mesh and blood vessel: We fabricated a silk fibroin substrate and analysed its mechanical properties. Moreover, using surgical mesh, we demonstrated optimal design and its safety to blood vessels by analyses of moduli between microneedle mesh and blood vessel.
■ Efficacy of microneedle meshes to vascular disease: After ex vivo animal study, the feasibility of microneedle meshes to deliver drugs into vascular tissue was confirmed. Finally, after in vivo animal study using injury model, we proved both high efficacy of microneedle mesh about intimal hyperplasia and safety to blood vessel after installation of our devices.
Expected Contribution
In this study, we developed a novel medical device technology, stretchable and programmable microneedle meshes, and its validity for the treatment of vascular disease by developing using a transfer molding method. This technology is expected to offer a new solution to the clinically unmet needs of preventing intimal hyperplasia with its sufficient drug delivery efficiency and efficacy. Efforts for filing additional intellectual properties and commercial development will be made.
