본 연구과제에서는 다음과 같은 연구결과를 달성. (1) 대사성 질환 마우스 모델의 조직에 대한 대규모 전사체 분석을 통하여 신규 오믹스 비만마커 유전자들의 발현량을 검증하였고, in vitro 모델에서 역할을 규명하였으며, 마커 유전자 변이 동물 모델을 디자인하고 제작. (2) 임상 데이터베이스를 구축하고 오믹스마커 단백질을 임상샘플에서 검증. (3) 오믹스마커 분비 단백질 진단 키트 제작을 위한 기반기술 마련. (4) 대사성 스트레스 동물모델을 대상으로 천연물 복합 조성물의 유효성을 검증. (5) 전통천연물의 독성 유발 가능성을

(8) 태음조위탕의 약재 중 하나인 길경이 동물실험결과 항비반/당뇨에 좋은 효능을 보이는 것으로 나타남. (9) 각 천연물질의 작용기전 규명을 위한 단백질체 분석실험을 통해 간조직에서 1,500개가 넘는 단백질을 동정함으로써 고품질의 단백질체 오믹스 데이터를 생산.

The ultimate goals of the study are to identify multi-component/multi-target (MC/MT) biomarkers through multi-omics data analysis and to establish a clinical database for the development of personalized diagnostic technology. Under the objective of identifying biomarkers from multi-omics data analysis using the mouse model with a metabolic syndrome, Stage I studies have been conducted as follows:
(1) Through transcriptome analysis of diet-induced and traditional natural compounds-administered obese mice tissues, omics-based new biomarkers were identified, which revealed significant changes in their expressions.
(2) By establishing in vitro models, the effect of traditional natural compounds, and the role and the mechanism of the identified biomarkers were investigated.
(3) In vivo animal models were generated in order to examine the roles of omics-based biomarkers and the effect of natural compounds on them.
(4) To establish a clinical database of the obesity-related biomarkers, clinical information and samples from obese patients and normal-weight healthy subjects were first collected. Correlation analysis between obesity and the expression level of secretory proteins identified as omics-derived new biomarkers was then performed.
(5) Effect of traditional natural compounds on obesity was analyzed using diet-induced obese mice blood plasma.
(6) An analysis kit that detects and measures a secretory protein newly identified from omics data started to be developed.
(7) Through genomic and transcriptomic data analyses of gastric cancer, one of digestive diseases, new biomarkers were identified, and their functions in gastric cancer were studied.
(8) Major compounds in traditional medicinal prescription have been profiled using bioinformatics tools.
(9) Phenotypic changes in a diet-induced obese subject were observed with respect to of obesity and inflammatory disease, and interactions among multi-tissues such as liver, adipose tissue, muscle, intestine and lung were analyzed.
(10) Toxicity-related biomarkers regulated by multicomponent natural products were identified using omics technology and combinatorial library for application to toxicity evaluation and development of toxicity prediction model.
(11) The identified biomarkers regulated by multicomponent natural products were validated in vitro and in vivo through established alternative toxicity test and high-throughput assay design.
(12) A rapid toxicity prediction system was constructed, and in vitro toxicity testing method was established for functional guidance bioindicator.
(13) Toxicity levels in the main organelles (mitochondria, ER, and Golgi) were predicted, and a network analysis of value index that enables people to interpret data was conducted. Mechanism of organelle toxicity caused by multicomponent natural product was also investigated.
(14) Metabolite structure and absorption/biotransformation mechanism of natural products were identified.
(15) Mass spectrometry-based endogenous metabolite profiling of natural product-treated in vivo model was conducted, and metabolite biomarkers in a network system were identified.
Taken together, these studies will further contribute to application of natural products and their combinations to suitable animal models, identification of biomarkers through integrative analysis of omics data, and validation of the identified biomarkers through in vitro experiments, in vivo preclinical experiments, clinical databases and construction of appropriate animal models. Such newly identified and validated biomarkers will further lead to the development of diagnosis kits for metabolic diseases, and natural product-derived drugs with low toxicity and high efficacy.
