본 연구는 벼의 이화학적 성분(아밀로스, 단백질), 흰잎마름병, 도열병, 저온발아성 및 밀의 단백질 조성에 대해 평가하고 DB를 구축하여 유용한 정보를 제공하기 위하여 수행하였다. 3년간 과제수행을 통해 NIR 검량식, 저온평가용 분자마커 개발, 흰잎마름병 저항성 유전자 탐색 등 대량평가 기술을 개발하였고 도열병 저항성 유전자, 밀 글루텐 점수체계 등 기존의 평가 기술을 적용하여 대량의 자원을 평가하고 유망자원을 선발하여 연구소재 확대에 기여하였다.

Plant genetic reources are essential for developing new varieties related to stress tolerant, climate change, and functional compound. Construction of database for information of agricultural important traits and supply of this information would increase use of genetic resources efficiently. this study was carried out for evaluation of rice physiochemical(amylose, protein), bacterial leaf blight, blast, low-temperature germinability and wheat protein composition, then supply for evaluation result through database. Rice, wheat genetic resources are the largest number of accesisons among germplasm conserved in GeneBank and required huge time and effort for evaluation of useful traits. So, we have conducted the mass-evaluation of these traits and developed new technique for future evaluation.
□ Results
Near infrared spectroscopy was applied for mass-evaluation of rice amylose and protein contents using enhanced calibration curve which was enlarged with application range for prediction of quantity. A total of 20,311 accessions were evaluated by this curve for the traits during 3 years. A total of 10,234 rice accessions were evaluate to Korean rice bacterial leaf blight race of K1, K2 K3 and K3a. Two accessions showed highly strong resistant to all pathogens and GWAS detected resistant gene loci for them. Low-temperature germination(LTG) characteristics of 2,000 accessions were evaluated and five SNP markers were developed through association analysis between phenotype and genotype. Combinations of these markers have elevated prediction efficiency for good LTG compared to single gene marker. Rice blast gene pattern in 9,153 germplasm collected from Japan and China were profiled using 12 rice blast genes. We found 4 accessions possessing maximum 10 genes and constructed DB for these profiling data. High molecular weight glutenin subunit related to the baking characteristics of 4,190 wheat accessions were investigated using SDS-PAGE and DNA markers. We selected 42 accessions with superb bread-baking quality and early maturity.
□ Expected Contribution
We have established mass-evaluation methods thorough application of NIR calibration curve and molecular markers. These technologies will be used in the future to enable rapid evaluation of genetic resources, especially in the large quantities such as food crops in order to obtain information on useful traits in a short period. Genetic resources selected using bioassay and existing evaluation system can be used as materials for breeding program and developing molecular markers.
